Trans-vatabnet: transcriptomic signatures for Parkinson's disease detection using deep learning.

Background: Early detection of Parkinson's disease (PD) is essential for timely clinical intervention. However, transcriptomic-based diagnosis faces challenges associated with high-dimensional gene expression data, redundant features, class imbalance, and limited interpretability. Objective: This study proposes TransVaTabNet, an integrated framework for PD detection and biomarker identification using transcriptomic analysis, deep learning, and explainable artificial intelligence. Methods: RNA-seq data were preprocessed using low-expression gene filtering, Trimmed Mean of M-values (TMM) normali

Generation of a CRISPR/Cas9-corrected isogenic control iPSC line (NIMHi001-A-1) from a Parkinson's disease Patient-Derived iPSC line carrying the LRRK2 I1371V mutation.

We report NIMHi001-A-1, a CRISPR/Cas9-corrected isogenic iPSC line derived from a patient line carrying a heterozygous LRRK2 missense mutation GRCh38 chr12:g.40308618A > G [(MANE transcript NM_198578.4:c.4111A > G); p.Ile1371Val] associated with familial Parkinson's disease. Sanger sequencing confirmed successful correction. The line retained iPSC morphology and expressed pluripotency markers (OCT4, NANOG, SOX2, TRA-1-60, SSEA4) by immunofluorescence, gene expression analysis, and flow cytometry. Normal karyotype (46, XY) and complete genetic identity with the parental line were established by

Disarming CH25H to clear α-synuclein.

α-Synuclein (α-Syn) accumulation is a defining feature of Parkinson's disease (PD). Zhang et al. identify cholesterol 25-hydroxylase (CH25H) as an astrocytic inhibitor of p62-dependent α-Syn degradation. Disrupting the CH25H-p62 interaction facilitates α-Syn clearance via autophagy and improves PD-like phenotypes, suggesting a previously unrecognized nonenzymatic function for CH25H with therapeutic potential.

Toward an Articulatory Weakness Index for Speech Kinematics in Parkinson's Disease.

Many acoustic speech metrics often reflect a combination of respiratory, laryngeal, and articulatory contributions, thereby preventing direct measurement of articulatory dynamics. We have developed an articulatory weakness index (AWI) derived from electromagnetic articulography (EMA) or acoustic data. We learn an articulatory-state coordinate Z a from healthy EMA data, where higher values indicate restricted articulator motion. In cohorts of individuals with Parkinson's disease (PD) and controls, AWI is elevated in PD and correlates with motor severity, while remaining stable across in heal

The A53T Parkinson's Disease Mouse Model Shows Abnormal Outer Retinal OCT Metabolic Biomarker Patterns.

PURPOSE: The purpose of this study was to test for abnormal optical coherence tomography (OCT) metabolic biomarker patterns in the A53T Parkinson's disease model, which accumulates α-synuclein in photoreceptor inner segments before overt motor symptoms. METHODS: Four-to-6-month-old transgenic A53T homozygous mice and B6C3H wild-type (WT) littermate controls were evaluated using 2 OCT metabolic biomarkers. The first was the external limiting membrane (ELM) to retinal pigment epithelium (RPE) thickness, a proxy for subretinal space acidification and retinoid deficiency. The second biomarker was

Automated Quantification of Parkinsonian Facial Expressions via a Transformer-Based Multi-View Multimodal Fusion Framework.

Facial hypomimia, a common symptom of Parkinson's disease (PD), is characterized by reduced facial expressiveness due to muscle rigidity. Its clinical assessment, typically based on the Movement Disorder Society-Unified PD Rating Scale, is challenging due to the limited clinician availability, subjectivity, and time consuming procedures. In this paper, we propose a transformer-based multi-view multimodal fusion framework for efficient and objective video-based automated assessment of PD facial expressions. The framework incorporates facial anatomical priors, jointly models global and local fea

Noninvasive brain stimulation for motor function and quality of life in Parkinson's disease: a systematic review and network meta-analysis.

BACKGROUND: Parkinson's disease (PD) seriously affects the motor function and quality of life of patients. Noninvasive brain stimulation (NIBS) is an effective non-pharmacological approach to improve the motor function and quality of life of PD patients, but the views about the optimal NIBS intervention remain controversial. This study aimed to compare and rank the relative efficacy of different NIBS interventions on motor function and quality of life. METHODS: Eight databases, including PubMed, Cochrane Library, Embase, Web of Science, SinoMed, CNKI, Wanfang, and VIP Database, were systemati

A comprehensive review of the neurobehavioral and neurotoxic effects of 6PPD-Q in fish, rodents, and humans.

