Phage Henu12-resistant mutant derives fitness trade-offs in Shigella dysenteriae.

UNLABELLED: The rapid emergence of multidrug-resistant Shigella dysenteriae strains necessitates new therapeutic approaches, such as phage therapy. Phages are considered a promising adjunct to antibiotics. The rapid emergence of phage-resistant bacterial mutants represents a major challenge in clinical practice. Nevertheless, this sometimes comes with fitness trade-offs. Thus, understanding the fitness trade-offs is crucial prior to phage clinical application. In the present study, a new lytic S. dysenteriae phage, Henu12, was isolated. Henu12 was assigned to the Caudoviricetes class and demon

Development and qualification of a multiplexed immunoassay to assess the immunogenicity of Shigella vaccines.

BACKGROUND: Shigella is a major cause of diarrheal disease in young children worldwide, particularly in low- and middle-income countries (LMICs). The emergence of antibiotic resistance and limited access to healthcare in LMICs underscores the urgent need for an effective Shigella vaccine. Several next-generation Shigella vaccines are under development, many of which target the O-antigen component of lipopolysaccharide (LPS) from multiple serotypes to achieve broad protective coverage. Reliable, multiplexed assays are needed to evaluate vaccine immunogenicity across Shigella serotypes and vacci

CagA-positive H. pylori may protect against disease of the hands, feet, and mouth.

BACKGROUND: The incidence of an infection with Helicobacter pylori, or H. pylori rises with age, mostly affecting children. The infection rate of Helicobacter pylori increases with age, primarily affecting children. The rate of cases of disease of the hands, feet, and mouth (HFMD), an infectious illness that mostly affects newborns and young children and is ubiquitous throughout the Asia-Pacific area, declines with age. Asthma, shigellosis, TB, anaphylactic disease, and other diarrheal illnesses are all prevented by H. pylori. It also has a strong correlation with infectious disorders brought o

Evaluation of cross-functional antibody responses elicited by a four-component GMMA Shigella vaccine.

Shigella is a major cause of morbidity and mortality in children in low- and middle-income countries. The O-antigen (OAg) component of the lipopolysaccharide is considered a protective antigen, however, their diversity challenges vaccine development, since more than 50 OAg serotypes have been identified. No licensed vaccine against shigellosis is available. A 4-component GMMA-based vaccine, altSonflex1-2-3, delivering S. sonnei and S. flexneri 1b, 2a and 3a OAg has been developed. Coverage is expected against non-vaccine serotypes, due to cross-reactivity, mediated by structural similarities a

Rise of the Resistance: A Case of Resistant Shigellosis in an Immunocompetent Female Patient.

Shigellosisis a common enteric infection with updated empiric treatment guidance due to the emergence of resistant isolates. Herein, we describe a case of resistant shigellosis in an immunocompetent female with no recent travel. The patient initially started ciprofloxacin, but due to ongoing gastrointestinal symptoms and intermediate resistance to fluoroquinolones on stool culture, she was later switched to azithromycin. We believe this case demonstrates the looming threat of multidrug-resistant Shigella, even among those without risk factors or recent antibiotic use. This case also highlights

Immunoinformatics-based design of a next generation multi-epitope vaccine candidate against Shigella boydii using a hierarchical subtractive proteomics approach.

Shigella boydii (strain CDC 3083-94 / BS512) is a Gram-negative facultative anaerobic bacterium that plays a significant role as a pathogen causing bacillary dysentery, especially in developing areas where hygiene is poor. Although less frequently reported compared to other Shigella species, S. boydii still contributes substantially to global shigellosis cases, particularly in low-income endemic regions. Despite its clinical relevance, there has been limited research dedicated on identifying strain-specific antigenic targets for vaccine development. The current study utilized the subtractive p

Climatic drivers of infectious diarrheal disease epidemics in China: An empirical dynamic modeling analysis of 21 million cases.

Infectious diarrheal diseases continue to impose a heavy public health burden in China, despite significant advancements in sanitation and economic development. While existing evidence has linked climate factors to the dynamics of these diseases, the heterogeneous climatic conditions and complex nonlinear interactions among meteorological variables give rise to intricate epidemic patterns that complicate the identification of causal drivers underlying the observed spatial and temporal variability in disease incidence. To address this gap, we conducted a nationwide study across 365 city-level r

Age-dependent acquisition of IgG antibodies to Shigella serotypes-a retrospective analysis of seroprevalence in Kenyan children with implications for infant vaccination.

