PROTAC degraders for lymphoid malignancies - current status and future directions.

INTRODUCTION: Despite remarkable advances in targeted therapy, some patients with lymphoid malignancies develop resistance to all available treatment modalities. Proteolysis-targeting chimeras (PROTACs), harness the ubiquitin-proteasome system to eliminate both catalytic and scaffolding protein functions of disease-causing proteins, offering a strategy to overcome this resistance. AREAS COVERED: This review discusses the mechanisms of PROTAC activity, current preclinical and clinical data across lymphoid malignancies, and challenges for this emerging therapeutic class. Factors Associated With Clinical Trial Participation in Adult Hematopoietic Stem Cell Transplantation Recipients: A Center for International Blood and Marrow Transplant Research (CIBMTR) Analysis.

BACKGROUND: Only 3%-5% of patients with cancer nationwide enroll in clinical trials. Although participation among patients with solid tumors has been well studied, data in the hematopoietic stem cell transplantation (HSCT) setting remain limited. We sought to (1) describe clinical trial participation rates by HSCT type; (2) examine associations between patient, disease, and transplant characteristics and trial participation; and (3) explore the association between trial participation and overall survival (OS). METHODS: We conducted a retrospective study using Center for International Blood an

Successful treatment with BrECADD in a patient with Dubin-Johnson syndrome and classic Hodgkin lymphoma: a case report.

Dubin-Johnson syndrome (DJS) results from ABCC2/MRP2 mutations causing impaired biliary transport. MRP2 also mediates hepatic elimination of chemotherapeutic agents, yet no data exist on anthracycline-based chemotherapy in adult DJS patients. We report a 26-year-old male with advanced-stage classic Hodgkin lymphoma and DJS harboring a novel homozygous ABCC2 variant (c.2291G>A, p.Gly764Asp). After a pembrolizumab prephase, full-dose BrECADD was administered, and samples were taken for therapeutic drug monitoring (TDM), which demonstrated normal etoposide and doxorubicin pharmacokinetics. Case Report: Light chain amyloidosis in a patient with low-grade B-cell lymphoma.

BACKGROUND: Light chain (AL) amyloidosis is typically associated with plasma cell dyscrasias but can rarely arise from low-grade B-cell non-Hodgkin lymphoma. Renal involvement may present with nephrotic-range proteinuria, acute kidney injury, edema, anasarca, and hypoalbuminemia. Recognizing lymphoma-associated AL amyloidosis is essential because diagnosis requires tissue confirmation, and treatment must target the underlying clonal B-cell disorder. CASE: We report a rare case of severe renal AL amyloidosis in a patient with known low-grade B-cell non-Hodgkin lymphoma previously treated with

N-AVD or BrECADD for Advanced-Stage Classical Hodgkin Lymphoma: Choosing the Right Treatment for the Right Patient.

Frontline therapy for advanced-stage classical Hodgkin lymphoma (cHL) has entered a new era. Two phase III trials have established highly effective but conceptually different strategies: nivolumab plus doxorubicin, vinblastine, and dacarbazine (N-AVD) in SWOG S1826 and PET-adapted BrECADD in GHSG HD21. Because these regimens were tested against different comparators and in different populations, their outcomes should not be interpreted as a head-to-head comparison. This review focuses on translating both trials into individualized treatment selection. N-AVD combines excellent disease control w

An in silico cortico-basal ganglia-thalamic testbed for comparing visual and deep brain stimulation in Parkinson's disease.

INTRODUCTION: Deep brain stimulation (DBS) is an established intervention for Parkinson's disease (PD), whereas the network-level effects of prescribed periodic visual input implemented with 10 Hz base pulses remain uncertain. METHODS: We developed a conductance-based cortico-basal ganglia-thalamic (CBT) model comprising 11 Hodgkin-Huxley neuronal populations and a prescribed periodic visual-input unit (RET) to compare 130 Hz subthalamic nucleus (STN) DBS with the archived periodic visual-input condition on the same calibrated PD network. Primary conditions used N = 10 independent runs each.

A panoramic study of circulating tumor DNA in diffuse large B-cell lymphoma.

Diffuse large B-cell lymphoma (DLBCL) is a frequently encountered aggressive form of non-Hodgkin's lymphoma, characterized by significant clinical and molecular heterogeneity. Owing to inherent limitations of traditional treatments for DLBCL, the advent of circulating tumor DNA (ctDNA) detection, coupled with the integration of multi-omics technologies, has brought about substantial advancements in the diagnosis and treatment of this disease. This review elaborates on the evolution of ctDNA detection and multi-omics integration, its applications in early diagnosis, prognostic model constructio

OGT regulates RNA-binding protein nucleocytoplasmic partitioning, but this fails in amyotrophic lateral sclerosis.

O-linked N-acetylglucosamine transferase (OGT) catalyzes the O-linked β-N-acetylglucosamine (O-GlcNAc) modification to proteins and has been implicated in a range of diseases, including amyotrophic lateral sclerosis (ALS), a rapidly progressive and currently incurable neurodegenerative disorder. However, the precise mechanisms by which OGT influences ALS pathogenesis remain largely unresolved. In this study, we used human-induced pluripotent stem cell (hiPSC)-derived spinal motor neurons (hiPSC-MNs), the principal cell type that undergoes degeneration in ALS, to systematically reveal that OGT-

Increased risk of cancer in patients with mixed connective tissue disease: a population-based cohort study in Korea.

