Curcumin-based Nanotherapeutics to Combat Neurodegenerative Disease: Drug Delivery Strategies and Therapeutic Potential.

Curcumin (CUR) has been used as a medicine, colouring agent, and flavouring agent for thousands of years. Polyphenolic active constituents of turmeric show antimicrobial, anticancer, antioxidant, anti-inflammatory, astringent, and other valuable properties. Numerous studies on animals have demonstrated that CUR is safe, with no harmful effects on the kidney and liver, even at higher doses. This review article aims to gather information about the properties and the use of active constituents of CUR in nanotechnology-based treatment strategies for neurodegenerative diseases (NDs). From scientifi

Bisphenol S induces hepatic steatosis and insulin resistance through AMPK inhibition and endoplasmic reticulum stress.

Bisphenol S (BPS), a common substitute for bisphenol A, has emerged as a widespread environmental contaminant, yet its metabolic effects remain poorly understood. Increasing evidence links BPS exposure to metabolic dysfunction-associated steatotic liver disease (MASLD), although the underlying mechanisms are unclear. Here, we investigated the metabolic effects of chronic BPS exposure (4, 25, and 50 μg/kg/day for 12 weeks) in C57BL/6 mice and evaluated its impact on human Huh-7 hepatocytes. In vivo, exposure to 25 μg/kg/day exhibited obesogenic potential, while all doses induced insulin resista

Evaluation of Umbelliferone Neuroprotective Effects Against Acrylamide-Induced Oxidative Stress, In Vivo and In Silico Study.

Acrylamide (ACR) is a heat-generated carcinogen, one of the most common toxins, with various effects on the biological system, including oxidative stress and related disorders. Umbelliferone (UMB) is a naturally occurring coumarin derivative that is present in edible fruits, known as an antioxidant, and reported for many other therapeutic effects. In the current study, UMB was evaluated for neuro- and hepatoprotective activity against oxidative stress toxicity induced by a 40 mg/kg daily dose of ACR in a mouse model over 15 days. Also, UMB was used as a ligand for the first time to predict its

The Regulatory Role of Iron Transporter SLC39A13 in Liver Fibrosis.

Liver fibrosis, driven by excessive collagen synthesis following hepatic injury, poses a significant health challenge. SLC39A13/ZIP13, a recently characterized intracellular iron transporter, is shown to provide iron to the ER/Golgi to help catalyze procollagen hydroxylation during collagen maturation. Here, we investigate whether ZIP13 plays a role during hepatic fibrogenesis modeled by CCl4 stress or other inducers. ZIP13 expression is induced during liver fibrosis. Germline disruption of Zip13 dramatically reduces fibrosis. Surprisingly, these mice do not benefit from ZIP13 loss after CCl4

Exploring the Impact of Polychlorinated Biphenyls (PCBs) on the Development of MASLD: A Comprehensive Review.

Metabolic dysfunction-associated steatotic liver disease (MASLD), formerly known as non-alcoholic fatty liver disease (NAFLD), is becoming the most common liver disease, affecting between 30 and 40% of the global population. MASLD is a multifaceted disease spectrum that is closely associated with obesity, insulin resistance, type 2 diabetes mellitus and, more broadly, metabolic syndrome. All these conditions increase the risk of liver-related mortality, which explains the intense research efforts in recent years to better elucidate its pathogenesis. The crucial impact of environmental pollutan

The mechanistic nexus between microplastics and chronic liver disease: A review of current evidence.

The progression of chronic liver disease results in severe complications. However, current treatment options remain limited. Research indicates that environmental pollutants, such as microplastics (MPs), pose a threat to the progression of chronic liver disease and represent a substantial challenge to human health. Current studies widely suggest that MPs tend to accumulate in the liver upon entering the body, potentially leading to the onset or acceleration of chronic liver disease. This article seeks to provide a comprehensive overview of the effects of MPs on chronic liver disease through va

Contaminating plasmid sequences and disrupted vector genomes in the liver following adeno-associated virus gene therapy.

Adeno-associated viruses (AAVs) are common vectors in gene therapy but can frequently cause liver complications in patients. The mechanisms underlying AAV-related liver toxicity remain poorly understood, posing challenges for effective prevention and intervention. Here we conducted a case study of a child with spinal muscular atrophy type 1 experiencing substantial hepatitis after receiving onasemnogene abeparvovec, undertaking long- and short-read metagenomic sequencing of liver tissue. We identified manufacturing plasmid sequences with complex structures and recombination. Vector genomes had

Integrated mesenchymal and extracellular cues drive bioengineered liver tissue formation and function.

Human tissue engineering holds great promise for creating physiological models while facing challenges replicating natural complexity, including cellular and extracellular cues. Current approaches often miss the incorporation of major bioengineering factors (i.e., cellular complexity, well-defined extracellular matrix (ECM) mimicry, dynamic stimuli). We bioengineered liver tissues (BLTs) utilizing human intrahepatic cholangiocyte organoids (ICOs), hepatic stellate cells (HSCs), and mesenchymal stromal cells (MSCs), a synthetic polyisocynide (PIC)- based hydrogel, and dynamic suspension culture

Bile acid retention in efferocytic macrophages shapes their inflammatory status during cholangitis.

