(4+3) annulation of enone-tethered indoles with α-alkylidene succinimides: access to seven-membered azepino[3,2,1-hi]indole scaffolds.

We have developed a straightforward synthesis of azepino[3,2,1-hi]indole architectures with four contiguous chiral carbons as single diastereomers. This metal-free protocol between indole-enones and alkylidene succinimides proceeds efficiently under ambient conditions, demonstrating a broad scope in good to excellent yields. The feasibility of the current methodology was illustrated by one-pot, gram-scale synthesis and further modifications.

Switchable Divergent Synthesis of Cyclopropane Scaffolds Enabled by Photoredox Catalytic Cascade.

Given the essential and valuable characteristics of cyclopropane scaffolds in pharmaceutical and organic chemistry, general strategies for their construction are highly desirable. However, the divergent synthesis of spirocyclopropanes and 1,1-substituted cyclopropanes using the same starting materials remains a challenge. Herein, we disclose a visible-light-promoted procedure for the concise and divergent construction of spirocyclopropyl succinimides (28 examples, yields up to 95%) and cyclopropyl-acetamides (18 examples, yields up to 82%) from N-substituted oxamic acids and methyl 4-chloro-2-

Oxonium/Sulfonium Ylide Rearrangements to Access Chromenes and Thiochromenes: Concerted or Stepwise Pathway?

Dirhodium(II)-catalyzed regio-divergent intramolecular rearrangement of oxonium and sulfonium ylides, generated from diazo arylidene succinimides (DAS), has been achieved at room temperature. Allyloxy-substituted DAS derivatives were strategically designed and synthesized to explore their reactivity under metal catalysis. The transformation is proposed to proceed via oxonium ylide intermediate, followed by [1,2]- and [1,4]-shifts, leading to the chromene framework. This strategy was further extended to thioallyl-substituted DASs to access thiochromenes under mild reaction conditions. For the f

Biointerface Strategies for COVID-19 Spike Protein Detection: A Surface Engineering Approach.

Rapid diagnosis or health monitoring biosensors have been developed with the advent of technology. Silicon and metal oxides are used as the base material for these biosensors as a point-of-care unit. However, surface modification is needed to introduce functional groups for anchoring the bioreceptor. This study aims to explore the differences in the spike protein binding efficiency directly on the bare amine self-assembled monolayers (SAMs) and further cross-linked with the N-ethyl-N'-(3-(dimethylamino)propyl)carbodiimide (EDC)/N-hydroxysuccinimide (NHS) and glutaraldehyde (GA). The specificit

One-step Immobilization of Human α-1-acid Glycoprotein on Magnetic Beads: A Rapid Method for Small Molecule hAGP Binding Study.

Human α-1-acid glycoprotein (hAGP) immobilized directly on NHS ester (N-hydroxysuccinimide) activated magnetic beads (hAGP-beads) was developed as a novel tool for studying small molecule hAGP binding. This method offers a straightforward, one-step immobilization process compared to traditional biotin-streptavidin immobilization technique. The hAGP-beads system provides a rapid and convenient alternative to conventional methods like equilibrium dialysis (ED) or ultracentrifugation (UF) for assessing hAGP small molecule interactions. Characterization and evaluation of the hAGP-beads system reve

Synthesis and evaluation of N-arylsulfonylated succinimides as activity-based probes.

Activity-based protein profiling (ABPP) technology has served as a powerful platform for studying proteins for more than two decades. However, the further growth of this field depends on the development of new probe structures to expand the proportion of the proteome that can be studied using these methods. Inspired by previous reports of succinimide-containing covalent inhibitors for proteases, we synthesized a panel of potential probe structures with a succinimide reactive group and a terminal alkyne tag suitable for subsequent azide-alkyne click chemistry. Members of this panel with an N-ar

13-Plex DeAla Isobaric Reagents for High-Throughput Proteome Quantification.

Isobaric labeling techniques are widely used in mass spectrometry-based quantitative proteomics to enable the simultaneous analysis of multiple samples. However, commercial isobaric tags are expensive due to complex synthesis and costly reagents, limiting their use in large-scale studies. Here, we introduce a novel, cost-effective diethylalanine-based isobaric reagent (DeAla), synthesized using diethylated alanine and β-alanine with N-hydroxysuccinimide. The DeAla tag offers several advantages, including improved peptide fragmentation, enhanced protein identification, and competitive pricing.

Synthesis and Screening of Bispecific Linker-based Combinatorial Libraries Identified Thiosuccinimide Lipids for mRNA Delivery.

Ionizable lipids are essential for lipid nanoparticle-mediated mRNA delivery. The discovery of novel ionizable lipids relies on combinatorial chemistry synthesis and screening. However, current libraries mainly focus on optimizing amine cores and hydrophobic tails, often neglecting linker optimization, which restricts novel ionizable lipid discovery. To address this, a one-pot, two-step, three-component reaction based on N-hydroxysuccinimide-ester-maleimide bispecific linkers is first developed for constructing combinatorial libraries. Two libraries containing 344 thiosuccinimide lipids are sy

Polyvalent Thioesters as Multifunctional Chemical Anchors: Toward Synthesis of Homo/Hetero Peptide or Miniprotein Multimers.

In this work, we develop a platform for multimerizing peptides/miniproteins using various polyvalent thioester cores derived from easy-to-synthesize N-hydroxysuccinimide esters. We employed native chemical ligation to attach multiple copies of peptide/miniprotein around a fixed multi-armed thioester core in a one-pot fashion, leading to the first-generation multimers. Further, using a reverse thioether ligation strategy, we synthesized second-generation branched homo/hetero multimers. The reactions proceed under an aqueous environment that closely mimics physiological conditions, forming multi

Click-linking: a cell-compatible protein crosslinking method based on click chemistry.

