Blunted arginine vasopressin secretion in individuals experiencing a major depressive episode with comorbid post-traumatic stress disorder: Results from an exploratory study using copeptin as a surrogate marker.

Arginine vasopressin (AVP) modulates stress responsivity and social-affective behaviors, but its role in mood and trauma-related disorders remains poorly defined due to challenges in peripheral measurement. This study used copeptin, a stable, reliable, and well-validated surrogate marker of AVP secretion, to assess vasopressinergic function in a transdiagnostic sample of individuals experiencing a major depressive episode (MDE) with and without post-traumatic stress disorder (PTSD), as well as healthy volunteers (HVs). Baseline levels of copeptin, corticotropin-releasing hormone (CRH), adrenoc

Impact of genetic variants on hippocampal volume among individuals with schizophrenia and bipolar disorders.

BACKGROUND AND HYPOTHESIS: Hippocampal volume reduction is a consistent finding in schizophrenia (SCZ) and bipolar disorder type I (BP-I), yet the role of genetic factors remains unclear. We investigated the influence of DISC1 (rs821616), AKT1 (rs1130233), COMT (rs4680), and GSK-3ꞵ(rs334558) polymorphisms on hippocampal morphology. STUDY DESIGN: Seventy-one participants (25 SCZ, 22 BP-I, 24 healthy controls, HC) underwent 1.5T MRI and genotyping. Bayesian multilevel models estimated associations between corrected hippocampal volume, diagnosis, hemisphere, and genetic variants. STUDY RESULTS:

Major depression and atherosclerotic disease: Linking shared genetics to pathways in blood, brain, heart, and atherosclerotic plaques.

The increased risk of atherosclerotic diseases (stroke, coronary artery disease [CAD]) observed in depression may stem from shared pathophysiology. We examined whether: 1) major depression (MD) and atherosclerotic traits share genetic risk, and 2) altered gene expression in various tissues linked to shared genetics has a potential causal role in depression etiology. Data from the largest genome-wide association studies of MD (N = 3,887,532) and 8 atherosclerotic traits (N = 26,909-1,308,460) were used in Two-Sample Mendelian randomization and colocalization to detect cross-trait causal associa

Genome-wide association study of major anxiety disorders in 122,341 European-ancestry cases identifies 58 loci and highlights GABAergic signaling.

The major anxiety disorders (ANX; including generalized anxiety disorder, panic disorder and phobias) are highly prevalent, often onset early and cause substantial global disability. Although distinct in their clinical presentations, they probably represent differential expressions of a dysregulated threat-response system. Here, we present a genome-wide association meta-analysis comprising 122,341 European ancestry ANX cases and 729,881 controls. We identified 58 independent genome-wide significant risk variants and 66 genes with robust biological support. In an independent sample of 1,175,012

A genetic atlas of relationships between circulating metabolites and liability to psychiatric conditions.

Observational studies have reported alteration of circulating metabolites across several psychiatric conditions, but these studies cannot resolve causal relationships. Emerging evidence suggests a genetic relationship exists between these traits requiring further investigation to identify clinically actionable biology. Here, we used the largest genome-wide association studies available to investigate genetic correlation and causal relationships between 10 psychiatric conditions and 249 circulating metabolites. This revealed 1,100 significantly correlated trait pairings, involving fatty acids,

Differentiation between psychotic and non-psychotic major depression by the tabular prior-data fitted network.

BACKGROUND: Misdiagnosing psychotic major depression (PMD) as non-psychotic major depression (NPMD) can lead to poor treatment outcomes. This study aims to develop and validate a machine learning-based model using electronic medical record (EMR) data and the Tabular Prior-data Fitted Network (TabPFN) model to distinguish between PMD and NPMD. METHODS: A total of 666 patients with PMD and 808 patients with NPMD from January 2020 to February 2025 were included. The Least Absolute Shrinkage and Selection Operator (LASSO) algorithm was first applied for feature screening, and the TabPFN model was

Pharmacogenomics of antiepileptic drug mood stabilizer treatment response in bipolar disorder: A MoStGen Consortium study.

Identifying biological and clinical factors associated with response to mood-stabilizing medications is critical for improving bipolar disorder (BD) treatment. The Mood Stabilizer Genomics (MoStGen) Consortium was established to investigate pharmacogenomic and clinical predictors of response to treatment of BD with antiepileptic drug mood stabilizers (AMS). Here we present the first pharmacogenomic analyses of AMS treatment outcomes based on MoStGen Consortium data, including 917 individuals across contributing sites. We performed genome-wide association analyses in subcohorts followed by meta

Causal relationship between mental illness and venous thromboembolism: a Mendelian randomization study.

