Recombinant Antigen for Newcastle Disease Virus Vaccine Focusing on Currently Circulating Virus Variants.

Newcastle disease virus (NDV) causes a highly contagious avian infection and is characterised by its high genetic diversity and mutation rate. Currently applied vaccines are based on NDV genotypes I and II, which are considerably distinct from the circulating pathogenic genotypes, of which genotype VII is predominant, and therefore, they may not provide complete protection for birds or stop virus transmission. This work is a proof-of-concept study aiming to develop a genotype VII-focused recombinant antigen for a vaccine targeting relevant NDV strains. The research primarily focuses on novel a

Viral Respiratory Infections: Upper and Lower Respiratory Tract Infections.

Respiratory infections are a major health concern and are divided into upper and lower respiratory tract infections. Upper respiratory tract infections are mostly viral and self-limiting; billions of cases occur globally each year. Lower respiratory tract infections such as pneumonia and bronchitis can cause significant illness, especially in children, older adults, and individuals with comorbidities or immunocompromise. In 2021, lower respiratory tract infections caused more than 2 million deaths globally, mainly in low- and middle-income countries. Preventive measures include hand hygiene, m

Designing a multi-epitope vaccine against La Crosse virus using immunoinformatics approach.

La Crosse virus (LACV) is a mosquito-borne Orthobunyavirus responsible for pediatric encephalitis, with no currently approved vaccine or targeted antiviral therapy. This study aimed to design a multi-epitope subunit vaccine targeting the conserved nucleoprotein of LACV using an integrated immunoinformatics approach. The nucleoprotein exhibited strong antigenicity and favorable physicochemical properties, including stability and hydrophilicity. Highly conserved CTL, HTL, and B-cell epitopes were initially predicted, from which high-affinity, non-allergenic, and non-toxic epitopes were selected.

A Self-Assembling β-Annulus Nanoparticle Vaccine Incorporating OX40L Confers Partial Protection Against NADC30-Like PRRSV in Piglets.

Porcine reproductive and respiratory syndrome (PRRS) remains one of the most economically significant viral diseases affecting the global swine industry, and currently available commercial vaccines provide only limited protection against circulating PRRS virus (PRRSV) strains. Therefore, alternative vaccination strategies are urgently needed to enhance protection against PRRSV infection. In this study, selected antigenic regions derived from the PRRSV structural proteins GP3, GP4, GP5, and M were incorporated into a β-annulus peptide-based nanoparticle scaffold derived from tomato bushy stunt

Early classical swine fever virus persistence is associated with cytotoxic and immunoregulatory transcriptional signatures distinct from protective C-strain vaccination.

Persistent classical swine fever virus (CSFV) infection represents a major challenge for disease control because persistently infected pigs remain clinically healthy while continuously shedding virus. However, the early immunological events associated with the establishment of viral persistence remain incompletely understood and require further investigation. In this study, we compared the peripheral blood mononuclear cell (PBMC) transcriptomes of neonatal piglets infected with the moderately virulent Catalonia01 CSFV strain, previously shown to induce postnatal persistent infection, with thos

An immunoinformatics-driven multiepitope subunit vaccine targeting Oropouche virus: Molecular docking, immune and dynamics simulation approaches.

The Oropouche virus (OROV) is an emerging zoonotic orthobunyavirus responsible for recurring outbreaks of febrile illness and neurological disorders in South and Central America. Despite its growing public health issues, no FDA-approved vaccines or antiviral therapies currently exist. To address this gap, a multiepitope subunit vaccine was designed to target OROV glycoproteins G1 (Gn), G2 (Gc), and the nucleocapsid (N) protein using a reverse-vaccinology (RV) approach. For vaccine construction, suitable epitopes (MHC-I, MHC-II, and linear B-cell) were selected based on percentile rank, antigen

Concurrent administration of a combined clostridial vaccine or ivermectin suppresses the antibody response to foot-and-mouth disease vaccine in sheep.

Foot-and-mouth disease (FMD) is a highly contagious viral disease affecting cloven-hoofed animals and poses severe economic losses to the sheep industry. In intensive sheep farming, simultaneous administration of the FMD vaccine with other vaccines or anthelmintics is frequently practiced to reduce handling stress and labor costs. However, whether such combined administration can influence the immunological efficacy of FMD vaccine still lacks sufficient research evidence. In this study, 96 healthy sheep were randomly allocated into three groups (n=32 each). Group A received FMD vaccine alone (

Engineering a dimeric porcine deltacoronavirus S-RBD subunit vaccine for enhanced immunogenicity and neutralizing antibody responses.

BACKGROUND: Porcine deltacoronavirus (PDCoV) is an emerging swine enteric coronavirus that has caused global epidemics and substantial economic losses, posing a potential risk of cross-species transmission, including to humans. However, commercial vaccines are currently scarce. The receptor binding domain (RBD) of the spike (S) protein is a key target for inducing neutralizing antibodies, but the RBD monomer typically exhibits low molecular weight and weak immunogenicity. This study aimed to develop a tandem dimeric RBD subunit vaccine and evaluate its immunogenicity and protective efficacy in

The human gut microbiome primes fever after vaccination.

