Epidemiological investigation of an acute gastroenteritis outbreak associated with norovirus GΙΙ.17[P17] in a cross-border travel group - Shanghai Port, China, 2024.

BACKGROUND: Norovirus is a leading cause of acute gastroenteritis and spreads efficiently in closed, mobile cohorts such as organized travel groups. This case is notable for the real-time detection and genomic confirmation of a cross-border outbreak at a port of entry, including near-identical Norovirus GΙΙ.17[P17] genomes and documented asymptomatic carriage, illustrating the practical value of integrated metagenomic surveillance in border health operations. CASE PRESENTATION: On July 21, 2024, 26 travelers arrived at Shanghai Port after a 12-day group tour in Europe. Clinical interviews ide

Progress in human intestinal organoid research: applications to acute gastroenteritis viruses.

Acute gastroenteritis viruses, such as rotavirus, human norovirus, human astrovirus, human adenovirus, human sapovirus, represent significant threats to global public health. Research on these pathogens has long been hampered by the limitations of conventional models. Animal and cell-based systems, widely used in virological studies, show limited efficiency in supporting rotavirus replication, while noroviruses remain largely non-cultivable in these settings. Organoids-complex, three-dimensional multicellular structures derived from stem cells-exhibit organ-specific characteristics and spatial

Clinical, Molecular and Evolutionary Characterization of Noroviruses in Hospitalized Children With Acute Gastroenteritis in Vietnam, 2016 to 2021.

Following rotavirus vaccine implementation in Vietnam, norovirus has gradually become equally an important as a cause of acute gastroenteritis (AGE) in children as rotavirus. This study aimed to investigate the genotype distribution and compare clinical manifestations of norovirus of children < 5 years of age hospitalized with AGE from 2016 to 2021. Of 6,718 specimens collected through active surveillance from 12/2016 to 05/2021, 2,317 (34.5%) were randomly selected for norovirus testing. Among the 477 (20.6%) norovirus-positive specimens identified from the previous surveillance, 390 (81.8%)

Epidemiologic Trends, Genetic Diversity, and Risk Factors of Norovirus Outbreaks in Beijing, China, 2016-2022.

Norovirus is the leading cause of acute gastroenteritis outbreaks worldwide. A total of 1859 norovirus outbreaks were reported from 2016 to 2022 in Beijing, China. GII.2[P16] was the predominant genotype during 2016-2021, and GII.3[P12] during 2021-2022. In the early stage of the COVID-19 epidemic (January to June of 2020), strict prevention and control measures were implemented, and only eight norovirus outbreaks were reported. Most norovirus outbreaks occurred in schools (95.6%, 1778). As the level of schooling rises, the scale of norovirus outbreaks also increases (median case numbers: 8 fo

Attitudes towards a potential future norovirus vaccine in a representative survey of U.S. adults, February 2025.

Norovirus is an important cause of acute gastroenteritis; vaccines to protect against norovirus illness are in development. In February 2025, we surveyed a representative sample of 4204 U.S. adults to assess attitudes towards norovirus vaccination. Overall, 38% said they would likely get a norovirus vaccine if available, 27% were unsure, and 35% were unlikely to get one. Among adults aged 65+, 54% were likely to vaccinate. Norovirus vaccine acceptance for was similar to COVID and RSV vaccines, and slightly lower than a flu vaccine. Among unsure and unlikely groups, side effects and distrust in

GPR182 and the reframing of lacteal chylomicron uptake.

OBJECTIVE: The introduction of rotavirus vaccines in the United States led to substantial reductions in rotavirus disease burden, whereas the relative burden of other viral pathogens causing acute gastroenteritis (AGE) increased. Our aim was to establish the community-level burden of enteric viral pathogens in a US cohort during the first 2 years of life. METHODS: From birth until age 2 years, weekly stools and AGE symptom surveys were collected from mother-child pairs in the PREVAIL (Pediatric Respiratory & Enteric Virus Acquisition and Immunogenesis Longitudinal) cohort. Stools were tested

Evaluation of Serum Antibodies as Correlates of Protection Against Norovirus Infection and Disease.

