New cosmetics patch test series: A proposal.

Contact dermatitis to cosmetic products is a common condition. It is likely the most frequent reason for performing patch tests because many substances used in cosmetics are potent allergens. Cosmetic contact dermatitis most frequently affects the face. Patch test series need to be continuously revised to identify outdated and emerging allergens. The aim of this study is to propose a reformulation of the patch test series for cosmetics. The list of European Surveillance System on Contact Allergies was initially used to select the allergens. Publications in PubMed from the last 10 years were so

The skin sensitization prediction model: an algorithm for real-world prediction of skin sensitization risk and minimization of human sensitization testing.

Skin sensitization testing to ensure the safety of skincare products for public consumption has largely relied on human repeat insult patch test (HRIPT). The desire to minimize reliance on HRIPT has prompted a search for alternative methods to assess the sensitization risk of consumer products to inform decision-making about their suitability before being brought to market. The novel Skin Sensitization Prediction Model (SSPM) is a methodology that draws upon a database consisting of more than 20 years of historical HRIPT data pertaining to 1274 unique product formulations, comprising 1226 comm

An extended Reference Chemical Potency List (RCPL) for characterising the performance of New Approach Methodologies (NAMs) in measuring the skin sensitisation potency of fragrance chemicals.

The development of a Reference Chemical Potency List (RCPL), and its purpose, has been described previously. That original RCPL comprised 33 chemicals, of varying skin sensitising activity, for each of which a discrete Potency Value (PV) was derived, based upon the best available human and/or animal (local lymph node assay) data. The purpose of the RCPL was to provide a reliable tool that would facilitate evaluation of the ability of New Approach Methodologies (NAMs) to measure skin sensitising potency. We here report the construction of an extended RCPL with 77 additional chemicals by applyin

Development of an Endpoint-Specific Fingerprint to Identify Structurally Similar Chemicals for Skin Sensitization Assessment by Read-Across.

Human health risk assessment of cosmetic ingredients must address skin sensitization due to their direct skin application. With the recent shift toward Next Generation Risk Assessment (NGRA), which utilizes animal-free alternative testing methods, the read-across (RAx) approach has gained prominence. RAx relies on information from structural analogues to fill data gaps and determine a point of departure (PoD). A major challenge in RAx is assessing "similarity" between the target chemical and its analogues, typically based on chemical structure, physicochemical properties, metabolic reactivity,

[Translated article] Should Hydroxyisohexyl 3-Cyclohexene Carboxaldehyde (Lyral®) Still be Part of the Baseline Series?

BACKGROUND AND OBJECTIVES: Hydroxyisohexyl 3-cyclohexene carboxaldehyde (HICC), or Lyral®, is a fragrance marker that is part of the Fragrance Mix II (FM II) and is still patched as an independent allergen within the European and other baseline series despite the European Commission banning its use in cosmetics in 2021. We aimed to study the prevalence of sensitization to the HICC in Spain and its simultaneous positivity with the FM II to determine whether it should be part of the Spanish baseline series. MATERIAL AND METHOD: We analyzed all consecutive patients simultaneously patch-tested wi

Quantitative next generation risk assessment for skin sensitization - application of regression models based on in vitro data to estimate point of departure.

Quantitative risk assessment (QRA) for dermal sensitization is essential for determining safe concentrations of skin sensitizers in consumer products. The fragrance industry developed the QRA2 approach, which uses the No Expected Sensitization Induction Level (NESIL) as a starting reference dose or point of departure (PoD). Animal alternatives for potency assessment have emerged to calculate quantitative PoDs. One such alternative is in vitro-based regression models. Herein, a framework for incorporating regression models into next-generation risk assessment (NGRA) is presented. The framework