INTRODUCTION: We conducted a nationwide, population-based cohort study to evaluate the risk of cancer in Korean patients with mixed connective tissue disease (MCTD) compared with the corresponding risk in the general population. METHODS: Using the Korean National Health Insurance Service data, we identified 2,094 patients with newly diagnosed MCTD between 2004 and 2020 (16,409 person-years). Standardized incidence ratios (SIRs) were calculated to compare the overall and site-specific cancer risks between patients with MCTD and the age- and sex-matched general population. RESULTS: The overall

Primary Extranodal Nodular Lymphocyte-Predominant Hodgkin Lymphoma of the Cubital Fossa: A Case Report.

Nodular lymphocyte-predominant Hodgkin lymphoma (NLPHL) is a rare B-cell lymphoma that typically presents as localized nodal disease. Primary extranodal involvement, particularly within soft tissue, is exceedingly uncommon and poses diagnostic and management challenges. We report a 25-year-old male who presented with a painless subcutaneous mass in the left cubital fossa. Core biopsy suggested a B-cell lymphoma with a differential diagnosis of NLPHL versus T-cell/histiocyte-rich large B-cell lymphoma (THRLBCL). Comprehensive staging, including bone marrow biopsy and PET-CT, demonstrated isolat

Potential Role of BCL2 and BCL6 Overexpression in Circulating Tumor Cells as Molecular Biomarkers in DLBCL After R-CHOP Therapy.

UNLABELLED: Diffuse large B-cell lymphoma (DLBCL) is the most prevalent non-Hodgkin lymphoma subtype. Although R-CHOP remains the standard first-line therapy, up to 30% of patients experience relapse or refractory disease. Currently, therapeutic monitoring relies on imaging, which is less sensitive than molecular techniques for detecting residual disease. AIM: To evaluate BCL2 and BCL6 oncogene expression in Circulating Tumor Cells (CTCs) as potential molecular biomarkers for clinical outcomes in DLBCL patients after R-CHOP therapy. METHODS: We analyzed BCL2 and BCL6 expression in CTCs from

Global Burden of Disease Study on the Changing Landscape of Hematologic Malignancies and the Growing Impact of Elevated Body Mass Index - Asia, 1990-2023.

WHAT IS ALREADY KNOWN ABOUT THIS TOPIC?: Hematologic malignancies constitute a growing health burden in Asia. Their long-term subregional patterns, future trajectories, and the contribution of high body mass index (BMI) have not been comprehensively evaluated within a unified framework. WHAT IS ADDED BY THIS REPORT?: From 1990 to 2023, leukemia mortality declined across Asia, whereas non-Hodgkin lymphoma and multiple myeloma incidence and mortality increased in most subregions with multiple myeloma showing the fastest growth through 2040. Mendelian randomization suggested a potential associat

Late cardiopulmonary morbidity in patients with Hodgkin lymphoma: a retrospective nationwide registry study.

Hodgkin lymphoma (HL) is a B-cell neoplasm with a cure rate of 90%. Previous data on late morbidity in patients with HL cover the era of older treatment modalities. This real-world study consisted of 2912 patients diagnosed with HL between 2000 and 2019 in Finland and aimed to clarify the incidence of late-onset cardiopulmonary morbidity compared to matched controls. Patient data were collected from four nationwide Finnish healthcare registries. We reported a higher occurrence of coronary artery disease in patients aged < 50 years at diagnosis than in controls (RR 3.80 (2.16-6.74), p < 0001).

[Emerging research progress on Epstein-Barr virus-related clinical diseases].

Epstein-Barr virus (EBV) infects over 95% of the global population and is closely associated with various tumor and non-tumor diseases. In recent years, significant progress has been made in the diagnosis and treatment of EBV related diseases. In terms of diagnosis, the sensitivity and specificity of the new serum marker p85 total antibody in screening high-risk areas for nasopharyngeal carcinoma are significantly better than traditional antibody combination testing, and have been included in the consensus of clinical experts. In terms of treatment, antibody based B-cell depletion therapy (suc

Epstein-Barr Virus Association and Prognosis in Taiwanese Hodgkin Lymphoma: A Nationwide Multicenter Study With Comparison to United States Whites.

Hodgkin lymphoma (HL) is associated with Epstein-Barr virus (EBV) infection in a subset of cases. We previously showed that the incidence of HL in Taiwan increased ~2.5-fold between 2001 and 2020, particularly among young women with the nodular sclerosis (NS) subtype. In this nationwide multicenter study, we evaluated EBV status in HL to determine its impact on clinicopathologic features and survival outcomes. We found that the proportion of EBV-associated HL in Taiwan declined over the past two decades, from 39% to 27%. EBV-positive frequency exhibited a bimodal age distribution, peaking in c

From Divergent Pathogenesis to Precision Medicine: The Road to Targeted Therapy in HIV-Associated and HIV-Unrelated Diffuse Large B-Cell Lymphoma.

Diffuse large B-cell lymphoma (DLBCL) is the most common and biologically heterogeneous subtype of non-Hodgkin lymphoma. While immunochemotherapy has improved outcomes, survival remains variable due to substantial molecular diversity. In people living with HIV (PLWH), DLBCL is the predominant lymphoma subtype and demonstrates more aggressive clinical behaviour, characterised by advanced-stage disease, extranodal involvement, and a potentially poorer prognosis despite antiretroviral therapy. Clarifying differences in the pathogenesis of HIV-associated and HIV-unrelated DLBCL is essential for in
