BACKGROUND AND PURPOSE: Brain arteriovenous malformations (bAVMs) carry a variable risk of hemorrhage that persists until complete obliteration is achieved. Gamma-knife radiosurgery (GKRS) is an established treatment, yet delayed treatment effects necessitate reliable follow-up imaging. Digital subtraction angiography (DSA) remains the gold standard, but non-invasive alternatives are desirable. Arterial spin labeling (ASL) MRI, which quantifies cerebral blood flow, has shown promise in detecting residual bAVM perfusion. This study aims to assess the diagnostic performance and prognostic value

Background/Objectives: Autism spectrum disorder (ASD) is a neurodevelopmental disease with both clinical and genetic heterogeneity. Several loss-of-function variants in the chromodomain helicase DNA-binding protein 8 (CHD8) gene have been identified in individuals with ASD and/or developmental delay/intellectual disability. These are associated with specific clinical manifestations, including overgrowth, macrocephaly, sleep disturbance, and gastrointestinal problems. Methods: We performed clinical exome sequencing in a female patient with ASD and macrocephaly. RNA analysis from peripheral bloo

Diagnosis and management of fetal neurologic conditions is evolving rapidly. This scoping review aims to synthesize existing literature characterizing parental preferences for fetal neurologic counseling. A literature search and screening of MEDLINE, Embase, and Web of Science databases resulted in 21 eligible articles. The most common fetal neurologic conditions studied included myelomeningocele (n = 6), anencephaly (n = 4), and Zika-related microcephaly (n = 3). Across conditions, parents valued receiving balanced prognostic information, support in decision making, and empathy from the care

INTRODUCTION: Fetal central nervous system (CNS) abnormalities have diverse etiologies, with genetic factors as a major contributor. Prenatal exome sequencing (ES) is a powerful tool for precise molecular diagnosis of CNS anomalies, but its diagnostic yield varies among studies. This study aimed to evaluate the additional diagnostic yield of prenatal ES compared with chromosomal microarray analysis (CMA) in fetuses with CNS anomalies detected by prenatal imaging.
MATERIAL AND METHODS: We collected ES results from fetuses diagnosed with CNS anomalies by prenatal imaging (2019-2024) who had neg

Congenital pituitary hormone deficiency (CH) is a rare disorder that usually involves deficient production of growth hormone (GH) and other pituitary hormones. Many cases are syndromic, affecting other aspects of craniofacial development and sometimes other organs. There are many genes implicated in congenital pituitary hormone deficiency, and some of them also cause more severe disorders like septo-optic dysplasia (SOD) and holoprosencephaly (HPE). It is not clear why mutations in the same gene can cause such variable clinical features. In addition, the molecular etiology of many cases is not

We report a prenatal case of rare dysplastic megalencephaly detected via ultrasound and magnetic resonance imaging (MRI), showing progressive macrocephaly and abnormal cortical development. Whole-exome sequencing (WES) identified a somatic AKT1 variant (c.49G>A), confirming the diagnosis. This case highlights the integration of imaging and genetic analysis in diagnosing fetal megalencephaly, underscoring the role of mTOR pathway disorders. Pregnancy was terminated after counseling.

UNLABELLED: Three-dimensional printing (3DP) has become an established adjunct in orthopaedic surgery. Its integration into planning, education, and intraoperative execution has evolved from isolated case reports to measurable clinical outcomes. Recent research highlights reductions in operative time, blood loss, and radiation exposure when three-dimensional printed guides or models are used, with the clearest benefits in deformity correction and spinal instrumentation. Applications now extend to patient-specific instruments, custom implants, and orthotic fabrication. Emerging work on biodegra

TUBB2B encodes a β-tubulin isotype essential for neuronal proliferation, migration, and organization during brain development. Pathogenic heterozygous variants in TUBB2B are associated with neurodevelopmental disorders including polymicrogyria and corpus callosum abnormalities. However, the phenotypic spectrum remains heterogeneous, most likely reflecting variant-specific effects on microtubule formation and stability. Homozygous TUBB2B variants are exceedingly rare, with one family reported to date. We describe five individuals in four families with rare TUBB2B variants. Four variants are des

BACKGROUND: Median craniofacial dysplasia comprises a rare and heterogeneous group of midline deformities. Although embryologic and anatomic classifications describe the morphologic spectrum, practical guidance for operative decision-making remains limited. Patterns of secondary deformity observed in referral practice suggest that operative strategy may not always align with the underlying tissue substrate.
METHODS: A descriptive retrospective observational case series spanning the spectrum of median craniofacial dysplasia was reviewed to develop a hypothesis-generating conceptual operative f

