Serological and molecular investigation of Toxoplasma gondii in domestic cats in Dhaka City, Bangladesh.

Toxoplasma gondii is a globally prevalent zoonotic protozoan parasite with a complex life cycle, using felids as definitive hosts and various warm-blooded animals as intermediate hosts. This study aimed to assess the sero-molecular prevalence and risk factors of T. gondii infection in domestic cats in the Dhaka Metropolitan Area, Bangladesh. Blood and fecal samples from 184 cats were collected and analyzed using both enzyme-linked immunosorbent assay (ELISA) and conventional polymerase chain reaction (PCR) targeting the REP-529 gene. The seroprevalence of T. gondii was determined as 9.78% (18/

Protist oxygen-sensing and the role of prolyl hydroxylase-regulation of E3(SCF) ubiquitin ligases.

A primary mechanism for sensing O2 in animals involves the O2-dependent prolyl hydroxylase PHD2, which generates a degron on the transcription co-factor HIF-α. In protists, its apparent evolutionary predecessor, PhyA, is dedicated to the modification of Skp1. Skp1 is a subunit of the E3(SCF) family of ubiquitin ligases whose impact on the proteome is mediated by multiple F-box protein (FBP) substrate receptors. In the social amoeba Dictyostelium, genetic manipulations of PhyA and Skp1 levels lead to reciprocal effects on an O2-dependent developmental transition. PhyA is also required for fitne

A Systematic Review and Meta-Analysis of Diagnostic Test Accuracy for Human Toxoplasmosis: Performance, Populations, and Validation Gaps.

BACKGROUND: Accurate and timely diagnosis of toxoplasmosis is crucial for clinical management, particularly in high-risk populations. Despite a diverse diagnostic landscape spanning laboratory-based serology, molecular assays, point-of-care (POC) devices, and emerging technologies, a consolidated quantitative synthesis of their performance and clinical readiness is lacking. METHODS: This systematic review and diagnostic test accuracy meta-analysis employed a dual-method design, integrating evidence mapping and quantitative synthesis. We searched Embase, PubMed, Scopus, IEEE Xplore, and other

Comparison of the efficacy of cell wall disruption methods for Saccharomyces cerevisiae INVSc1 cells expressing Toxoplasma gondii recombinant ROP6 protein.

Toxoplasma gondii infects both humans and animals, causing severe clinical manifestations. Due to the lack of a safe vaccine, the development of a safe and effective vaccine against toxoplasmosis remains a significant need. Saccharomyces cerevisiae is a well-established recombinant protein expression system with Generally Recognized as Safe status. It supports post-translational glycosylation and is employed in several licensed vaccines, including hepatitis B (Engerix-B), human papillomavirus (Gardasil), and malaria (Mosquirix). Recombinant T. gondii proteins expressed in S. cerevisiae have be

Minimum inoculum of resistance assay for evaluating for anti-toxoplasmosis compounds that target phenyl alanine tRNA synthetase.

Toxoplasma gondii is a globally important intracellular parasite, and treatment regimens are limited by the failure of drugs to target latent tissue cysts. Developing new candidates for treatment also needs to address the potential for resistance to arise. Here, we developed a Minimum Inoculum for Resistance (MIR) assay as a quantitative metric for evaluating inhibitors of T. gondii. The MIR assay, adapted from assays used in malaria drug discovery, measures the frequency for pre-existing resistance alleles by exposing different sized parasite populations to drug pressure. We profiled a series

Source attribution of foodborne pathogens in the Netherlands using structured expert elicitation.

Foodborne pathogens represent a significant public health burden. Quantifying the relative importance of various potential sources of foodborne infection is challenging due to data scarcity and uncertainty in empirical studies. Structured expert judgment (SEJ) provides a valuable methodological alternative to gain insights into source attribution of foodborne pathogens. We conducted a SEJ study to attribute human cases of 26 foodborne pathogens in the Netherlands to seven major transmission pathways, 20 food groups, and two animal groups, in a typical post-COVID19 year, using Cooke's classical

Subretinal parasitic lesion mimicking viral retinitis managed with combined medical therapy and laser photocoagulation.

Parasitic infections can present with variable manifestations and may mimic viral retinitis, particularly when intraocular inflammation is prominent. We report a case of a 54-year-old woman experiencing worsening blurred vision in her right eye for over a month. She was previously diagnosed with acute retinal necrosis and treated with corticosteroids and acyclovir, but her condition continued to decline. Fundus examination showed dense vitritis and an ill-defined area of retinitis along the superior temporal arcade. Serologic tests were positive for Toxocara IgG and Toxoplasma gondii IgG, whil

Host-directed therapeutic strategies against apicomplexan parasites: targeting purinergic P2 receptors.

Apicomplexan parasites establish intracellular infections that profoundly alter host cell physiology and elicit complex immune responses. The long-standing coevolution between these parasites and vertebrate hosts has resulted in extensive overlap between parasite and host metabolic pathways, limiting the feasibility of conventional parasite-centered therapeutic approaches. Increasing evidence indicates that host-derived signals generated during infection play a decisive role in shaping parasite survival and dissemination. Among these signals, extracellular nucleotides released in response to c

Emerging insights into chemistry and therapeutic potentials of functionalized hexahydroquinolines.

