Locus coeruleus abnormalities on neuromelanin-sensitive MRI in Parkinson's disease: a systematic review and meta-analysis.

Neuromelanin-sensitive magnetic resonance imaging (NM-MRI) enables in vivo assessment of locus coeruleus (LC) integrity, but findings in Parkinson's disease (PD) vary across cohorts and methods. We searched PubMed/MEDLINE, Embase, Web of Science Core Collection and Scopus from inception to 5 September 2026 and pooled independent PD-healthy control (HC) comparisons using random-effects models. Sixty-five reports were included, of which 17 contributed to at least one main pooled outcome. The primary LC-signal analysis included 14 independent cohorts (1,078 participants with PD and 543 HCs) and s

MiR‑204‑5p modulates glial morphology and apomorphine‑induced rotational asymmetry without dopaminergic neuroprotection in a preclinical mouse model of Parkinson's disease.

Parkinson's disease (PD) is characterized by progressive nigrostriatal dopaminergic degeneration and a dysregulated neuroinflammatory environment. Here, we investigated whether intranigral delivery of a microRNA‑204‑5p mimic (miR‑204-5p) at the time of lesion modulates dopaminergic‑dependent rotational asymmetry, glial morphology, and striatal medium spiny neurons (MSNs) recruitment in a unilateral 6‑OHDA mouse model. Adult C57BL/6 mice received a unilateral striatal 6‑OHDA injection and simultaneous intranigral infusion of a synthetic miR‑204‑5p mimic or vehicle, and apomorphine‑induced rotat

Prevalence of alpha-synuclein seeding activity in the Olfactory Mucosa following SARS-CoV-2 infection: results from a pilot study.

Olfactory dysfunction (OD) is a hallmark of SARS-CoV-2-infection where it is believed to result from neuroinflammation induced by viral invasion via the olfactory mucosa (OM). Hyposmia also affects the majority of patients with Parkinson's disease (PD) where it is caused by α-synuclein (aSyn) pathology in the olfactory system, and aSyn-seeding-activity can be detected in the OM using aSyn-seeding-amplification-assays (aSyn-SAA). The present pilot study aimed to investigate whether SARS-CoV-2-infection induces local aSyn-misfolding and -aggregation in OM samples using aSyn-SAA. We conducted an

Longitudinal olfactory and neuropsychological trajectories across the GBA1 spectrum.

Tracking non-motor symptoms and neuropsychological function is critical for assessing neurodegenerative progression. We characterized differences among individuals with Gaucher disease, GBA1 variant carriers and GBA1-associated Parkinson disease to identify candidate markers of Parkinson disease-related change in Gaucher disease or GBA1 variant carriers. We analysed data from 161 adults evaluated at the National Institutes of Health between 1983 and 2024, including 79 with Gaucher disease, 52 GBA1 variant carriers and 30 with GBA1-associated Parkinson disease. Assessments included questionnair

How system justification sustains sexual inequality: The case of the orgasm gap.

INTRODUCTION: Parkinson's disease (PD) is the second most common neurodegenerative disorder after Alzheimer's disease, and is characterized by motor symptoms such as bradykinesia, rigidity, and reduced movement automaticity. Caregiver contributions (CC) to self-care in PD represent an important resource for supporting people with PD. However, this topic has not yet been fully explored. AIM: To explore CC to self-care in PD across the domains of CC to self-care maintenance, monitoring, and management. METHOD: A qualitative study design was conducted using a combined deductive and inductive ap

Deep brain stimulation as neuroimmune-metabolic modulation in Parkinson's disease and beyond: tissue response, glial remodeling, and disease modification.

Deep brain stimulation (DBS) is an established symptomatic therapy for Parkinson's disease (PD), yet its biological effects extend beyond modulation of abnormal basal ganglia circuitry. This review conceptualizes DBS in PD as a neuroimmune-metabolic intervention acting across the electrode-tissue interface, glial networks, peripheral immunity, and activity-dependent metabolic systems. PD is characterized by convergent immune and metabolic vulnerability. These processes create a disease environment in which electrical stimulation may interact with inflammation, excitotoxicity, bioenergetic stre

An examination of the effect of dual task on gait in Parkinson's disease and REM sleep behavior disorder.

BACKGROUND: Gait variability is a hallmark of Parkinson's disease (PD) and has been linked to cognitive deficits and fall risk. Rapid eye movement sleep behavior disorder (RBD) is a strong predictor of synucleinopathies, yet evidence for gait changes in RBD is inconsistent. Performing a dual task increases gait variability, an effect that can be quantified using a cost function. OBJECTIVE: Determine the degree to which dual task cost differs between control, RBD, and PD participants at baseline, and between RBD converters versus non-converters at follow-up. METHODS: 46 RBD, 23 control, and 1

Epidemiological comparison of cirrhosis to other long-term health conditions in the United Kingdom.

BACKGROUND: Although cirrhosis is a serious long-term health condition (LTC), it tends to receive little attention from healthcare policymakers. Here, we sought to describe how cirrhosis compares to other LTCs to inform priority-setting discussions. METHODS: We analysed data from the Our Future Health (OFH) study, a cohort of ∼1.7 million individuals from England, United Kingdom. Self-reported diagnosis status for cirrhosis and 15 benchmark LTCs (including type 1 diabetes, type 2 diabetes, Parkinson's disease, chronic kidney disease, schizophrenia, and stage 4 cancer) were ascertained from th

Parkinson's Disease-Associated Genetic Alterations Disrupt Behavior and Diurnal Activity in Drosophila melanogaster.

