An integrative clinical and bioinformatic analysis identifies MicroRNAs as biomarkers of ischemic stroke severity.

Identifying reliable circulating biomarkers is crucial for improving the diagnosis and risk stratification of patients with ischemic stroke. In this study, we evaluated several whole-blood circulating miRNAs (miR-106b-5p, miR-16-5p, miR-15b-5p, let-7e-5p, and miR-125a-3p/-5p) to determine their diagnostic and disease severity in acute ischemic stroke (AIS). Sixty AIS patients and thirty age- and sex-matched controls were included. Whole-blood miRNAs were quantified at admission and on day 7. Statistical analyses included ROC curves, multivariate logistic regression, and SHAP-based machine lear

Neuroprotective Mechanisms of Hyperbaric Oxygen Therapy in Cerebral Ischemia-Hypoxia Injury Following Cardiopulmonary Resuscitation.

Despite significant advancements in cardiopulmonary resuscitation (CPR) techniques, the global burden of sudden cardiac death remains high, with post-CPR survival rates persistently below 8%. Hypoxic-ischemic brain injury (HIBI) is the predominant cause of mortality, accounting for 68% of fatalities following resuscitation. Hyperbaric oxygen (HBO) therapy, which enhances oxygen dissolution in plasma, has demonstrated efficacy in focal cerebral ischemia conditions such as stroke. However, its potential in addressing global cerebral ischemia following CPR-a condition pathophysiologically distinc

Quantitative and morphological analyses reveal microglia-specific neuroinflammation in brain regions linked to cognitive function in ovariectomized and chronic intermittent hypoxia-exposed female rats.

Women affected by obstructive sleep apnea (OSA) face an increased risk of cognitive impairment and mood disorders, with emerging evidence suggesting that neuroinflammation plays a significant role in the pathophysiology. OSA causes intermittent hypoxia and activates the immune response in the brain. Additionally, menopause is a separate risk factor that can put women with OSA at an even higher risk of cognitive decline. Although neuroinflammation occurs in OSA, the underlying mechanisms of the neuroinflammatory response are not well understood, and it remains unknown whether ovarian hormones m

Acute blood biomarker responses to consensual sexual choking/strangulation in young adult women: a randomized crossover study.

INTRODUCTION: Sexual strangulation, commonly referred to as "choking", has become increasingly common among young adults, yet its neurobiological consequences remain poorly understood. Preclinical and clinical evidence suggests strangulation may trigger axonal injury, neuroinflammation, and blood-brain barrier dysfunction. Blood biomarkers of neural injury and inflammation provide a sensitive means to detect subtle effects. AIM: To examine whether consensual sexual choking/strangulation acutely alters blood biomarkers of neural injury and inflammation compared to non-choking sexual activity i

Racial, ethnic and sex-specific mechanisms of obstructive sleep apnea and Alzheimer's disease risk.

BACKGROUND: Obstructive sleep apnea (OSA) is associated with Alzheimer's disease (AD) risk. Racial-, ethnic-, and sex-specific mechanisms of OSA and AD risk were examined. METHODS: We analyzed data from 3978 polysomnography patients without cognitive decline aged ≥ 60 including 663 OSA+ patients (284 non-Hispanic White, 207 Black, 172 Hispanic) matched to OSA- cohorts (1:1, n = 663; 1:4, n = 2652) and followed for AD through 2013. RESULTS: During the 8.5 (standard deviation 1.4) year period, 358 patients developed AD. AD risk was higher for Black (adjusted hazard ratio [aHR] 2.24 [1.24-2.71]

Influence of the hydroxytyrosol/triterpenes ratio on the neuroprotective effect of olive oil in an experimental model of diabetes mellitus.

