Consensus-based detection of aetiologic copy number variants for syndromic orofacial clefts utilising whole exome sequencing of case parent trios.

BACKGROUND: Orofacial clefts (OFCs) are the most common craniofacial congenital anomalies, with complex aetiology involving both genetic and environmental factors. Most genetic studies on the condition have focused on the contribution of single nucleotide variants (SNVs) and small insertions and deletions (indels). However, the contribution of copy number variants (CNVs), especially in African populations, remains underexplored despite their known contribution to congenital anomalies. This study aimed to identify high-confidence CNVs potentially contributing to the aetiology of syndromic OFCs

Facial Morphology Assessment in Brazilian Children With Congenital Zika Virus Syndrome: A Cross-Sectional Anthropometric Study.

OBJECTIVES: This study aimed to conduct an anthropometric analysis of children with microcephaly caused by Zika virus infection and to compare results with those of a control group. MATERIALS AND METHODS: The study included 12 children with congenital microcephaly (experimental group) and 15 normosystemic children (control group), all aged 4-5 years. Eight anthropometric facial points were marked to assess the upper, middle, and lower thirds of the face. STATISTICS: To assess normality, the Shapiro-Wilk test was used, followed by the Student's t-test to compare groups at the 95% confidence l

Laser treatment of oral vascular anomalies. A retrospective observational study.

BACKGROUND: Oral vascular anomalies, though benign, may impact comfort and aesthetics, particularly in visible or functional areas. Laser therapies have gained prominence due to their precision, minimal invasiveness, and favourable outcomes. This study aimed to analyse the laser treatment modalities for oral vascular anomalies and their respective outcomes in a population of the north of Portugal. MATERIAL AND METHODS: This retrospective study analysed 111 vascular anomalies in 95 patients treated between January 2011 and May 2025 in the University Clinics of IUCS-CESPU. Data on demographics,

'This is the Test of Our Faith': An Exploratory Study of the Religio-spiritual and Cultural Construction of Cleft Lip and Palate Among Muslim Parents in Punjab, Pakistan.

One of the most common congenital defects affecting the orofacial areas worldwide is cleft lip and/or cleft palate (CL/P), which causes a range of social, psychological, and medical challenges for affected children and their families. Several socio-cultural beliefs and superstitions are attached to this condition, but little is known about how CL/P is framed by Muslim parents. The religious, spiritual, and cultural construction of CL/P is a significant yet often overlooked aspect of the subject covered in this study. It also discusses how parents of children with CL/P use spirituality and reli

Understanding Appearance Concerns in Young People With Cleft Lip ± Palate: A Photo Elicitation Study.

ObjectiveYoung people (YP) with a cleft lip with or without cleft palate have expressed concerns about their appearance and image-sharing on social media. This study aimed to explore and understand YP's concerns, views, and experiences regarding photographs of themselves.DesignThis was an exploratory qualitative study using photo-elicitation during semi-structured interviews conducted virtually. YP provided 3 photographs of themselves they liked and 3 they did not, which were used to facilitate discussion of their appearance, thoughts, and feelings about their photographs, and social media. IRF6 Gene Polymorphisms Influence the Susceptibility of Nonsyndromic Cleft Lips With or Without Palate: A MOOSE-Compliant Meta-Analysis.

ObjectiveThe present meta-analysis has been conducted with the objective of conducting a precise examination of the association between 6 IRF6 gene polymorphisms and the risk of nonsyndromic cleft lip with or without palate (CL ± P) among various ethnic groups.MethodsThe meta-analysis incorporated a total of 53 published case-control studies. Pooled odds ratios (ORs) and 95% confidence intervals (CIs) were computed. A leave-1-out sensitivity analysis was conducted, in conjunction with an investigation into publication bias, under the dominant model. Subgroup analysis based on ethnicity was per

Unusual Association of Orofacial Cleft and Finger-Like Skin Appendages With or Without Constriction Rings: A Disorganization-Like Phenotype?

The mouse disorganization (Ds) was described in mutant mice with multiple congenital anomalies arising from the 3 germinative layers. The human homologue of Ds was proposed in a case presenting multiple congenital anomalies associated with the unique feature of a finger-like skin appendage. Since then, several cases have been reported suggesting the occurrence of a disorganization-like (Ds-like) phenotype in humans. Here, we report a case series of 7 patients presenting with the association of orofacial clefts and finger-like skin appendages. These cases, despite the similarities with Ds and a

[Research progress in regulatory variants in non-coding regions in non-syndromic cleft lip with or without cleft palate].

Non-syndromic cleft lip with or without cleft palate (NSCL/P) is one of the most common congenital craniofacial malformations, characterized by significant genetic heterogeneity and polygenic features. Historically, genetic studies of NSCL/P have primarily focused on coding region variants. However, only approximately 2% of the human genome consists of coding sequences, with the majority of variants located in non-coding regions. Recent large-scale genome-wide association studies have indicated that risk loci associated with NSCL/P are predominantly enriched in regulatory elements within non-c

Barriers and delays in access to cleft surgery among children with cleft deformities in Rwanda: a qualitative study.

OBJECTIVES: Although timely surgery is essential for improving general health and psychosocial outcomes, delays in cleft surgery remain common in low- and middle-income countries, including Rwanda, where little is known about the underlying causes. This study aimed to explore the factors influencing delays in cleft surgery in Rwanda using the four-delay framework. DESIGN: A phenomenological qualitative study was conducted between April and July 2024. We conducted 29 in-depth interviews with 15 caregivers of children with orofacial clefts and 14 healthcare providers involved in cleft care. Tra

The Suggestive Association Between the NOG rs227731 Polymorphism and Non-Syndromic Cleft Lip With or Without Palate Subtypes in a Japanese Cohort.

Non-syndromic cleft lip with or without palate (NSCL/P) ranks among the most prevalent congenital craniofacial anomalies globally. In Japan, reported rates of cleft lip and/or palate range from 1 in 500 to 1 in 700 live births. This study aims to explore potential sex-specific interactions of NOG rs227731 across NSCL/P subtypes in a Japanese population. This case-control study included 420 participants recruited from the Cleft Lip and Palate Centre at Aichi Gakuin Dental Hospital in Nagoya, Japan. The study population was divided into three groups: 110 individuals with non-syndromic cleft lip

Unilateral Cleft Lip Repair: Reproducibility, Accessibility, and Durability.

Unilateral cleft lip (UCL) repair has evolved significantly over the past decades. In certain areas of the world, barriers such as limited surgical expertise, lack of equipment, and restricted access to general anesthesia make cleft care challenging. Simplified, reliable, reproducible, and time-tested techniques are essential to ensure equitable outcomes and reduce the need for revision surgeries. With over 4 decades of refinement, the technique described in this chapter incorporates key maneuvers that yield consistent results. It is easy to teach and applicable to all cleft severities, includ
