BACKGROUND: Childhood pneumonia remains a leading cause of morbidity and mortality worldwide. Prenatal Zika virus (ZIKV) infection has been associated with severe outcomes such as Congenital Zika Syndrome (CZS), linked to excess hospitalisations. However, the long-term respiratory consequences of prenatal ZIKV exposure across the full clinical spectrum, including children without CZS, remain poorly understood. We aimed to examine the association between prenatal ZIKV exposure and community-acquired pneumonia-related hospitalisation or death during childhood.
METHODS: We conducted a population

Zika virus (ZIKV) is an Orthoflavivirus that poses a significant public health threat due to its unique capacity to cause congenital malformations, neurological disorders, and reproductive complications. Upon infection, host cells sense viral RNA and danger signals through pattern recognition receptors, including Toll-like receptors, RIG-I-like receptors, inflammasomes and the cGAS-STING pathway, leading to the production of type I interferons and the subsequent expression of interferon-stimulated genes. Additional intrinsic restriction mechanisms, such as apoptosis, autophagy, RNA interferenc

Zika virus is a mosquito-borne flavivirus that caused a major epidemic in the Americas in 2015 and 2016 and is associated with congenital birth defects and neurological disease. Like other flaviviruses, it depends on conserved sequence and RNA structural elements that carry out essential steps in its life cycle and in evading host immune defenses. Public sequence databases and annotation tools give free access to Zika sequence data, but comparable structural information is sparse. We built a computational model of Zika sequence and secondary structure for use with the VADR software package, wh

Congenital arboviruses represent a significant public health concern because of their ability to cross the placental barrier and adversely affect fetal development. In this review, we delineate the epidemiology, pathogenesis, clinical manifestations, and diagnostic challenges associated with key congenital arboviral infections, including Zika, dengue, chikungunya, yellow fever, Oropouche, and West Nile virus. While vertical transmission mechanisms vary among arboviruses, inflammation, placental disruption, and viral replication within fetal tissues are common pathological features. Early diagn

The recent outbreak of Oropouche virus (OROV) in Latin-America with unprecedented reports of vertical transmission resulting in congenital malformations and fetal death, redefines the threat posed by this arbovirus. We discover that injection of a low inoculum of the epidemic OROVIRCCS-SCDC_1/2024 strain into the placenta of Swiss mice results in viral replication across embryonic organs, in particular the brain. Infected embryos present with multi-organ pathology, pneumonitis, myocarditis, steatohepatitis and severe neuropathology, including microcephaly, ventriculomegaly and neural loss. Neu

The Zika virus (ZIKV), a mosquito-borne flavivirus, has been linked to congenital Zika syndrome (CZS), a condition characterized by severe neurological abnormalities in infants born to infected mothers, with microcephaly being the most prominent manifestation. Despite extensive research, the molecular mechanisms driving CZS-associated microcephaly remain poorly understood. In this study, we utilized genetic network analysis to unravel the complex molecular interactions underlying CZS pathogenesis. By analyzing transcriptomic data from CZS-affected and unaffected individuals, we identified dist

BACKGROUND: Neutralizing antibodies (nAbs) play a central role in protection against Zika virus (ZIKV) infection. Higher maternal ZIKV nAb titers during pregnancy have been associated with reduced risk of congenital anomalies. However, limited data exist on the long-term persistence of these antibodies in women infected during pregnancy and in children exposed in utero.
METHODOLOGY/PRINCIPAL FINDINGS: We conducted a cross-sectional serological study using stored serum samples from a cohort of mother-child pairs with confirmed maternal ZIKV infection during pregnancy in Campo Grande, Brazil. Z

Zika virus (ZIKV) has emerged as a significant public health concern due to its association with severe neurological outcomes in infants, including microcephaly and congenital Zika syndrome (CZS). However, while the majority of ZIKV infections during pregnancy do not result in CZS, the potential long-term neurological effects of mild or inapparent maternal infections remain poorly understood. In this study, we adapted a model of maternal ZIKV infection in human STAT2 knock-in (hSTAT2) mice to investigate the effects of ZIKV infection during mid-gestation, aiming to mirror typical asymptomatic