Previous findings on the relationship between mental disorders and venous thromboembolism (VTE) are controversial, and the precise causal relationship remains unclear. This study investigated the association between mental disorders and VTE based on the analysis of pooled data from large-scale genome-wide association studies (GWAS). Mendelian randomization (MR) was used to assess the causal relationships between bipolar disorder, schizophrenia, and VTE, with inverse variance weighting (IVW) as the primary statistical method. Furthermore, the F-statistic checks instrument stability; Cochran’s Q

In-Depth characterization of the shared genetic architecture of suicide attempts with other major psychiatric disorders.

Suicide is a significant public health problem that usually co-occurs with major psychiatric disorders. Suicidal behaviors have heritability of 30-50%, and the largest genome-wide association studies identified 12 loci linked to suicide attempts (SA). These findings indicate shared genetic architecture among SA and psychiatric disorders. We analyzed public GWAS summary statistics of SA, major depressive disorder (MDD), bipolar disorder (BIP), schizophrenia (SCZ), and attention deficit hyperactivity disorder (ADHD) to quantify genetic overlap using statistical genetics methods: MiXeR for polyge

Single Nucleotide Polymorphisms in Orexin-1 and BDNF Receptor Genes are Associated with Increased Risk of Developing Postpartum Depression among Women with Gestational Diabetes Mellitus.

INTRODUCTION: Global studies have shown a bidirectional association of gestational diabetes mellitus (GDM) with postpartum depression (PPD). Despite high GDM prevalence in Pakistan (3.3%-17.8%), no prior studies have explored its link with PPD. In this study, association between GDM and risk of developing PPD was investigated and risk factors for PPD were identified using the gold-standard Edinburgh Postnatal Depression Scale (EPDS). Evidence suggests that PPD has strong genetic basis. The BDNF gene is a known candidate for PPD pathogenesis, while the orexin system is linked to arousal, energy

The role of sex hormones in severe mental illness: A genetic exploration.

INTRODUCTION: Prominent sex differences exist in severe mental illness (SMI), with increasing evidence pointing towards a pivotal role for sex hormones. Elucidation of these hormonal influences is crucial to tailor sex-specific prevention and treatment. METHODS: To investigate potential shared genetics and bi-directional causal effects between sex hormone traits and SMI (depressive disorder, bipolar disorder and schizophrenia-spectrum disorder), we computed genetic correlations using linkage disequilibrium score regression and bi-directional summary-level Mendelian Randomization (MR). A range

Genomic Study of Resilience to Posttraumatic Stress Disorder in the Million Veteran Program.

BACKGROUND: Resilience following combat exposure is an important factor in understanding posttraumatic stress disorder (PTSD), associated risk, and resilience more generally. Identifying underlying genetic factors requires large samples; most biobanks lack extensive resilience assessments, although data regarding trauma and psychiatric symptoms are frequently present that allow computation of a resilience measure. METHODS: We leveraged the Million Veteran Program cohort to calculate discrepancy-based psychiatric resilience (DBPR) scores by regressing PTSD symptoms (PTSD Checklist for DSM-IV)

Modulatory effects of ketamine on EEG source-based resting state connectivity in treatment resistant depression.

Treatment-resistant depression (TRD) accounts for approximately 30% of major depressive disorder cases and has been characterized by altered functional connectivity within and between the Default Mode (DMN) and Frontoparietal networks (FPN). Ketamine can be an effective treatment for TRD, and its antidepressant response has been associated with alterations in resting state functional connectivity (rsFC). Here, we evaluated the effect of a single subanesthetic dose of racemic ketamine (0.5 mg/kg) on electroencephalogram (EEG) derived source-based measures of rsFC from 24 participants with TRD (

Leveraging the genetics of psychiatric disorders to prioritize potential drug targets and compounds.

Genetics can inform biologically relevant drug development and repurposing, which may improve patient care. Here, we leverage the genetics of psychiatric disorders to prioritize potential drug targets and compounds. We used the genome-wide association studies of four psychiatric disorders [attention deficit hyperactivity disorder (ADHD), bipolar disorder, depression, and schizophrenia] and genes encoding drug targets. We conducted drug enrichment analyses incorporating the novel and biologically specific GSA-MiXeR tool. We conducted multiple molecular trait analyses using large-scale transcrip
