Distinct affective neural phenotypes in breast cancer survivors: implications for personalized supportive care after chemotherapy.

PURPOSE: Anxiety and emotional dysregulation remain among the most prevalent unmet supportive care needs in breast cancer survivorship, yet reported affective outcomes are strikingly inconsistent, with some survivors showing elevated distress and others displaying resilience or post-traumatic growth. Because distress screening in survivorship relies almost exclusively on self-report, the neural mechanisms shaping how chemotherapy-exposed survivors perceive and report emotional experience remain poorly understood, limiting mechanism-informed psychosocial support. METHODS: Forty female breast c

GP perspectives on genomics in primary care: a qualitative study on using polygenic risk scores to evaluate cancer risk.

BACKGROUND: A polygenic risk score (PRS) enables personalisation of cancer risk and supporting risk stratification for melanoma, colorectal, breast, and prostate cancers. Including a PRS in a cancer risk assessment can facilitate risk-appropriate cancer screening by incorporating an individual's age, sex, family history, and genomic test results. GPs are the likely healthcare professionals to order PRS tests and deliver results to patients within existing preventative health models. AIM: To elucidate GPs' perspectives on the use of PRSs to tailor cancer screening in the Australian primary car

False-Negative Screening and Diagnostic Mammograms in the National Mammography Database From 2010 to 2022.

BACKGROUND. False-negative (FN) mammograms typically delay breast cancer diagnoses and may impact clinical outcomes. However, systematic evaluations of FN mammograms are challenging to conduct due to the low incidence of interval cancers. OBJECTIVE. The purpose of this study was to evaluate the rates of FN screening and diagnostic mammograms in the NMD and to assess associations of FN rates with patient- and facility-level characteristics. METHODS. This retrospective study included all screening and diagnostic mammograms in the National Mammography Database (NMD) obtained from January 1, 2010,

Mammographic Density as a Mediator for Breast Cancer Risk: Pooled Analysis of Four Population-Based Case-Control Studies on Women with Screening Mammograms before Age 50.

BACKGROUND: Multiple risk factors influence breast cancer risk, but the role of percent mammographic density (PMD) in mediating these associations remains unclear. METHODS: In this pooled analysis of women who had screening mammograms before age 50 years, we assessed whether PMD mediates associations between reproductive, structural, and lifestyle factors and breast cancer risk. Causal mediation analysis was used to estimate direct and indirect effects and the proportion mediated by PMD. RESULTS: PMD partially mediated 29% of the association between breast calcifications and breast cancer ri

A qualitative study exploring Malaysian women's preferences for authority or emotional appeals in public health messages promoting breast cancer screening.

Persuasive health communication is key to promoting breast cancer screening. Due to existing gaps over how public health messages should be framed, a study was conducted to explore the preferences and perceptions of Malaysian women towards messages that incorporate different types of appeals to promote breast cancer screening. In-depth qualitative interviews were conducted among Malaysian women aged 40 and above with no previous history of breast cancer or other cancers. They were exposed to messages that promoted breast cancer screening using seven different appeals; authority, hope, social,

Factors Associated With Inaccurate Recall of Inherited Cancer Genetic Test Results Among Individuals With Germline Pathogenic Variants.

BACKGROUND: Germline pathogenic and likely pathogenic variants (GPV) in cancer susceptibility genes may guide medical care. However, inaccurate recall of test results by patients may impact the uptake of appropriate medical care, underscoring the importance of accurate recall of results. METHODS: Among individuals with a confirmed cancer susceptibility GPV that alters cancer risk management, individuals were categorized based on their accuracy of recall. Multiple logistic regression was performed to determine demographic and psychosocial factors associated with recall of test results. RESULT

STK11 and DNA Repair Gene Mutations Define Hereditary Subset of Middle Eastern Papillary Thyroid Cancer.

Papillary thyroid cancer (PTC) is the most common endocrine malignancy with especially high incidence in Middle Eastern populations. While classical hereditary syndromes explain a minority of cases, the broader germline landscape of non-syndromic PTC remains unclear. Whole-exome sequencing was performed on 245 unselected Saudi PTC patients to identify germline pathogenic or likely pathogenic variants (PVs/LPVs) in cancer predisposition genes. Clinical and molecular characteristics, and family history were integrated to assess phenotypic correlations. Eleven patients (4.5%) harbored germline PV

Molecular and clinical characteristics of glioma patients with germline pathogenic mutations of Lynch syndrome genes: a large-scale multicenter study.

BACKGROUND: Lynch syndrome (LS) is an inherited disorder caused by germline mutations in mismatch repair (MMR) genes or EPCAM deletions and is primarily associated with an increased risk of gastrointestinal and endometrial cancers. However, LS-associated gliomas are rare, and their clinical and molecular characteristics remain poorly defined. The objective of this study was to systematically characterize the prevalence, genetic landscape, molecular features, and clinical outcomes of LS-associated gliomas. METHODS: We retrospectively analyzed a multicenter cohort of 5,594 glioma patients who un

Two decades of PARP inhibitor synthetic lethality in cancer.

Two decades ago, two papers in Nature described how PARP inhibitors selectively killed cells deficient in the BRCA1 or BRCA2 tumour suppressor genes, observations that led to the first clinically approved treatment of a cancer with a targeted therapy selected based on a germline biomarker. This work was recognized by Nature as one of the top 20 discoveries in cancer in the twenty-first century and provides a compelling example of leveraging fundamental biology discovery for patient benefit. For people with specific forms of breast, ovarian, prostate or pancreatic cancer, these discoveries chan

Phenotypic heterogeneity in carriers of a pathogenic MSH2 variant: implications for the diagnosis of Lynch syndrome.

Lynch syndrome is an inherited cancer predisposition syndrome caused by germline pathogenic variants in mismatch repair (MMR) genes and is primarily associated with colorectal and endometrial cancers. Classically, Lynch syndrome-associated tumors exhibit microsatellite instability (MSI) and loss of MMR protein expression on immunohistochemistry (IHC); therefore, MSI and IHC are routinely used in screening algorithms to support diagnostic evaluation. ; Methods: In this study, we present the molecular and pathological characteristics of four cases from three unrelated families carrying the same