BACKGROUND: Norovirus is a major cause of acute gastroenteritis. Several vaccines are in development. Identifying a reliable immunological correlate of protection (CoP) would aid vaccine development and evaluation; however, no strong CoP has been established to date. METHODS: We evaluated the relationships between prechallenge genogroup I, genotype 1 (GI.1) and GII.4 norovirus antibodies and clinical outcomes in two human vaccine-challenge studies. We analyzed genotype-specific histo-blood group antigen blocking titers, serum IgA and IgG, and total antibody ELISA. We used Bayesian regression

Wastewater-based surveillance of norovirus GI and GII: Comparative performance of commercial and in-house RT-qPCR Assays.

Noroviruses, the leading cause of nonbacterial acute gastroenteritis outbreaks, are widely detected in wastewater worldwide and reflect community-level viral circulation. These RNA viruses exhibit high mutation and recombination rates, favoring the emergence of novel variants with epidemiological relevance. In this study, wastewater-based epidemiology was applied to monitor the circulation of norovirus genogroups GI and GII in raw sewage samples collected biweekly throughout 2024 from a wastewater treatment plant serving the Metropolitan Region of Rio de Janeiro, Brazil. In parallel, the analy

Recombinant AcHERV-baculovirus vaccine elicits robust immune responses against human norovirus.

Human norovirus (HuNoV) is a leading global cause of acute viral gastroenteritis, placing a particularly high burden on pediatric and elderly populations. Despite significant progress in HuNoV vaccine development, including advanced clinical trials of virus-like particle (VLP)-based candidates, no vaccine has received regulatory approval to date. In this study, we developed a recombinant baculovirus-based vaccine, AcHERV-HuNoV, designed to express the human norovirus (HuNoV) VP1 protein. This vaccine utilizes the AcHERV platform, which consists of Autographa californica multiple nucleopolyhedr

Bile acids accumulate norovirus-like particles and enhance binding to and entry into human enteric epithelial cells.

Human norovirus (HuNoV) is a leading cause of acute viral gastroenteritis, but despite high impact on public health and healthcare, the mechanisms of viral attachment to and entry into target cells are not yet fully understood. Recent reports indicate that saliva and bile contribute to the transmission of HuNoV. For example, human bile acids increase cell surface ceramide levels in human enteroids, which improves norovirus entry into cells, resulting in enhanced replication. Bile acids can also interact directly with the norovirus capsid, but it is not known whether bile or other gastrointesti

Seasonal dynamics and genetic diversity of non-polio enteroviruses in hospitalized children with acute gastroenteritis in Pune, India.

Acute gastroenteritis (AGE) in children is primarily associated with pathogens such as Human Rotavirus A (HRVA), Human norovirus (HNoV), Human adenovirus (HAdV), Human astrovirus (HAstV), Human parechovirus (HPeV) and increasingly, Human enteroviruses (EVs). EVs are non-enveloped RNA viruses belonging to the Picornaviridae family, comprising over 100 subtypes classified into four species. Despite their clinical relevance, the prevalence of EVs in AGE remains underexplored in India, resulting in limited data on their epidemiology and molecular diversity. A total of 450 stool samples were collec

CBC-derived inflammatory indices predict adverse outcomes in geriatric emergency patients with acute infectious diarrhea: a MIMIC-IV analysis.

Acute infectious diarrhea remains a leading cause of morbidity and mortality worldwide. Similar to pediatric populations, geriatric patients represent a highly vulnerable group with increased hospitalization rates, prolonged length of stay, heightened dehydration susceptibility, diminished physiological reserve, and elevated mortality risk. While pediatric populations benefit from well-established clinical severity scores such as Vesikari and Clark scales, analogous prognostic tools specifically validated for geriatric patients with acute infectious diarrhea remain lacking. Specific inflammato

Norovirus-specific monoclonal antibodies that block histo-blood group antigen binding isolated from healthy donors.

Human noroviruses are a leading cause of acute gastroenteritis, yet no licensed vaccine or antiviral is currently available. The extensive genetic diversity of noroviruses and the limited cross-protective immunity between genotypes complicate the development of broadly effective interventions. To systematically characterize norovirus-specific B cell responses from healthy individuals, we applied linking B cell receptor to antigen specificity through sequencing (LIBRA-seq) to peripheral blood mononuclear cells using a genetically diverse panel of norovirus capsid protruding (P) domain antigens.