The aim of the study is to analyse the possible neuroprotective effect of two types of olive oils, one with a high content of alcoholic phenols (POO) and the other with a high content of alcoholic phenols and triterpenes (PTOO), in an experimental model of diabetes mellitus. We also aimed to assess the participation of triterpenes, alone or in association with hydroxytyrosol, in the neuroprotective effect of these olive oils. We administered 0.5 mL/kg/d of both oils to type 1 diabetic animals and performed a hypoxia-reoxygenation model in brain slices. Both oils reduced cell death in this expe

Using Near Infrared Light to Detect Hypoxia in Retinal Images.

INTRODUCTION: Acute hypoxia poses a serious risk in military aviation because it can lead to impaired performance of safety-critical tasks. Early detection of hypoxia is therefore critical. Oxygen saturation (Spo2) of the retinal blood vessels may serve as a surrogate biomarker for brain hypoxia. This study investigates the feasibility of detecting hypoxia through retinal imaging in the near-infrared (NIR) spectrum, which does not interfere with the pilot's vision. METHODS: There were 12 subjects who participated in a hypobaric flight simulation to lower systemic Spo2. Retinal images were cap

Use of Laser Speckle Contrast Imaging for Distribution of Animals by Severity of Brain Tissue Damage in a Neonatal Hypoxia-Ischemia Model in Mice.

Background/Objectives: Inter-individual variability in injury severity represents a major barrier to reproducibility in neonatal hypoxia-ischemia (HI) models. Objective early postoperative stratification of animals is therefore essential for standardized group allocation and reliable assessment of experimental outcomes. This study aimed to evaluate whether laser speckle contrast imaging (LSCI) can be used as a rapid, noninvasive tool for early post hoc stratification of ischemic brain damage severity in neonatal mice following HI. Methods: Neonatal CD-1 mice (postnatal day 9; n = 60) underwent

In Vitro Analysis of TWEAK/Fn14 Axis in the Blood-Brain Barrier Models during Oxygen-Glucose Deprivation and Reoxygenation.

Upregulation of tumor necrosis factor-like weak apoptosis-inducing factor (TWEAK) and its receptor fibroblast growth factor-inducible 14 (Fn14) was observed in stroke patients and murine models, contributing to neuronal apoptosis and blood-brain barrier (BBB) disruption. This study aimed to investigate the TWEAK/Fn14 signaling axis in cerebral ischemia and reperfusion using different in vitro oxygen-glucose deprivation (OGD) durations and cellular models. Western blot and RT-qPCR were used to evaluate TWEAK/Fn14 expression in monocultures, co-cultures, and triple-cultures of human immortalized

lncRNA LUCAT1 regulates DNA damage response in glioma stem cells under hypoxia.

BACKGROUND: Genomic stability is essential for cell survival, particularly under stress conditions like hypoxia, which disrupt DNA repair. Glioblastoma (GBM) is markedly resistant to DNA-damaging therapies, primarily due to glioma stem cells (GSCs) and the hypoxic tumor microenvironment. Long non-coding RNAs (lncRNAs) play a significant role in maintaining genome integrity, but their involvement in the DNA damage response (DDR) under hypoxic conditions in GSCs remains unclear. We previously identified LUCAT1 as the most highly induced lncRNA in GSCs under hypoxia and a key regulator HIF1α acti

Targeting hypoxic exosomal IGFBP2 overcomes CD47-mediated immune evasion in glioblastoma.

Glioblastoma (GBM) acquires malignant traits through complex molecular adaptations that sustain immune evasion, often characterized by hypoxia and overexpression of the phagocytosis checkpoint CD47. However, the role of hypoxic drivers coordinating CD47-dependent immune evasion remains poorly defined. Here, we integrated single cell RNA sequencing and proteomic analysis to identify that insulin-like growth factor binding protein 2 (IGFBP2) was co-expressed with CD47 in hypoxic mesenchymal-like GBM subpopulations, synergistically promoting tumor progression and immune evasion. Mechanically, hyp

HIF-1α/BNIP3-mediated mitophagy mitigates cerebral ischemia/reperfusion injury in rats by suppressing NLRP3 inflammasome activation.

