BACKGROUND AND OBJECTIVES: Oligodontia, the congenital absence of multiple permanent teeth, is frequently linked to LRP6 variants. However, the genotype-phenotype correlations remain unclear and non-European cohorts are underrepresented. The objectives of this study were to: (1) characterize the molecular and clinical features of LRP6 variants identified in unrelated Thai individuals with tooth agenesis; (2) conduct a scoping review of previously published cases; and (3) refine the understanding of LRP6 genotype-phenotype correlations.
MATERIALS AND METHODS: A detailed case series analysis of

Patients with hypodontia often undergo orthodontic treatment, with single tooth pontics often used to replace missing teeth. However, single tooth pontics can often rotate on the archwire, leading to compromised stability. 'Blocks of pontics' can provide prosthetic replacement during orthodontic treatment for patients with hypodontia, overcoming common issues with large edentulous spans. After impression taking, blocks of pontics can be waxed up and finished in acrylic. These can then be ligated to the archwire intraorally. Blocks of pontics can be life-changing, restoring adequate aesthetics

Congenital hypogonadotropic hypogonadism (СНH) is a group of diseases caused by impaired synthesis or secretion of gonadotropin-releasing hormone (GnRH) and gonadotropin hormones. At present, more than twenty genes involved in the development of СНН have been described. In the structure of HGH, the most common forms of the disease are caused by pathogenic variants in genes involved in the ontogenesis, migration and survival of GnRH neurons, whereas pathology of genes involved in the action/transmission of GnRH signals in normally developed GnRH neurons is less common. This article describes a

This study aimed to sequence the PAX9 and MSX1 genes in a group of Jordanian patients diagnosed with non-syndromic hypodontia. The study also examined whether the identified genetic variants in these key odontogenic genes could be used to assess susceptibility to hypodontia. Fifty Jordanian individuals diagnosed with non-syndromic hypodontia were enrolled in this study. Genomic DNA was extracted, and all exons of PAX9 (Exons 1,2, 3, and 4) and MSX1 (Exons 1 and 2) were amplified using PCR. The Sanger sequencing technique was used to detect genetic variants, which were analyzed for their clinic

INTRODUCTION: Management of missing anterior teeth can pose significant challenges for the orthodontist, as the maxillary incisors are key to esthetic orthodontic treatment planning. Two treatment modalities for patients that are unilaterally missing a maxillary lateral incisor that offer definitive functional and esthetic natural-tooth replacements for patients, regardless of age, are orthodontic canine substitution (OCS) and autotransplantation (AT) of a lower incisor. Using validated indices and an electronic questionnaire, the purpose of the study was to assess, objectively and subjectivel

Hypohidrotic ectodermal dysplasia (HED), caused by mutations in genes such as EDAR, is a genetic disorder characterized by hypodontia, hypotrichosis, and hypohidrosis. Dental anomalies in HED patients lead to functional and aesthetic impairments, necessitating multidisciplinary interventions. This case report highlights the role of combined orthodontic and prosthodontic approaches in managing HED-related dental deficiencies. CASE PRESENTATION: This clinical report describes the multistage treatment of a 32-year-old woman diagnosed with a dominant case of ED with c.175-2(IVS 3)A > G mutation of

The aims of this retrospective radiographic study were to assess the occurrence of dental anomalies in children and patients with disabilities and to investigate the association of these anomalies with pre-, peri-, and post-natal medical history. A total of 289 panoramic radiographs of children (0-14 years) and patients with disabilities (including all age groups) were used to identify developmental dental anomalies, and information was collected from medical records for statistical analysis. Data were expressed as relative frequency (percentage). The chi-square test, Fisher's exact test, rela

We present a genome assembly from an individual Anodontia alba (buttercup lucine; Mollusca; Bivalvia; Lucinida; Lucinidae). The genome sequence has a total length of 1 862.85 megabases. Most of the assembly (99.28%) is scaffolded into 18 chromosomal pseudomolecules. The mitochondrial genome has also been assembled, with a length of 18.48 kilobases. Gene annotation of this assembly by Ensembl identified 12 083 protein-coding genes. From the metagenome data, we recovered four bins, of which three were high-quality MAGs.

Congenital tooth agenesis impairs masticatory function and aesthetics and adversely affects craniofacial development. Although largely considered genetic in origin, its exact etiology remains unclear. This study reports three familial cases of nonsyndromic congenital tooth agenesis (NSTA). Whole genome sequencing (WGS) revealed five pathogenic variants: filamins-B (FLNB) (c.5186C>A, p.Ser1729Ter), methylcrotonyl coenzyme a carboxylase 2 (MCCC2) (c.91C>T, p.Gln31Ter; c.484C>T, p.Gln162Ter; c.340C>T, p.Gln114Ter), laminin subunit alpha 2 (LAMA2) (c.1084A>T, p.Arg362Ter), cathepsin C (CTSC) (c.74
