Association of lifestyles and mental health with adult acne: A population-based cross-sectional study.

BACKGROUND: Acne is a multifactorial inflammatory disorder which is influenced by lifestyle and psychosocial factors. Comprehensive evidence is lacking on modifiable behavioural risks and the mediating role of psychological distress. OBJECTIVE: This population-based cross-sectional study investigated the associations between sedentary behaviour, physical exercise, sleep and acne in adults living in Southern China, and quantified the mediating effects of depressive and anxiety symptoms. METHODS: Participants were recruited from 10 districts in Shenzhen who completed investigator-administered

Spatial transcriptomics identifies differentiation, lipid metabolism, and retinoid pathway alterations in acne vulgaris.

Acne vulgaris is a common skin condition involving complex interactions among lipid-secreting sebaceous glands, keratinocytes, immune cells, and microbiota. While retinoids are effective for treating acne, disease pathogenesis remains poorly understood. In particular, it remains unclear how different subtypes of acne, including inflammatory (pustular) and noninflammatory (comedonal) lesions, vary in gene expression, signaling, and sebaceous gland involvement. Here, we performed spatial transcriptomics on healthy, nonlesional, comedonal, and pustular acne skin using a custom panel targeting seb

Combination Effects of Clindamycin and Benzoyl Peroxide Against Cutibacterium acnes.

This study aimed to evaluate the clinical efficacy of combination therapy with clindamycin and benzoyl peroxide in treating acne vulgaris. We assessed the antimicrobial susceptibility of Cutibacterium acnes isolates obtained from these patients. In addition, the potential risk of C. acnes developing resistance to clindamycin and benzoyl peroxide following exposure was investigated in vitro. We analyzed 182 C. acnes isolates from patients with acne to evaluate the clindamycin susceptibility and resistance determinants and to examine the association between topical clindamycin use and resistance

Nitric oxide-releasing hyaluronic acid as an antibacterial and immunomodulatory acne therapeutic.

Acne vulgaris (acne) is a common skin disorder associated with significant psychosocial impact. Current clinical therapies include topical and systemic antibiotics, benzoyl peroxide, and retinoids. While moderately effective, these clinical therapies fail to target all four major pathogenic causes of acne and are associated with painful side effects. Nitric oxide (NO), an endogenous signaling molecule, represents a promising alternative to conventional acne treatments due to its innate antibacterial and immunomodulatory functions. As NO is highly reactive, macromolecular NO donors are required

Azure A as a promising candidate for antimicrobial photodynamic therapy of acne-related bacteria.

Antimicrobial photodynamic inactivation (aPDT) is an effective technique for treating acne vulgaris, due to its efficiency in inactivating multiresistant bacteria and the absence of systemic side effects. This study investigated the potential of the dye Azure A (AZA), belonging to the phenothiazine class, as a photosensitizer (PS) in aPDT to combat the gram-positive bacteria Staphylococcus epidermidis and Cutibacterium acnes, both associated with inflammatory acne vulgaris. The efficiency of AZA was compared to Orthotoluidine Blue (OTB), evaluating the absorbance spectra, singlet oxygen genera

Microbiome-Derived Indole-3-Lactic Acid Attenuates Cutibacterium Acnes-Induced Inflammation via the Aryl Hydrocarbon Receptor Pathway.

Acne vulgaris is a chronic inflammatory dermatosis where conventional therapies often face limitations in efficacy and safety, necessitating the development of microbiome-targeted interventions. This study investigated the immunomodulatory potential of microbiome-derived tryptophan metabolites as a novel therapeutic strategy for Cutibacterium acnes (C. acnes)-induced inflammation, focusing on the aryl hydrocarbon receptor (AHR) pathway. We evaluated indole-3-lactic acid (ILA), indole-3-acrylic acid (IAA), and indole-3-propionic acid (IPA) in comparison to tapinarof, utilizing C. acnes-stimulat

Dermocosmetics in Acne Vulgaris: South African Consensus Recommendations With a Focus on Skin of Color.

BACKGROUND: Acne vulgaris is one of the most common dermatological disorders worldwide, affecting both adolescents and adults. It frequently leads to significant psychosocial and physical sequelae, including acne-induced hyperpigmentation and scarring. Beyond pharmaceutical therapies, dermocosmetics-topical formulations enriched with active ingredients in cosmetically elegant vehicles-have emerged as essential partners in acne management. They optimize clinical outcomes by supporting skin barrier repair, reducing irritation associated with conventional treatments, and targeting key pathogenic

Placebo response and its influencing factors in acne vulgaris: A model-based meta-analysis.

AIMS: The high placebo response in acne clinical trials has slowed down the progress of new drug development. Therefore, this study used a model-based meta-analysis method to quantitatively analyse the placebo response and explore its influencing factors. METHODS: This study included 21 627 subjects from acne-related randomized double-blind placebo-controlled trials. Pharmacodynamic models were developed for four endpoints: total lesion counts absolute change, noninflammatory lesion counts absolute change, inflammatory lesion counts absolute change and treatment success rate defined by the in

A Case of Pseudogout in an Adolescent on Isotretinoin.

Pseudogout or calcium pyrophosphate dihydrate deposition disease rarely occurs in the young. Known risk factors for pseudogout include age, previous surgery, trauma, metabolic conditions, and medications. Isotretinoin, a retinoid frequently used to control acne vulgaris, is known to cause arthralgia, arthritis, and myalgia. We describe a case of an adolescent using isotretinoin who presented with acute left upper extremity pain and weakness. Birefringent calcium pyrophosphate dihydrate crystals were seen on synovial fluid analysis. The patient's symptoms resolved after discontinuing isotretino