BACKGROUND: The hypoxia-inducible factor-1α (HIF-1α)/Bcl-2/adenovirus E1B 19-kDa interacting protein (BNIP3) pathway, a key regulator of mitophagy, has demonstrated protective effects in renal and cardiac ischemia/reperfusion (I/R) injury. However, its specific role and mechanism in cerebral I/R injury (CIRI) are not fully understood. METHODS: An oxygen-glucose deprivation/reoxygenation (OGD/R) model in SH-SY5Y neuroblastoma cells and a transient middle cerebral artery occlusion (MCAO) model in rats were established to simulate CIRI. HIF-1α overexpression plasmids was then introduced into the

Identification of Glioma Phenotypic Subtypes From Multimodal MRI Data Using Hierarchical Multi-Kernel Learning.

BACKGROUND: Gliomas are the most common primary brain tumors, exhibiting significant phenotypic variability even within the same grade. Identifying glioma subtypes through non-invasive methods could improve patient management. METHODS: In this study, we applied hierarchical multi-kernel learning to identify glioma phenotypic subtypes using MRI data (T1CE and T2FLAIR) from the First Hospital of Shanxi Medical University (FHSXMU) and Shanxi Provincial People's Hospital (SPPH) (n = 246). We further validated our findings using an independent dataset of similar tumor characteristics from The Canc

A multifunctional and ROS response CO-gas delivery platform for spinal cord regeneration.

Spinal cord injury (SCI) initiates a cascade of pathological events in which neuroinflammation and hypoxia critically impair functional recovery. Although exogenous carbon monoxide (CO) exhibits anti-inflammatory potential, its translation has been hindered by the lack of a safe and effective delivery strategy. Here, we report the development of a multifunctional therapeutic platform (COPH), composed of CO-releasing molecules-401 (CORM-401) as the CO donor, peptide dendrimer nanogels (PDNs) as the carrier, and hyaluronic acid (HA) as a microglia-targeting ligand. COPH selectively accumulates i

Riluzole preserves brain endothelial integrity in ischemic stroke via SLC7A11-dependent GPX4 and HIF-1α/VEGFA signaling.

Riluzole, an FDA-approved neuroprotective agent, was investigated for its therapeutic potential in ischemic stroke. Transcriptomic profiling of human brain microvascular endothelial cells (hBMECs) subjected to oxygen-glucose deprivation/reperfusion (OGD/R) identified a pivotal role for the cystine/glutamate antiporter SLC7A11. We found that riluzole activates SLC7A11, thereby triggering a dual protective mechanism: it strengthens cellular antioxidant capacity by upregulating GPX4 while simultaneously enhancing proangiogenic signaling through the HIF-1α/VEGFA pathway. Consequently, riluzole att

Neutrophils in glioblastoma: orchestrators of the tumor microenvironment and immune evasion.

Glioblastoma (GBM) is the most aggressive primary brain tumor in adults and remains refractory to current therapies. Beyond profound immunosuppression, GBM is characterized by a complex tumor microenvironment (TME) in which neutrophils have emerged as critical yet understudied regulators of tumor progression and immune evasion. Tumor-associated neutrophils (TANs) display marked functional plasticity, acquiring pro-tumor or anti-tumor phenotypes depending on microenvironmental cues. GBM recruits and reprograms infiltrating neutrophils through chemokine-driven trafficking, hypoxia, and tumor-der

NADPH exerts neuroprotection in ischemic stroke by reinforcing blood-brain barrier integrity and stimulating angiogenesis.

Ischemic stroke (IS) represents a significant global health burden with increasing incidence, creating an urgent need for novel therapeutic approaches. This study explored the neuroprotective effects of nicotinamide adenine dinucleotide phosphate (NADPH) in preserving blood-brain barrier (BBB) integrity and promoting angiogenesis after IS. Through network pharmacology analysis and molecular docking, five key molecular targets of NADPH in IS were identified: HIF-1α, SRC, NLRP3, CASP3, and AKT1. In vivo, NADPH treatment conferred significant protection against cerebral ischemia in the transient