Quinolines are well-established scaffolds in drug discovery and form the core of many FDA-approved medicines. Among their reduced analogues, hexahydroquinolines (HHQs) have gained increasing attention due to their structural adaptability and broad pharmacological relevance. Research has largely focused on the 1,4,5,6,7,8-hexahydroquinoline framework, although interest in other variants, including 2-oxo derivatives, is steadily growing. This review summarizes recent progress in HHQ chemistry, with emphasis on synthetic strategies and emerging therapeutic applications. Traditional multicomponent

Toxoplasma gondii affects trait anxiety in adult ADHD.

BACKGROUND/OBJECTIVE: Growing evidence emerges that Toxoplasma gondii (T. gondii) is associated with mental disorders like anxiety disorders or attention-deficit/hyperactivity disorder (ADHD). In ADHD patients around 25% suffer from comorbid anxiety disorders. As the impact of a latent T. gondii infection on anxiety in adult ADHD remains unknown, this study aims to investigate this relationship. METHODS: In a case-control study, including 140 participants, venous blood samples were taken of 70 adult ADHD patients and 70 controls for serological analysis of markers of infection and inflammatio

Immunity to helminths in complex environments: co-infection disrupts effective granuloma formation and parasite clearance.

INTRODUCTION: The intestinal roundworm Heligmosomoides bakeri establishes chronic infections in susceptible C57Bl/6 mice, yet repeated ('trickle') infections confer immunity and promote worm clearance. We previously linked this acquired immunity to enhanced intestinal granuloma formation. Here, we focused on effector molecules (antibodies) and cells (eosinophils and macrophages) known to immobilize and damage developing parasites. METHODS AND RESULTS: We used flow cytometry and immunofluorescence to show that granulomas in trickle-infected female mice contain elevated levels of IgG1, SiglecF+

Current research trends towards the control of protozoans in foods.

Protozoan parasites such as Cryptosporidium spp., Giardia duodenalis, Toxoplasma gondii and Cyclospora cayetanensis remain difficult-to-control hazards in food due to environmental persistence, low infectious doses, and the interpretability gap between nucleic acid detection and infectivity. This review synthesizes 4-year research trends shaping protozoan control in food systems, focusing on three critical pillars: matrix-adapted front-end processing (concentration, lysis, inhibitor management); inhibitor-resilient quantification; and sequencing-based attribution for outbreak investigation and

Glancing at the role of Toxoplasma gondii in neuro-oncology: from risk factor to therapeutic target.

BACKGROUND: Cancer is a leading cause of morbidity and mortality, with brain tumors posing significant challenges due to their site and complexity. Therefore, exploring tumor pathogenesis and the discovery of novel therapeutic agents have drawn attention to the role of infectious agents. Toxoplasma gondii is a highly prevalent protozoan parasite that establishes a chronic latent infection in neural tissues. Several epidemiological studies have linked infection with Toxoplasma gondii to a higher risk of developing cancer, particularly brain tumors. However, research findings are controversial,

A Unique Protein Adjuvant for Precision Immunotherapy to Prevent Recurrence of Surgically Resected Colorectal Cancer.

Effectively activating protective CD8+ T cell immunity specifically against cancer antigens is an important pathway to prevent the growth of various types of cancers. A major obstacle in this approach is variations in cancer antigens among patients. A valuable material to overcome the antigen variation among cancer patients is the use of each individual's own cancer cells for immunization. In colorectal cancer (CRC), approximately one-third of the patients who receive curative surgical resection have a recurrence of cancer. Therefore, the use of surgically resected CRC for immunotherapy to spe

Spinal Cord Toxoplasmosis: Mapping the Journey of a Rare Entity Through a Case Report and Review of the Literature.

Toxoplasmosis remains the most frequent cause of cerebral lesions in patients with acquired immunodeficiency syndrome (AIDS), especially in those not receiving prophylaxis. Medullary involvement, although rare, can cause irreversible neurological damage. When associated with fever in the returning traveler, the etiological diagnosis of spinal cord lesions can be challenging due to the wide range of diagnostic possibilities. We report a unique case of spinal cord toxoplasmosis associated with Salmonella non-typhi bacteremia after a trip to Cameroon, revealing an advanced human immunodeficiency

Cerebral Toxoplasmosis in a Renal Transplant Recipient-A Rare Complication.

Cerebral toxoplasmosis is a rare but potentially fatal opportunistic infection in renal transplant recipients receiving long-term immunosuppressive therapy. It may result from donor-derived transmission or reactivation of latent infection. We report the case of a 70-year-old female who underwent kidney transplantation from a deceased donor in 2004 for end-stage renal disease due to glomerulonephritis. She was maintained on cyclosporine, mycophenolate mofetil, and prednisone. In September 2024, she presented with headache, mood changes, and right-sided hemiparesis. Brain multislice computed tom