Parkinson's disease (PD) is a progressive neurodegenerative disorder characterized by the loss of dopaminergic neurons, resulting in motor dysfunction and a variety of non-motor symptoms. Multiple genetic alterations, including increased α-synuclein expression and loss-of-function of parkin, PINK1, and DJ-1, have been associated with PD pathogenesis; however, their effects on stress tolerance, motor behavior, and daily activity remain incompletely understood. This study investigated the effects of PD-associated genetic alterations on stress tolerance, motor behavior, and diurnal activity in Dr

Genome-wide cross-trait analysis to compare genetic architecture between Parkinson's disease and kidney-related traits.

Epidemiological studies link kidney function to Parkinson's disease risk, but the genetic basis of this relationship remains unclear. Here, we leverage genome-wide association data for Parkinson's disease and five UK Biobank-derived kidney-related traits in individuals of European ancestry, combining genetic correlation, pleiotropy mapping, colocalization and enrichment across sex-combined and sex-stratified analyses. Genome-wide genetic correlations with Parkinson's are modest, with only a significant correlation with urinary potassium-to-creatinine ratio significant, yet we identify shared l

Dysregulated cholesterol trafficking through ABCA1 drives neurodegeneration in Parkinson's disease.

Although brain cholesterol dysregulation is implicated in Parkinson's disease (PD), targeting cholesterol synthesis has shown limited efficacy. Here we identify ATP-binding cassette transporter A1 (ABCA1) as a critical regulator of cholesterol homeostasis in PD. We show that ABCA1 expression is elevated in the substantia nigra of postmortem PD brains and in a PD mouse model, with regional analyses revealing preferential alterations in nigral and striatal circuits. Mechanistically, astrocytic ABCA1 drives excessive cholesterol efflux in mice, promoting neuronal cholesterol overload. In neurons,

Parkinson's disease LRRK2 mutations dysregulate iron homeostasis and promote oxidative stress and ferroptosis in human neurons and astrocytes.

BACKGROUND: Iron accumulation is a hallmark of sporadic and familial Parkinson's disease (PD) and correlates with clinical motor symptom severity. The biochemical mechanisms driving iron dyshomeostasis in PD brain and whether these are early or late events in the neurodegenerative process remain unknown. Nigral iron levels in LRRK2-PD patients have been reported to be even higher than in idiopathic PD, and greater in non-manifesting LRRK2 carriers than controls, suggesting that iron accumulation precedes clinical onset in LRRK2-associated PD. However, the cells affected and mechanisms governin

Modifications to cognitive behavioural therapies for depression and anxiety in adults with neurological disorders: A qualitative analysis of intervention protocols.

Cognitive and Behavioural-based Therapies (CBT) can effectively manage symptoms of depression and anxiety. However, adults with neurological disorders (NDs) often face cognitive, physical, and social challenges that make it difficult to engage with traditional forms of these treatments. To address this, CBT protocols have been modified to better suit the needs of this population. This study analysed 41 randomized controlled trials with modified CBT-based protocols for treating symptoms of depression and/or anxiety in adults with a ND. It spanned 10 different NDs : stroke, epilepsy, migraine, P

Training Non-Neurologist Healthcare Professionals in the Recognition and Management of Movement Disorders in Malawi.

BACKGROUND: Many patients with neurological disorders in Malawi are seen by non-neurologist healthcare professionals due to the low number of neurologists available in the country. Movement disorders are a growing concern at neurology clinics and patients often present late, having spent years before getting a neurology referral. In line with the WHO's global action plan on improving neurological care, we organized a workshop for non-neurologist healthcare professionals on the clinical overview of common movement disorders. METHODS: The 2-day in-person workshop was held in Blantyre city and i

Daily regulation of glucocerebrosidase activity and impact of fixed-time rotarod training in a mouse model of Gba1-associated Parkinson's disease.

INTRODUCTION: Mutations in the glucocerebrosidase (GBA1) gene are a major genetic risk factor for Parkinson's disease (PD) and result in dysfunctional glucocerebrosidase (GCase), the lysosomal enzyme responsible for cleaving glucosylceramide into glucose and ceramide. The pathogenic L444P variant disrupts the enzyme structure and activity, leading to lysosomal accumulation of glucosylceramide and ultimately impairing the clearance of monomeric α-synuclein (α-syn). Given the functional crosstalk among lysosomal homeostasis, the molecular circadian clock, and physical activity, we investigated w

Hierarchical reorganization of cortical and cerebellar functional gradients in Parkinson's disease.

INTRODUCTION: Parkinson's disease (PD) is a common movement disorder with heterogeneous clinical manifestations, and its tremor-predominant presentations may overlap with essential tremor (ET), complicating clinical differentiation in some patients. Functional gradient analysis provides a continuous framework for characterizing macroscale brain hierarchical organization and may offer candidate imaging features for disease characterization. METHODS: We first investigated surface-based resting-state functional measures and cerebral perfusion in 50 patients with idiopathic PD and 33 healthy cont

Rotigotine transdermal patch in Parkinson's disease: a narrative review of pharmacology, clinical evidence, special-population management, and health economics.

BACKGROUND: Parkinson's disease (PD) requires lifelong dopaminergic therapy, yet oral routes are frequently compromised by gastrointestinal dysfunction, dysphagia, and perioperative fasting. Abrupt withdrawal of oral dopaminergic drugs may precipitate severe motor deterioration or parkinsonism-hyperpyrexia syndrome. Rotigotine transdermal patch delivers 24-h continuous dopaminergic stimulation (CDS) via a non-ergot dopamine agonist (DA) formulation that bypasses first-pass metabolism and gastrointestinal absorption, and may be particularly useful when oral dopaminergic delivery becomes unrelia