Crosslinking mass spectrometry (XL-MS) has the potential to map the human interactome at high resolution and with high fidelity, replacing indirect, error-prone sampling methods such as affinity pulldown MS. However, the sampling depth of XL-MS remains stubbornly low. We present a crosslinking strategy that splits the crosslinking reaction into two sequential and orthogonal coupling events. The method involves pre-stabilizing the spatial proteome with a fixation protocol inspired by immunofluorescence imaging, followed by a stepwise process that begins with extensively labeling surface-accessi

Site-selective addition of succinimide motif through nitro-assisted C-H functionalization of (hetero) arenes under rhodium catalysis.

Succinimide motifs are recognized as privileged cores in anticonvulsants and antipsychotics such as phensuximide, ethosuximide, and lurasidone. These succinimides can be readily converted into pharmaceutically important pyrrolidines and γ-lactam scaffolds, making them highly promising compounds in drug discovery. Nitroarenes are also important chemical feedstocks and have attracted increased attention owing to their versatile applications in pharmaceuticals, functional materials, and agricultural pharmacology. Therefore, directly combining succinimides with nitroarenes is a valuable approach f

Access to Succinimidyl Spirocyclobutenes via Spiroannulation of Bicyclo[1.1.0]Butanes and Azaoxyallyl Cations.

We herein disclose a base-mediated annulation reaction between azaoxyallyl cation and arylbicyclo[1.1.0]butane carboxylates (BCBs) to access succinimidyl spirocyclobutenes. The base played a dual role in forming the electrophilic azaoxyallyl cation species and also generating an enolate species from BCB and HFIP also assisted the stabilization of the intermediates and cyclization. Control experiments reveal the explicit involvement of the azaoxyallyl cation.

Optimized In-Solution and Gas-Phase Chemistry Enables High-Efficiency Interactome Mapping by DSBSO-Based Cross-Linking Mass Spectrometry.

Cross-linking mass spectrometry (XL-MS) allows characterizing protein structures and interactions in highly complex samples. The enrichable disuccinimidyl bissulfoxide (DSBSO) cross-linker has enabled comprehensive XL-MS studies of human cells. However, existing DSBSO workflows demand multi-day sample preparation with high input requirements and provide insufficient detection sensitivity. Here, we systematically optimize the in solution and gas-phase chemistry of azide-A-DSBSO-based XL-MS. Importantly, we reduce sample preparation time to 10 h and introduce StageTip-based strong cation exchang

Co(II)-Catalyzed Picolinamide-Directed C(sp3)-S Bond Formation with N-(phenylsulfanyl)succinimides.

Herein, we disclose a novel and efficient cobalt-catalyzed cross-coupling strategy for picolinamide-directed direct C(sp3)-H bond formation with N-(phenylsulfanyl)succinimides. This method enables the direct construction of C(sp3)-S bonds under mild conditions and exhibits excellent functional group tolerance along with a broad substrate scope. Notably, the catalytic system achieves oxidative C-H functionalization without relying on costly or environmentally detrimental oxidants, offering a more sustainable and practical alternative for C-S bond formation.

Afterglow ice formed by phosphorescent luminophore-protein conjugates and complexes in aqueous solution at freezing temperature.

Unlike the widely reported fluorophore-biomacromolecule systems in nucleic acid testing and immunoassay, organic afterglow luminophore-biomacromolecule conjugates and complexes remain rarely explored. Here we report the observation of organic afterglow from aqueous solutions of luminophore-protein conjugates and complexes at freezing temperature, named as afterglow ice for abbreviation. Reliable N-hydroxysuccinimide ester protocol, as well as β-diketone chemistry, are applied for protein labeling to form luminophore-protein conjugates, which exhibit intriguing afterglow at freezing temperature

Clickable Multitargeting Cross-Linkers Based on SuFEx for In Vivo Cross-Linking Mass Spectrometry.

Cross-linking mass spectrometry (XL-MS) is a powerful technique to study protein structures and protein-protein interactions. The coverage of amino acids determines the depth of the XL-MS analysis and accuracy of the structural resolution. In this work, we designed and synthesized a pair of novel trifunctional cross-linkers, succinimidyl-propargyl-aryl sulfonyl fluoride (SPSF), for multitargeting cross-linking in vivo. The highly reactive succinimide ester reacts rapidly with Lys residues first; then, the less reactive sulfonyl fluoride reacts with multiple nucleophilic amino acid side chains

Bioinspired nanocellulose-based ultrathin hydrogel bioadhesives with sweat-resistant properties.

Hydrogels have gained prominence as a distinctive material for smart healthcare devices and wearable electronics, owing to their inherent biocompatibility, tissue-like mechanical properties and superior water and oxygen permeability. In most application scenarios, hydrogels are required to have certain adhesion properties. However, current hydrogel adhesive systems are fundamentally constrained by two intrinsic material restrictions: low mechanical strength, and compromised adhesion in humid environments (under conditions such as sweating and bleeding), which severely restrict their practical

Investigation of the Effect of C-Terminal Adjacent Phenylalanine Residues on Asparagine Deamidation by Quantum Chemical Calculations.

The deamidation rate is relatively high for Asn residues with Phe as the C-terminal adjacent residue in γS-crystallin, which is one of the human crystalline lens proteins. However, peptide-based experiments indicated that bulky amino acid residues on the C-terminal side impaired Asn deamination. In this study, we hypothesized that the side chain of Phe affects the Asn deamidation rate and investigated the succinimide formation process using quantum chemical calculations. The B3LYP density functional theory was used to obtain optimized geometries of energy minima and transition states, and MP2
