The Impact of Glucagon-Like Peptide-1 Receptor Agonists on Risk of Retinal Vascular Occlusions.

Purpose: To investigate whether glucagon-like peptide-1 receptor agonists are protective against retinal vascular occlusions. Methods: Using a large aggregated, de-identified electronic health records data platform in the United States, patients with type 2 diabetes ages ≥18 years prescribed glucagon-like peptide-1 receptor agonists or non-glucagon-like peptide-1 receptor agonists (controls) and having ophthalmology follow-up of at least 1, 3, or 5 years were included. The glucagon-like peptide-1 receptor agonist and control cohorts were propensity score matched (1:1) on demographics and risk

H3K9me3 inhibition reverses Alzheimer's progression by restoring synaptic and immune proteostasis across the brain-retina axis.

INTRODUCTION: Epigenetic dysregulation is increasingly linked to aging and neurodegeneration, yet its contribution to retinal and brain pathology in Alzheimer's disease (AD) remains unclear. METHODS: We quantified the repressive epigenetic mark histone H3 lysine 9 trimethylation (H3K9me3) in post mortem retinas and brains from donors spanning normal cognition, mild cognitive impairment, and AD dementia. Using a multi-enzyme methyltransferase panel, we characterized the in vitro inhibitory activity of ETP69, an epidithiodiketopiperazine, against suppressor of variegation 3-9 homologue 1 (SUV39

Retinal thickness is associated with global cognition and executive function in mild cognitive impairment due to Alzheimer's disease.

Alzheimer's disease (AD) is a neurodegenerative disease that leads to cognitive impairment and dementia, and retinal imaging is being explored as a biomarker. We examined the differences in retinal thickness by optical coherence tomography (OCT) between 47 participants with mild cognitive impairment due to Alzheimer's disease (Aβ+MCI) and 23 age-matched controls, as well as associations between cognition and OCT. There was no significant difference in retinal thickness between groups. However, retinal measures were robustly associated with global cognition and executive function in those with

The A53T Parkinson's Disease Mouse Model Shows Abnormal Outer Retinal OCT Metabolic Biomarker Patterns.

PURPOSE: The purpose of this study was to test for abnormal optical coherence tomography (OCT) metabolic biomarker patterns in the A53T Parkinson's disease model, which accumulates α-synuclein in photoreceptor inner segments before overt motor symptoms. METHODS: Four-to-6-month-old transgenic A53T homozygous mice and B6C3H wild-type (WT) littermate controls were evaluated using 2 OCT metabolic biomarkers. The first was the external limiting membrane (ELM) to retinal pigment epithelium (RPE) thickness, a proxy for subretinal space acidification and retinoid deficiency. The second biomarker was

Ciliary epithelium detachment during lens subluxation extraction surgery after blunt eye trauma: A case report.

PURPOSE: To document the ciliary epithelial detachment identified intraoperatively during lens-subluxation extraction in a patient with blunt eye trauma. METHOD: A 60-year-old Chinese man presented 14 days after right eye blunt trauma with blurry vision, elevated intraocular pressure (IOP), temporal shallowing of the anterior chamber, and iris bombé 1. To relieve pupillary-block-related IOP elevation, phacoemulsification was performed. Intraoperatively, the lens capsule ruptured, and portions of the nucleus and cortex dropped into the vitreous cavity. The surgeon switched to a cutter to remov

Artificial intelligence (AI) and the choroidal challenge: A comprehensive review of AI applications in choroidal imaging.

The choroid is a densely vascular layer that sustains the outer retina and may reflect systemic microvascular health. With the advent of new imaging techniques, choroidal imaging has become increasingly detailed, generating vast data volumes. Conventional analytic tools often fall short in managing these high-dimensional datasets. Artificial intelligence (AI) automatically enhances, segments, and analyzes choroidal images, facilitating the discovery of choroidal biomarkers relevant to ocular and systemic disease. Yet, anatomical complexity, heterogeneous protocols, and a lack of large, annotat

Enrichment of Retinal Endothelial Cells and Müller Glia Enable Comprehensive scRNA-Seq Profiling of the Blood-Retinal Barrier.