Nuclear factor I (NFI) transcription factors regulate neural stem and progenitor differentiation during brain development. While NFIA, NFIB, and NFIX are linked to neurodevelopmental disorders, the role of NFIC (MIM: 600729) in human disease remains unclear. This study aimed to determine whether NFIC contributes to a neurodevelopmental syndrome, define its phenotype, and assess dosage-dependent effects. We established the first cohort of 11 individuals, including NFIC deletions and single nucleotide variants. Genotype-phenotype correlations, including critical region mapping, were performed. M

In schizophrenia spectrum disorders (SSD), structural alterations of the gray matter (GM) and white matter (WM) have been widely described. However, the complex interplay between early disease-related changes and ongoing brain maturation challenges our ability to identify early biomarkers. In this study, we investigated structural abnormalities and their association with symptoms in a drug-naïve or minimally medicated sample comprising 113 patients with SSD and 112 neurotypical controls. Specifically, we derived centile scores using normative modelling from cortical thickness (CT), and subcort

Syndromic megalencephaly comprises a heterogeneous group of genetic disorders characterized by abnormal brain overgrowth and variable structural brain abnormalities. Prenatal recognition remains challenging because imaging findings overlap with those of isolated macrocephaly, while genotype-phenotype correlations are incompletely understood. This review summarizes the current literature on fetal magnetic resonance imaging (MRI) findings in molecularly confirmed syndromic megalencephaly and discusses their relationship with the underlying genetic pathways. The available evidence indicates that

BACKGROUND: From the radiographic perspective, the septum pellucidum (SP) and septum verum (SV) Complex (SPVC) has been tacitly understood. Microdissection and diffusion tensor imaging (DTI) have now well established that they are not mere membranes but contain septal nuclei and nerve fibers; the Superior (SF) and Inferior fascicles (IF) forming the SP, and precommissural fibers of the fornix (PrCFx) in SV.
OBJECTIVE: We aimed to delineate the topography of normal and abnormal SPVC using ultrasound (US), T2-weighted magnetic resonance imaging (MRI), and DTI in fetuses and provide an algorithm

The corpus callosum (CC) is the largest structure of white matter, connecting the two cerebral hemispheres. Agenesis of the corpus callosum (AgCC) is a rare but well-recognized neurodevelopmental malformation. It can be caused by genetic and chromosomal anomalies, metabolic and toxic causes, intrauterine developmental disorders, hypoxic-ischemic insults, infections, and other causes. While attention deficit hyperactivity disorder (ADHD) is frequently associated with AgCC, there is also a significant and disproportionate prevalence of autism spectrum disorder (ASD) in these individuals compared

Following traumatic brain injury, the ability of conventional diffusion-weighted MRI analysis techniques to resolve tract-specific white matter damage, particularly in crossing fibre regions, is limited. Using fixel-based analysis, this study aimed to identify white matter abnormalities in chronic traumatic brain injury patients and to resolve the effects of traumatic brain injury on distinct white matter tracts, especially in crossing fibre regions. In this cross-sectional study, diffusion-weighted MRI were acquired from adults with chronic moderate-to-severe traumatic brain injury (N = 29; m

BACKGROUND: Prenatal alcohol exposure can cause fetal alcohol syndrome, but genetic variation can account for some of the observed differential susceptibility to alcohol-induced birth defects. Here we investigate an inbred mouse strain, 129S1/SvImJ (129S1), for its potential susceptibility to alcohol- and Sonic hedgehog pathway inhibitor-induced craniofacial birth defects and the ways complex genetic factors work to alter susceptibility compared to the highly alcohol-susceptible strain, the C57BL/6J (B6J).
METHODS: 129S1 pregnant females were treated with alcohol (two doses of 2.9 g/kg), CP-5

RATIONALE: Neonatal encephalopathy (NE) affects 1.15 million newborns annually and is associated with a high incidence of mortality and long-term neurodevelopmental disability (NDD). Therapeutic hypothermia (TH) is the only proven therapeutic intervention available. However, it is only partially effective, and evidence suggests it is not safe in low-income countries. Melatonin is a promising neuroprotective treatment in NE as it provides anti-inflammatory properties. Pre-clinical models have demonstrated improved outcomes with melatonin. However, its effect on human newborns is unclear.

UNLABELLED: Polyhydramnios, Megalencephaly, and Symptomatic Epilepsy syndrome (PMSE/STRADA-related disorder) is a rare neurodevelopmental disorder characterized by megalencephaly (ME), early-onset drug-resistant epilepsy, neurocognitive impairment, and high early mortality, often due to status epilepticus. PMSE is caused by a multi-exon deletion in STRADA , encoding STRADA, which regulates the mechanistic target of rapamycin (mTOR) pathway. GABAergic inhibitory interneurons (INs) critically modulate the excitatory:inhibitory balance in cortical and hippocampal networks, and IN deficits contrib