The clearance of apoptotic cells by phagocytes is crucial for restoring tissue balance after injury. In autoimmune liver diseases like primary sclerosing cholangitis, cell death is thought to result from accumulation of toxic bile acids within parenchymal cells. Whether, in this context, bile acid-loaded dying cells impact the efficiency of phagocytic macrophages in restoring tissue balance remains unknown. Here, we demonstrate that in a murine model of cholangitis, bile acids accumulate in a subpopulation of efferocytic macrophages with pro-inflammatory features. Our in vitro results indicate

Neurotoxicity of antimony: A review of epidemiological evidence and the underlying molecular mechanisms.

Antimony (Sb) is a toxic metalloid and a global pollutant. Sb exposure is known to cause pulmonary, cardiovascular, liver and kidney damage, as well as cancer. In addition, data showing neurotoxic effects of Sb have been also obtained recently. Therefore, the objective of the present review was to discuss existing epidemiological findings linking Sb exposure to brain diseases and the underlying molecular mechanisms of Sb neurotoxicity. Laboratory findings revealed neurotoxic effects of high-dose Sb exposure. Specifically, in vitro and in vivo studies show that Sb induces neuronal apoptosis thr

Silibinin Modulates NF-κB Activation and Heat Shock Protein in Preeclampsia-Like Rat Model.

PROBLEM: Preeclampsia (PE) is a serious human pregnancy complication and remains the most complex hypertensive disorder due to its multifactorial nature and multisystemic effects. Despite the incomplete understanding of the PE pathophysiology, it is well-accepted that placental changes in PE are associated with oxidative stress, increasing the excessive production of cytokines and other inflammatory mediators. Thus, agents such as silibinin, a non-toxic natural polyphenolic flavonoid with antioxidant, anti-inflammatory, and hepatoprotective properties could modulate this profile for better dis

Efficacy of peptide receptor radionuclide therapy in patients with oligodiscordant gastroenteropancreatic neuroendocrine tumours.

BACKGROUND: The use of peptide receptor radionuclide therapy (PRRT) is well established in the treatment of advanced or unresectable neuroendocrine tumours (NETs) after progression on somatostatin analogues (SSA), with randomised clinical trials such as NETTER-1 and NETTER-2 showing improvement in progression free survival (PFS) as well as tumour response [12, 13]. Current landmark trials only considered patients suitable for PRRT if all known lesions displayed SSTR expression, via avidity on planar scintigraphy or Gallium-68 DOTATATE PET/CT. However, there is a select group of patients with o

Pharmacokinetics, pathology and efficacy of SARS-CoV-2 main protease inhibitor VPC285785 in a murine model of coronavirus infection.

Herein we report the development of novel covalent inhibitors of the SARS-CoV-2 main protease (Mpro). The developed compounds VPC285785 and VPC285786 demonstrated moderate inhibition of Mpro (IC50 0.8 µM vs. Nirmatrelvir 0.03 µM), whereas VPC285786 additionally inhibited human cathepsin L (CatL; IC50 4.2 µM vs Nirmatrelvir > 100 µM). In vitro metabolic stability studies in human and mouse microsomes revealed that VPC285786 demonstrated enhanced metabolic stability compared to Nirmatrelvir, with minimal turnover observed during the experimental window. Subsequent mass spectrometry analysis iden

DOACs in cirrhosis: Progress, pitfalls, and the path forward.

Direct oral anticoagulants (DOACs) were historically avoided in cirrhosis due to concerns regarding impaired hepatic metabolism, cumulative toxicity, and the systematic exclusion of patients with cirrhosis from pivotal randomized trials. However, over the past decade, a growing body of retrospective cohorts, meta-analyses, and small prospective studies - including limited randomized comparisons - have evaluated DOACs against vitamin K antagonists (VKAs) for management of venous thromboembolism (VTE), splanchnic vein thrombosis (SVT), and atrial fibrillation (AF) in cirrhosis. In Child-Pugh A a

Stepwise Interpretation of Albumin Fraction Abnormalities on Serum Protein Capillary Electrophoresis in Nephrotic Syndrome: A Case Series.

Serum protein electrophoresis (SPEP) is commonly used in the diagnostic evaluation of nephrotic syndrome (NS) to rule out monoclonal gammopathies. However, NS itself and various other factors may cause transient bisalbuminemia, resulting in albumin fraction irregularities in capillary SPEP and complicating interpretation. This study proposes a stepwise approach for interpreting albumin fraction irregularities, illustrated through three representative cases of NS. Case 1: a 44-year-old woman with isolated proteinuria, Case 2: a 36-year-old woman with diabetic foot infection, and Case 3: a 58-ye

Dapsone-Induced Systemic Drug Reaction in a Patient With Immunoglobulin A Vasculitis.

Dapsone is widely used for inflammatory dermatoses and vasculitides but may cause severe systemic hypersensitivity reactions. Diagnosis can be challenging when clinical features overlap with underlying inflammatory disease and when eosinophilia is absent. We report a case of a young man with biopsy-confirmed immunoglobulin A vasculitis (Henoch-Schönlein purpura) who developed facial and neck edema, a diffuse morbilliform eruption, and hepatocellular liver injury approximately four weeks after initiation and dose escalation of dapsone. Laboratory evaluation demonstrated elevated aminotransferas