Obtaining high-quality single-cell suspensions from neural tissues such as the murine retina remains technically challenging due to cellular fragility, tight intercellular junctions, and the heterogeneous composition of the tissue. These challenges are further compounded when studying rare cell populations, including vascular endothelial cells and Müller glia, which each account for less than 2% and are underrepresented in standard dissociation workflows, often lost during processing. Here, we present an optimized method for isolating murine retinal cells that preserves cellular integrity and

Extensive monolayer formation after transplantation depends on an RPE subpopulation derived from human iPSCs.

Loss of retinal pigment epithelium (RPE) in the eye leads to photoreceptor dysfunction, death, and eventually, vision loss. Cell replacement strategies using RPE cells derived in vitro from pluripotent stem cells are currently evaluated as a potential therapeutic strategy. Generation of polarized monolayers represents an essential prerequisite for proper RPE function; however, monolayer formation following transplantation of RPE suspensions has not been systematically assessed. Using the sodium iodate mouse model of acute RPE depletion, significant increase in monolayer formation capacity of p

Acute macular neuroretinopathy: is the name still appropriate?

BACKGROUND: Acute macular neuroretinopathy (AMN) is a rare but increasingly reported retinal disorder that predominantly affects young women and is characterized by acute-onset paracentral scotomas and distinctive parafoveal macular lesions. Following its first report in 1975 as an inner retinal disease, a considerable body of work has been produced, expanding on its clinical presentation and systemic associations. Aim of this review is to synthesize the current understanding of AMN, including existing controversies on nomenclature. METHODS: A literature search was conducted in PubMed/MEDLINE

Ifi27l2a Facilitates the Recruitment of Pathogenic T Cells by Retinal Microglia in the Initiation Phase of Experimental Autoimmune Uveoretinitis.

The mechanisms by which pathogenic T cells enter the immune-privileged retina at the onset of uveitis remain unclear. To identify regulatory targets that promote retinal inflammation, we established an experimental autoimmune uveoretinitis (EAU) model by immunizing C57BL/6J mice with IRBP651-670 and dynamically evaluated retinal inflammation after induction; fundus lesions were prominent, with the first 14 days representing the initiation phase, Days 14-28 the amplification phase, Days 28-35 the peak phase, and Days 35-42 the regression phase. We characterized retinal cellular alterations from

The role and mechanism of macrophage-to-myofibroblast transition in fibrotic diseases.

Fibrosis is a pathological process primarily characterized by excessive deposition of extracellular matrix (ECM), which leads to disruption of tissue architecture and progressive loss of organ function, serving as a key pathological basis for various chronic diseases. Recent studies have revealed that, in addition to conventional sources, macrophages can directly differentiate into ECM-producing myofibroblasts through a process termed macrophage-to-myofibroblast transition (MMT), thereby actively participating in fibrogenesis. The concept of MMT was initially proposed in the context of renal f

Macular Holes Secondary to Macular Telangiectasia Type 2: Pathogenesis, Clinical Features, Imaging and Surgical Management.

Full-thickness macular holes (FTMHs) are a rare but recognized complication of macular telangiectasia type 2 (MacTel2). Unlike idiopathic FTMHs, which are primarily driven by vitreofoveal traction, MacTel2-associated holes appear to arise predominantly from progressive neurodegenerative changes. Müller cell dysfunction, intraretinal cavitation, and outer retinal atrophy progressively compromise foveal architecture, ultimately predisposing to tissue dehiscence and full-thickness defects. Although vitreofoveal traction may contribute in individual cases, it is unlikely to be the principal mechan

Unveiling Glial Heterogeneity in the Inner Retina: Non-Visual Photic Spectrum Responses and Calcium Dynamics in Müller Glial Cells vs. Neurons.

Traditional calcium (Ca2+) imaging analysis often overlooks the non-stationary temporal complexity inherent in retinal circuits, reducing dynamic signaling traces to static summary descriptors. To resolve these dynamic signatures, we implemented a reproducible analytical framework combining Continuous Wavelet Transform (CWT) time-frequency decomposition with Sinc baseline filtering and Ward's agglomerative hierarchical clustering, applied directly to standardized high-dimensional feature spaces, reserving Uniform Manifold Approximation and Projection (UMAP) strictly for low-dimensional visuali
