Pseudo-Psoriatic Nail Dystrophy Associated With Cosmetic Nail Products.

Pseudo-psoriatic nail dystrophy, a form of allergic contact dermatitis (ACD) secondary to nail product use, often mimics nail changes associated with psoriasis, leading to diagnostic delays. This review focuses on identifying dystrophic nail changes, particularly those resembling pseudo-psoriatic alterations, caused by cosmetic nail products and offers diagnostic and management strategies. Among the allergens listed as culprits, triethylene glycol dimethacrylate, hydroxyethyl acrylate, and hydroxyethyl methacrylate were the most common. This poses a challenge in differentiating between ACD and

USB1 deficiency disrupts neutrophil maturation via RNA dysregulation independent of global pre-mRNA splicing.

BACKGROUND: U6 biogenesis 1 (USB1) gene mutations cause poikiloderma with neutropenia (PN), which is clinically characterized by skin hyperpigmentation, nail dysplasia, neutropenia, and an elevated risk of cancer. USB1 functions as an RNA exonuclease involved in RNA maturation and stability regulation, although its precise mechanism of action in the hematopoietic system remains unclear. METHODS: We established a myeloid cell-specific USB1 knockout mouse model (USB1fl/fl-Lyz2-cre) using CRISPR/Cas9. Using a combination of research methods, including Western blot, flow cytometry, messenger RNA

Marked improvement of isolated acrodermatitis continua of hallopeau with upadacitinib: a case report and literature review.

Acrodermatitis continua of Hallopeau (ACH) is a rare and chronic variant of pustular psoriasis characterized by sterile pustules, periungual inflammation, and progressive nail destruction. Due to its rarity and overlapping clinical features with other nail disorders, ACH is frequently underrecognized and remains difficult to treat, with no standardized therapeutic guidelines. Here, we report a case of long-standing, isolated ACH showing marked improvement after treatment with the selective Janus kinase 1 (JAK1) inhibitor upadacitinib. A 54-year-old man presented with a 13-year history of recur

The Changing Trends in the Incidence and Clinical Presentation of Immunobullous Disorders Before and During the COVID-19 Era in a Tertiary Healthcare Centre in Andhra Pradesh.

Background Autoimmune immunobullous disorders (AIBDs) represent a heterogeneous group of chronic blistering dermatoses mediated by autoantibodies against desmosomal and hemidesmosomal adhesion molecules. The COVID-19 pandemic has generated significant interest in the interaction between SARS-CoV-2 infection, vaccination-related immune modulation, and the clinical behaviour of autoimmune diseases. Objective This study's primary objective is to compare hospital-based temporal trends in the frequency, subtype distribution, and clinical presentation of AIBDs before and during the COVID-19 era in a

Efficacy of Dupilumab in Treating Trachyonychia and Other Nail Dystrophies: A Systematic Review of Case Reports.

Trachyonychia and related nail dystrophies are often associated with inflammatory skin diseases such as atopic dermatitis, psoriasis, and alopecia areata. Management remains difficult, as standard therapies frequently yield limited benefit. Dupilumab, an IL-4Rα antagonist, is effective for Th2-driven conditions, but its role in nail dystrophies has not been systematically assessed. This review evaluates the existing evidence on dupilumab for trachyonychia and related nail disorders. A systematic search of PubMed, Wiley, Web of Science, MEDLINE, Google Scholar, and EBSCO was performed from ince

Responsibility sharing among children with Type 1 Diabetes and their caregivers: a cross-sectional survey from North India using the Diabetes Family Responsibility Questionnaire (DFRQ).

INTRODUCTION: Reconstruction has evolved long way from primary closure to flaps. As time evolved, better understanding of vascularity of flap has led to the development of innovative reconstructive techniques. These flaps can be raised from various parts of the body for reconstruction and have shown least donor site morbidity. We use one such peroneal artery perforator flap for tongue reconstruction with advantage of thin pliable flap, minimal donor site morbidity and hidden scar. MATERIALS AND METHODS: Our patient 57yrs old lady underwent wide local excision with selective neck dissection. P

Successful management of pediatric Pythium insidiosum keratitis - A case report highlighting the role of targeted antimicrobial therapy and mechanical debridement.