N-(1,3-dimethylbutyl)-N'-phenyl-p-phenylenediamine (6PPD) is a widely used tire additive that undergoes environmental transformation to 6PPD-quinone (6PPD-Q), which is ubiquitously detected in environmental matrices. Increasing evidence demonstrates that exposure to 6PPD-Q adversely affects organisms through neurobehavioral disruption and neurotoxicity. While studies document behavioral abnormalities and molecular alterations, a comprehensive synthesis linking these outcomes remains limited. The objective of this review was to synthesize current literature reporting neurobehavioral and neuroto

Epigenetic processes in Parkinson disease.

Numerous investigations performed over the last 20 years revealed various epigenetic modifications in both postmortem brain samples and in peripheral tissues of individuals with Parkinson disease (PD), as well as in neuronal and animal models of the disease. These include changes in DNA methylation and hydroxymethylation, histone-tail modifications, and a dysregulated expression of both short- and long-noncoding RNA molecules. Overall, there is no doubt that epigenetic modifications occur within the neurodegenerative process linked to PD, resulting in altered expression of PD-linked genes. How

Visual feedback increases expiratory muscle training effort in Parkinson's disease: a randomised crossover trial.

BACKGROUND: Disorders of airway protection, including impaired cough and swallowing, are clinically important complications of Parkinson's disease. Expiratory muscle strength training (EMST) is an established non-pharmacological intervention targeting these deficits. The aim of this study was to evaluate the acute effect of visual feedback provided by the SpiroGym mHealth app on expiratory muscle activation during EMST in people with Parkinson's disease. METHODS: We conducted a single-center, randomised, two-period crossover trial in people with Parkinson's disease. Participants performed EMS

Longitudinal Changes in the Digitized Archimedes Spiral Drawing Test Following iTBS of the Left DLPFC in Parkinson's Disease: An Exploratory Feasibility Study.

Background: The Digitized Archimedes Spiral Drawing Test (DAST) provides objective assessment of fine motor function, but its utility as a biomarker for neuromodulatory interventions in Parkinson's disease (PD) remains underexplored. Objective: To explore longitudinal changes in a comprehensive set of DAST parameters following a course of intermittent theta-burst stimulation (iTBS) of the left dorsolateral prefrontal cortex (DLPFC) in PD patients as a feasibility study. Methods: Twenty-seven participants (17 PD patients, 10 healthy controls) underwent DAST testing at baseline and at 1.5 and 6

Interaction Between Cigarette Smoking and PARK16 on Parkinson's Disease.

Parkinson's disease (PD) is influenced by genetic and environmental factors. Epidemiological evidence indicates that smoking is associated with reduced PD risk, potentially through oxidative stress-related pathways. PARK16 is a PD-susceptibility locus also implicated in oxidative stress regulation. This study examined gene-environment interaction involving smoking and a PARK16 single nucleotide polymorphism (SNP). A case-control study involving 999 Chinese subjects (497 cases, 502 controls) was conducted. PARK16 SNP rs6679073 (NC_000001.11:g.205787356C>A) was identified in a genome-wide associ

Elevated erythrocyte membrane pS42-α-synuclein in Parkinson's disease and its changes following plasma cross-incubation.

BACKGROUND: Blood biomarkers for pathological α-synuclein in Parkinson's disease (PD) are confounded by hemolysis and hemoglobin. Erythrocyte membrane-bound α-synuclein may overcome these challenges. OBJECTIVE: To investigate whether erythrocyte membrane-bound α-synuclein is altered in PD and whether it changes following plasma cross-incubation. METHODS: Erythrocyte membranes from patients with PD and healthy controls were isolated and analyzed by phosphoproteomics to identify phosphorylation sites. Digital western blotting was used for targeted quantification of α-synuclein species. Plasma

Modulating neuroinflammation with glucagon-like peptide-1 receptor agonists: a scoping review exploring their therapeutic potential in schizophrenia.

Neuroinflammation is a key contributor to the pathophysiology and symptomatology of schizophrenia spectrum disorders. Glucagon-like peptide-1 receptor agonists (GLP-1RAs), clinically approved anti-diabetic and -obesity medication, have been associated with anti-inflammatory properties. It remains unclear whether GLP-1RA treatment can improve neuroinflammation within psychiatric disorders, including schizophrenia. This scoping review synthesizes evidence from clinical and preclinical studies on the anti-inflammatory effects of GLP-1RAs in psychiatric, neurodegenerative and neurological disease
