Towards credible digital twins for basic and preclinical research.

Atomoxetine (Strattera(R)) is a selective norepinephrine (noradrenaline) reuptake inhibitor that is not classified as a stimulant, and is indicated for use in patients with attention-deficit hyperactivity disorder (ADHD). Atomoxetine is effective and generally well tolerated. It is significantly more effective than placebo and standard current therapy and does not differ significantly from or is noninferior to immediate-release methylphenidate; however, it is significantly less effective than the extended-release methylphenidate formulation OROS(R) methylphenidate (hereafter referred to as osm

Infectious medical waste (IMW) and disease transmission: A critical public health challenge in Africa and the global implications - A systematic review.

BACKGROUND: Infectious Medical Waste (IMW) from both human and animal healthcare sources have the potential to transmit diseases in human populations due to its pathogenic composition. Conversely, a paucity of data regarding cause-and-effect relationships inhibits our understanding of the extent to which disease transmission occurs. In parts of Africa where the management of IMW is not prioritized, the risks to public health, along with its global implications cannot be ignored. METHODOLOGY: A systematic literature search following the PRISMA guidelines was conducted across five electronic da

The impact of bacterial and viral diseases on dromedary camel (Camelus dromedarius) welfare: a comprehensive review.

Dromedary camels (Camelus dromedarius) are vital to food security, livelihoods and cultural identity in arid and semi-arid regions of Africa, the Middle East and South Asia, with global populations exceeding 35-40 million and continuing to rise as climate change favors livestock adapted to harsh environments. Despite their expanding roles in meat and milk production, transport, tourism and racing, camel health and welfare remain comparatively underexplored, particularly regarding infectious diseases. This review synthesizes current knowledge on major bacterial infections, including brucellosis

Parabens and estrogen-mediated cancers: unraveling coincidence, co-occurrence, and causality.

Trabeculectomy remains the 'gold standard' intraocular pressure (IOP)-lowering procedure for moderate-to-severe glaucoma; however, this approach is associated with the need for substantial post-operative management. Micro-invasive glaucoma surgery (MIGS) procedures aim to reduce the need for intra- and post-operative management and provide a less invasive means of lowering IOP. Generally, MIGS procedures are associated with only modest reductions in IOP and are targeted at patients with mild-to-moderate glaucoma, highlighting an unmet need for a less invasive treatment of advanced and refracto

Lysosomal metabolism in pancreatic cancer and its crosstalk with host physiology.

Transcription factors from the family of Signal Transducers and Activators of Transcription (STAT) are activated by numerous cytokines. Two members of this family, STAT5A and STAT5B (collectively called STAT5), have gained prominence in that they are activated by a wide variety of cytokines such as interleukins, erythropoietin, growth hormone, and prolactin. Furthermore, constitutive STAT5 activation is observed in the majority of leukemias and many solid tumors. Inactivation studies in mice as well as human mutations have provided insight into many of STAT5's functions. Disruption of cytokine

The Plague of Athens: Investigating the Enigmatic Epidemic of Ancient Greece.

The ancient Greek historian Thucydides described in his book "History of the Peloponnesian War" one of the earliest epidemics in known human history, the "Plague of Athens". The plague, which lasted from 430-426 B.C., had a death toll estimated at around 75,000 - 100,000. The importance of the plague was immense. Pericles, the historical leader of Athens, was among its victims, and his death was just the beginning of the fall of the Golden Age of Athens. The enormous death toll from the plague weakened Athens, leading to its eventual defeat in the war against Sparta. So far, the cause of this

Climate-sensitive infectious diseases in transition: a scoping review of key diseases and their epidemiology, data applications and mechanism of outbreak response in Saudi Arabia.