BACKGROUND: Shigellosis mainly affects children under 5 years of age living in low- and middle-income countries, who are the target population for vaccination. There are, however, limited data available to define the appropriate timing for vaccine administration in this age group. Information on antibody responses following natural infection, proxy for exposure, could help guide vaccination strategies. METHODS: We undertook a retrospective analysis of antibodies to five of the most prevalent Shigella serotypes among children aged <5 years in Kenya. Serum samples from a cross-sectional serosur

Putative correlates of protection against shigellosis assessing immunomarkers across responses to S. sonnei investigational vaccine.

Shigella spp. are a leading bacterial cause of diarrhea. No widely licensed vaccines are available and there is no generally accepted correlate of protection. We tested a S. sonnei Generalized Modules for Membrane Antigen (GMMA)-based vaccine (1790GAHB) in a phase 2b, placebo-controlled, randomized, controlled human infection model study (NCT03527173) enrolling healthy United States adults aged 18-50 years. We report analyses evaluating immune responses to vaccination, with the aim to identify correlates of risk for shigellosis among assessed immunomarkers. We found that 1790GAHB elicited S. s

Prolonged Ileus Due to Underlying Shigella Infection After Bilateral Open Inguinal Hernia Repair.

We present a rare case of prolonged ileus caused by underlying Shigella infection after surgical hernia repair. Infectious disease is an uncommon cause of postoperative prolonged ileus in adults. Our 48-year-old male patient underwent bilateral open inguinal hernia repair and open umbilical hernia repair without complication at an academic institution, with same-day discharge. Eight days later, he presented to the emergency department with complaints of severe cramping abdominal pain, nausea, emesis, and watery diarrhea. Physical examination, computed tomography scan of the abdomen and pelvis,

Testing S. sonnei GMMA with and without Aluminium Salt-Based Adjuvants in Animal Models.

Shigellosis is one of the leading causes of diarrheal disease in low- and middle-income countries, particularly in young children, and is more often associated with antimicrobial resistance. Therefore, a preventive vaccine against shigellosis is an urgent medical need. We have proposed Generalised Modules for Membrane Antigens (GMMA) as an innovative delivery system for Shigella sonnei O-antigen, and an Alhydrogel formulation (1790GAHB) has been extensively tested in preclinical and clinical studies. Alhydrogel has been used as an adsorbent agent with the main purpose of reducing potential GMM

Development of a visual Adhesion/Invasion Inhibition Assay to assess the functionality of Shigella-specific antibodies.

INTRODUCTION: Shigella is the etiologic agent of a bacillary dysentery known as shigellosis, which causes millions of infections and thousands of deaths worldwide each year due to Shigella's unique lifestyle within intestinal epithelial cells. Cell adhesion/invasion assays have been extensively used not only to identify targets mediating host-pathogen interaction, but also to evaluate the ability of Shigella-specific antibodies to reduce virulence. However, these assays are time-consuming and labor-intensive and fail to assess differences at the single-cell level. OBJECTIVES AND METHODS: Here

Investigation of the vaccine potential of an in silico designed FepA peptide vaccine against Shigella flexneri in mice model.

BACKGROUND: Shigellosis is one of the significant causes of diarrhea in Bangladesh. It is a global health problem; approximately 1.3 million people die yearly from Shigellosis. The current treatment method, using different antibiotics against Shigellosis is ineffective. Moreover, it becomes a worrying situation due to the emergence of antibiotic-resistant pathogenic microbes responsible for these diarrheal diseases. METHODOLOGY: Previous immunoinformatics study predicted a potential peptide from the Ferric enterobactin protein (FepA) of Shigella spp. In this study, we have chemically synthesi

The effect of temperature on infectious diarrhea disease: A systematic review.

This study aimed to ascertain the delayed effects of various exposure temperatures on infectious diarrhea. We performed a Bayesian random-effects network meta-analysis to calculate relative risks (RR) with 95 % confidence intervals (95 % CI). The heterogeneity was analyzed by subgroup analysis. There were 25 cross-sectional studies totaling 6858735 patients included in this analysis, with 12 articles each investigating the effects of both hyperthermia and hypothermia. Results revealed that both high temperature (RRsingle = 1.22, 95%CI:1.04-1.44, RRcum = 2.96, 95%CI:1.60-5.48, P < 0.05) and low