Human pythiosis keratitis is a rare but potentially sight-threatening infection. We report the case of an 8-year-old boy who developed a nonhealing corneal ulcer after finger nail trauma that did not respond to broad-spectrum antibiotics. Initial cultures identified Staphylococcus warneri and Streptococcus mitis. Worsening clinical signs prompted repeat investigations, which confirmed Pythium insidiosum through polymerase chain reaction. Despite intensive topical (natamycin and voriconazole) and systemic antifungals (voriconazole), the ulcer progressed with biofilm formation. Manual debridemen

Expanding the spectrum of Darier Disease variants: 13 novel ATP2A2 genetic variations discovered in an Italian retrospective analysis.

Darier Disease (DD) is a rare autosomal dominant genodermatosis (prevalence: 1/30.000-100.000) characterized by hyperkeratotic papules in seborrheic areas, palmoplantar pits, distinctive nail dystrophy and in some cases neurological symptoms. ATP2A2, encoding for the sarco/endoplasmic reticulum calcium transporting ATPase2 (SERCA2), is the causative gene of DD. It is characterized by high penetrance but variable expressivity and the onset is usually in childhood/puberty. A cohort of 50 suspected DD subjects have been analysed by the Medical Genetics Unit of 'Mauro Baschirotto' Institute for Ra

Genome-wide association study of atopic and autoimmune comorbidities in alopecia areata.

INTRODUCTION: Alopecia areata (AA) is a chronic autoimmune-mediated disorder characterized by hair loss from the scalp and/or body. AA patients frequently present with comorbid chronic inflammatory disorders (CIDs), particularly atopic and autoimmune diseases. Genome-wide association (GWA) studies have suggested a genetic link. However, no studies to date have examined genetic factors that are associated with the comorbid development of CIDs directly in individuals affected by AA. METHODS: We performed an exploratory GWA study in Central European AA patients stratified by self-reported comorb

Cutaneous lichen planus beyond the skin: tricho-onychoscopic and systemic insights.

Cutaneous lichen planus (CLP) often involves the scalp and nails subclinically. Early appendageal signs are underrecognized despite their potential for timely diagnosis and prevention of irreversible damage. To determine the frequency of mucosal and appendageal involvement in CLP, assess potential contributing factors (e.g., medications, systemic diseases, vaccines), and identify early trichoscopic and onychoscopic features. A retrospective study was conducted on 108 patients with histopathologically confirmed CLP. Data included clinical, trichoscopic, onychoscopic, and systemic parameters. Dr

Clinical evaluation of a rapid antigen detection test for the diagnosis of tinea unguium.

INTRODUCTION: Currently, diagnosis of dermatophyte onychomycosis relies on direct microscopic examination (DME) and fungal cultures of nail material. Herein, we aim to evaluate the clinical performance of the rapid antigen detection (RAD) assay PreventID Dermatophyte in comparison to conventional methods for the diagnosis of tinea unguium. METHODS: Between February and July 2024, 48 nail samples, collected at the Navarra University Hospital, Spain, were included in the evaluation. In addition to DME and culture, the RAD test was executed. In case of discrepancies, the qPCR assay Novaplex™ Der

Cutaneous Toxicities of T-Cell-Redirecting Therapies in Multiple Myeloma: An EADV Task Force Position Paper and Practical Recommendations.

T-cell-redirecting immunotherapies, including bispecific antibodies and chimeric antigen receptor (CAR) T-cell therapies, have rapidly become major treatment options of relapsed or refractory multiple myeloma but are associated with a distinctive spectrum of cutaneous toxicities. Dermatologic manifestations are particularly prominent with GPRC5D-targeting agents, reflecting on-target, off-tumor activity against hard-keratinizing tissues, whereas BCMA-targeted bispecific antibodies and anti-BCMA CAR T-cell therapies are associated with milder and less phenotypically distinct cutaneous adverse e