BACKGROUND: Climate-sensitive infectious diseases (CSID), of both vector- and water-borne diseases present growing public health concerns in Saudi Arabia. By virtue of the absence of evidence-based vaccine for most of these diseases, adequate management should primeirly rely on timely and reliable data integration, robust disease surveillance, and temporal and spatial vector control strategies. OBJECTIVE: This scoping review aimed to examine the scale and scope of CSID in Saudi Arabia, with attempts to elicit all diseases' trends and patterns, the availability of associated epidemiological, e

Use of Rift Valley Fever Virus Expressing NanoLuc Luciferase for the Assessment of Neutralizing Antibodies and Antivirals.

Rift Valley fever (RVF) is an arboviral zoonotic disease affecting many African countries with the potential to spread to other geographical areas. In this chapter we describe the use of a replication-competent recombinant (r)RVFV expressing NanoLuc Luciferase (Nluc) for in vitro studies. The determination of parameters such as neutralizing antibodies in serum samples, or the antiviral activity of drugs is usually carried out using standard assays based on the assessment of cytopathic effect on cell cultures. The use of a virus encoding a traceable reporter protein allows to correlate the pres

Molecular Diagnosis of Phlebovirus riftense Infection Using an L-Segment-Based Real-Time RT-PCR Method.

Rift Valley fever (RVF) is one of the main vector-borne zoonotic diseases that affects a wide range of ruminants and humans in Africa and the Arabian Peninsula.Several techniques involving cell culture and molecular biology methods have been developed to diagnose RVF infection. Success partly relies on sending samples to a national or reference laboratory in good conditions and having the capacity to perform the appropriate diagnostic test in the matrix of interest during the period of viremia where high loads of viral particles are present. Conventional and duplex/multiplex real-time reverse

Cryogenic Electron Microscopy of Rift Valley Fever Virus.

Rift Valley fever virus (RVFV) is an important livestock and human pathogen. It is also a potential bioweapon owing to its ability to spread by aerosols. It is an enveloped virus containing surface protrusions composed of two viral glycoproteins, Gc and Gn; the viral core contains ribonucleoprotein complexes. We describe the use of cryogenic electron microscopy (Cryo-EM) of single particles to visualize the three-dimensional (3D) organization of RVFV. The spherical RVFV virions have a diameter of 100 nm with icosahedral symmetry (T = 12) and 720 prominent protrusions on their surface. In this c

Vector Competence Assays for RVFV in Mosquitoes.

Rift Valley fever virus (RVFV) is the causative agent of Rift Valley fever, a mosquito-borne zoonotic febrile illness that primarily affects ruminants, camels, and humans. It is endemic in sub-Saharan Africa and the Arabian Peninsula, albeit with the potential to spread to and establish in non-endemic countries where mosquito vectors are present. Assessing the vector competence (VC) of local mosquito populations is essential for risk evaluation and to develop more accurate control and surveillance strategies. Herein, parameters for VC experimental design, as well as protocols for mosquito rear

Recombinant Protein Production and Purification of Rift Valley Fever Virus Nucleoprotein from Escherichia coli Expression Systems.

The recombinant expression and purification of viral proteins are a key component in the study of the immune response of viruses, as well as the creation of diagnostic techniques for the detection of viruses. For structurally simple proteins, one commonly used technique is the production of recombinant proteins in bacterial expression systems, which enable the large-scale synthesis and purification of recombinant viral proteins. In this technique, the cDNA encoding for a viral protein is cloned into a bacterial expression vector (with an appropriate purification tag), produced in a modified ba

Bunyaviruses: Expression of Recombinant Glycoproteins from Insect Cells.

High-density suspension cultures of insect cells offer a scalable and serum-free system for the expression of recombinant proteins. Rift Valley fever virus (RVFV), an arthropod-borne virus spread by mosquitoes, contains two envelop glycoproteins Gn and Gc. These glycoproteins are crucial for eliciting neutralizing antibodies that can offer protection against RVFV infection. Here, we describe the production of individual RVFV glycoproteins and empty RVFV particles using S2 Drosophila insect cells, followed by purification and analysis of the purified proteins.