BACKGROUND: Zika virus (ZIKV) can cause severe neurological disorders such as congenital microcephaly, and there are currently no approved prevention or treatment measures. Elucidating the antiviral mechanism of the host interferon-stimulated genes (ISGs) is crucial for the development of therapeutic strategies.
METHODS: This study integrated multisystem transcriptomic data [human brain organoids, monocyte-derived dendritic cells (moDCs), and peripheral blood mononuclear cells (PBMCs) from infected patients], combined with cellular experiments and bioinformatics analysis, to systematically in

Congenital and early-life Zika virus (ZIKV) infection can result in neurologic deficits. Precise mechanisms of injury, especially in the more subtle presentation of postnatal infection, are not fully elucidated. Here, we defined the effects of ZIKV on the developing brain using single cell transcriptomics, histopathology and design-based stereology, diffusion MRI, and neurobehavioral assessments in infant rhesus macaques. ZIKV upregulated interferon-stimulated genes in activated microglia and cell death pathways in neurons and downregulated metabolism and differentiation genes in mature oligod

Zika virus (ZIKV) can traverse the placental barrier, leading to fetal microcephaly and congenital zika syndrome (CZS). The viral E protein mediates host-cell interactions and infection. Here, we demonstrated that cord blood natural killer cell-derived extracellular vesicles (CBNK-EVs) potently inhibit ZIKV infection in vitro without compromising cellular viability. Mechanistically, CBNK-EVs engage ZIKV through ITGB2, a surface-enriched integrin that interacts with the viral E protein, facilitating nanoparticle-virion contact or membrane fusion. This interaction triggers antiviral activity via

BACKGROUND: Zika virus (ZIKV) is primarily known for its impact on the fetal central nervous system potentially leading to Congenital Zika Syndrome (CZS). Emerging evidence suggests ZIKV may also affect cardiac development. We conducted a follow-up study evaluating cardiologic findings in infants from ZIKV-exposed mothers.
METHODS: Infants born to mothers with PCR-confirmed ZIKV infection during pregnancy and/or who had positive ZIKV PCR results at birth received echocardiograms in the first year of life. Repeat imaging within 12 months was requested for infants with identified abnormalities.

Background: Zika virus (ZIKV), a mosquito-borne flavivirus, is associated with congenital malformations and neuroinflammatory disorders, highlighting the need to identify host factors that shape infection outcomes. Macrophages, key targets and reservoirs of ZIKV, orchestrate both antiviral and inflammatory responses. Methods: Vitamin D (VitD) has emerged as a potent immunomodulator that enhances macrophage antimicrobial activity and regulates inflammation. To investigate how VitD shapes macrophage responses to ZIKV, we reanalyzed publicly available RNA-seq and miRNA-seq datasets from monocyte-

BACKGROUND: Zika virus (ZIKV) is a flavivirus that has gained global attention due to its association with congenital microcephaly and neuroinflammatory responses. Markers of endothelial activation and purinergic signaling have been identified in the context of ZIKV neuropathogenesis, although the underlying mechanisms remain poorly understood.
METHODS: Brain tissue samples from fatal cases of ZIKV-induced microcephaly were analyzed using immunohistochemistry to detect endothelial activation markers (E-selectin, P-selectin, ICAM-1, and VCAM-1) and purinergic receptors (P2X4, P2X7, and P2Y2).

IMPORTANCE: Epilepsy is a major clinical concern in children with congenital Zika syndrome (CZS). However, whether in utero exposure to Zika virus (ZIKV) without development of CZS is associated with increased epilepsy risk compared with unexposed children remains unclear.
OBJECTIVE: To compare the risk of epilepsy-related hospitalizations during the first 4 years of life among children exposed to ZIKV during pregnancy (with and without CZS) vs an unexposed group.
DESIGN, SETTING, AND PARTICIPANTS: This was a population-based cohort study conducted in Brazil among live-born singleton childre

Although congenital Zika virus (ZIKV) syndrome is well-characterized, the neurodevelopmental consequences of postnatal infection are less understood. Here we used a rhesus macaque model to investigate the developmental consequences of ZIKV infection during infancy on the brain and behavior, building on our prior research. Male and female infant rhesus macaques infected with ZIKV at 1 month of age were compared to sex-, age-, and rearing-matched uninfected controls and infants treated with the TLR3 agonist PolyIC as a control for activation of the innate immune system. Longitudinal behavioral a