Fever is a common adverse reaction to vaccination, contributing to vaccine hesitancy and reduced uptake. To understand variation in fever risk, we longitudinally profiled fecal microbiota, oral temperature, and serological markers in 171 healthy adults receiving Severe acute respiratory syndrome coronavirus 2 mRNA vaccines. Fever risk correlated with low-grade intestinal inflammation, increased abundance of flagellated Lachnospiraceae bacteria, and increased flagellin expression prevaccine. Microbiomes from fever-high donors triggered stronger inflammation in human intestinal organoids and dro

Reconsidering vaccinate-to-live strategies in EU foot-and-mouth disease control.

In January 2025, FMD was detected in water buffalos near Berlin. A few weeks later, a different FMDV-strain was confirmed on dairy farms in Slovakia and Hungary. Since February 2026, Cyprus and Lesvos are confronted with FMD-outbreaks. While Europe is rethinking its FMD-strategies, the two affected member states, Cyprus and Greece, take opposite approaches to control the disease. FMD-vaccination was pivotal for its eradication in many parts of the world. Such regions fought new outbreaks by traditional animal-disease-control. In 2001, a devastating FMD-epizootic swept across the UK and contine

A recombinant Newcastle disease virus-vectored multi-epitope vaccine confers protection against QX-like infectious bronchitis virus in chickens.

Infectious bronchitis virus (IBV) QX-like (GI-19) genotype dominates global poultry flocks, yet Mass-type vaccines fail to confer cross-protection. We developed a recombinant Newcastle disease virus (NDV) LaSota vector expressing conserved IBV S1 B-cell hotspots paired with S1-derived BF2-restricted CTL epitopes (rNDV-IBV-T/B). Specific-pathogen-free chickens received single or booster ocular-nasal immunization (106 EID50) followed by QX-like challenge. Booster immunization increased the frequency of splenic CD3+CD8+ T cells (41.3% vs. 31.9% in the sham-vaccinated challenge control; Tukey-adju

Characterization and immunogenicity of nanoparticle vaccines displaying embecovirus spike proteins.

Endemic human coronaviruses OC43 and HKU1 cause widespread respiratory infections and can be associated with severe illness in immunocompromised and elderly individuals. Frequent adaptive evolution in the spike proteins of these embecoviruses and the potential for zoonotic transmission from a large animal reservoir necessitates the characterization of the immunogenic landscape of the spike proteins of embecoviruses. Here, we constructed nanoparticle vaccines displaying the spike antigens from OC43, HKU1 A, or HKU1 B, as well as a bivalent formulation incorporating spike antigens from OC43 and

Mycotoxins as silent modulators of enteric viral susceptibility and vaccine responsiveness.

Enteric viral infections remain a major cause of morbidity and mortality worldwide, particularly in children and intensively raised livestock. Although live oral vaccines have reduced disease burden, vaccine efficacy varies greatly across regions. Factors such as host genetics, microbiota, maternal antibodies, and nutrition have been widely studied, whereas chronic exposure to dietary toxicants has received less attention. Mycotoxins, fungal metabolites contaminating staple crops including maize, wheat, and peanuts, are commonly consumed at low but persistent levels in regions heavily affected

Design and immunoinformatics characterization of a novel multi-epitope vaccine rPRRSV-35N targeting NADC30-like and NADC34-like porcine reproductive and respiratory syndrome virus variants.

BACKGROUND: Porcine reproductive and respiratory syndrome (PRRS), caused by the genetically diverse porcine reproductive and respiratory syndrome virus (PRRSV), remains a major economic burden to the global swine industry. Current commercial vaccines, including modified live virus (MLV) and inactivated vaccines, have shown limited effectiveness against newly emerging PRRSV variants, particularly NADC30-like and NADC34-like strains. There is therefore a need for improved vaccine strategies targeting these circulating strains. In this study, we used immunoinformatics approaches to design a multi

Integrated Single-Cell and TCR Profiling Reveals Protection-Associated CD8+ T Cell Subsets Linked to Viral Control in PRRSV.

Porcine reproductive and respiratory syndrome virus (PRRSV) remains a major threat to global swine industry, yet the immune mechanisms underlying protective vaccination are incompletely understood. Here, we applied integrated single-cell RNA sequencing and T cell receptor (TCR) profiling to characterize immune responses in a PRRSV vaccination-challenge model spanning complete, partial, and non-protection outcomes. We identified distinct CD8+ T cell subsets that were selectively enriched in protected animals vaccinated with modified live vaccine (MLV) and marked by clonal expansion, strong cyto

Development and evaluation of a virus-like particle PEDV vaccine based on MS2 bacteriophage.

Porcine epidemic diarrhea virus (PEDV) can infect pigs of all ages, causing porcine epidemic diarrhea and resulting in significant losses to the global pig farming industry. The virus causes epidemic diarrhea and represents a significant threat to the global swine industry. To enhance vaccine safety and protective efficacy, a PEDV nanoparticle vaccine (PEDV-MS2 VLP) based on MS2 bacteriophages was developed. The MS2 phage coat (MS2 VLP) self-assembles into nanoparticles within Escherichia coli. It is conjugated to the PEDV RBD antigen via the SpyTag-SpyCatcher system, forming a structure mimic
