Prevalence of alpha-synuclein seeding activity in the Olfactory Mucosa following SARS-CoV-2 infection: results from a pilot study.

Olfactory dysfunction (OD) is a hallmark of SARS-CoV-2-infection where it is believed to result from neuroinflammation induced by viral invasion via the olfactory mucosa (OM). Hyposmia also affects the majority of patients with Parkinson's disease (PD) where it is caused by α-synuclein (aSyn) pathology in the olfactory system, and aSyn-seeding-activity can be detected in the OM using aSyn-seeding-amplification-assays (aSyn-SAA). The present pilot study aimed to investigate whether SARS-CoV-2-infection induces local aSyn-misfolding and -aggregation in OM samples using aSyn-SAA. We conducted an

Beyond RSV: why the bronchiolitis burden in infants will not simply disappear.

Nirsevimab and maternal respiratory syncytial virus (RSV) vaccination are delivering real-world important reductions in RSV-bronchiolitis hospitalisations, a landmark achievement in paediatric preventive medicine. We argue, however, that these successes should not be mistaken for the resolution of severe bronchiolitis as a clinical and public health problem. First, RSV's epidemiological dominance may partly reflect its transmission efficiency rather than a unique pathogenic superiority: rhinovirus, human metapneumovirus, adenovirus, and bocavirus already cause substantial disease, and genetic

Serological response to field rabies vaccination in owned dogs in the immune belt of Peninsular Malaysia.

Rabies vaccination is conducted along the northern border of Peninsular Malaysia, but paired field serological data are limited. We analysed routine programme data from 154 owned dogs sampled immediately before vaccination and 30-83 days later (median, 43 days). Rabies virus-neutralising antibody levels were measured using the fluorescent antibody virus neutralisation test. Paired levels in IU/mL were compared using the Wilcoxon signed-rank test, and benchmark proportions were compared using the exact McNemar test. The proportion with levels ≥ 0.5 IU/mL increased from 72.7% (112/154; 95% confi

Toll-like receptor expression across histological states of epidermodysplasia verruciformis.

Epidermodysplasia verruciformis (EV) is characterized by susceptibility to cutaneous β-human papillomaviruses, persistent viral lesions, and an increased risk of cutaneous squamous cell carcinoma (cSCC). We investigated whether expression of Toll-like receptors 3, 4, 5, and 9 (TLR3, TLR4, TLR5, and TLR9) differs across histological states within EV. Immunoreactivity was quantified by digital H-score using QuPath in 97 specimens from 28 patients, comprising 13 lesion-free skin samples, 15 flat warts, and 69 cSCCs. Prespecified comparisons evaluated flat wart versus lesion-free skin, cSCC versus

The role of 2'-5'-oligoadenylate synthetase (OAS) proteins in HIV infection: innate antiviral immunity, viral persistence, and therapeutic opportunities.

Human immunodeficiency virus type 1 (HIV-1) establishes persistent infection despite robust innate immune responses and effective antiretroviral therapy, highlighting the importance of host antiviral mechanisms in regulating viral replication and persistence. Among interferon-stimulated genes, 2'-5'-oligoadenylate synthetase 1 (OAS1) is a key antiviral effector that senses viral double-stranded RNA and activates the OAS-RNase L pathway. Beyond its canonical antiviral activity, emerging evidence implicates OAS1 in immune regulation, viral latency, neuroinflammation, and retroviral disease progr

Development of a risk score to predict encephalitis by Herpes Simplex Virus and Varicella Zoster Virus.

Encephalitis guidelines recommend repeating a lumbar puncture (LP) 3-7 days after an initial negative cerebrospinal fluid (CSF) herpes simplex virus (HSV) polymerase chain reaction (PCR) test in patients with high suspicion for HSV or varicella zoster virus (VZV), but this subgroup has not been specified. Retrospective observational study of 602 adults with encephalitis from 2002 to 2023 from two large U.S. healthcare systems. Multivariate logistic regression models were developed and internally validated to identify clinically relevant and statistically significant features predicting HSV, VZ

Virus transmission required for maintenance and host adaptation mirrors neurophysiological differences in rabies-derived peptides.

Pathogen adaptations to new hosts are central to the disease ecology of cross-species transmission and host switching of viruses. The rabies virus is a model for adaptations to different hosts as it induces behavioral changes that facilitate transmission, while being maintained in distinct species-specific cycles of infection. Here, we demonstrate that the rabies virus requires distinct transmission dynamics in two hosts with different ecologies to establish maintenance, using agent-based modeling, analyses of virus glycoprotein-mediated manipulation of neuroreceptor function, and phylogenetic

Oral uracil treatment increases susceptibility to rectal SIV infection.

The intestinal microbiomes of men who have sex with men (MSM) are distinct as compared to non-MSM and may increase susceptibility to rectal HIV infection. We recently demonstrated that dysbiosis-associated disruptions to host antimicrobial immunity increase macaque susceptibility to rectal SIVmac239X acquisition. The described susceptibility-associated network identified Dual Oxidase 2 (DUOX2) - an epithelial sensor that catalyzes reactive oxygen species (ROS) production upon sensing pathobiont-derived uracil - as a likely contributor to increased SIV susceptibility. We hypothesized that intes

Incidence and predictors of mother-to-child transmission of HIV among HIV-exposed infants in public General hospitals of Tigray, Ethiopia, 2020: Retrospective cohort study.

BACKGROUND: Mother-to-child transmission (MTCT) of human immunodeficiency virus (HIV) remains a major public health challenge, particularly in low-income countries. Previous studies conducted in Ethiopia were largely limited to single health facilities and did not adequately assess important predictors, including maternal viral load, paternal enrollment in antiretroviral therapy (ART), and breastfeeding duration. Therefore, this study aimed to determine the incidence and predictors of MTCT of HIV among HIV-exposed infants in public general hospitals of Tigray, Ethiopia. METHODS: Hospital-base

Knowledge, attitudes and practices about dengue vaccines in Latin America and the Caribbean: a scoping review.

Dengue remains a major public health concern in Latin America and the Caribbean, where vaccination has emerged as a key strategy to reduce severe disease. However, its successful implementation depends on population knowledge, attitudes, and practices (KAP). This study aimed to synthesize available evidence on KAP related to dengue vaccination in the region. A scoping review was conducted following JBI guidelines. Searches were performed in international and regional databases, including MEDLINE/PubMed, Scopus, Embase, Web of Science, SciELO, and the Virtual Health Library. Original studies we

Implementing a First Nation data-sovereign and agile modelling platform for pandemic management: lessons from Manitoba.

This paper describes lessons learned from a partnership between the First Nations Health and Social Secretariat of Manitoba (FNHSSM) and researchers at the University of Manitoba to develop a community-led COVID-19 modelling platform. The envisioned tool would allow First Nations leaders to input local data and generate outputs to support decision-making using a stochastic, age-stratified agent-based model of COVID-19 transmission. Although the platform was not fully operationalized during the project, several important lessons emerged. First, the COVID-19 pandemic placed significant demands o
