BACKGROUND: Fetal alcohol spectrum disorder (FASD) represents a range of neurodevelopmental outcomes from prenatal alcohol exposure for which there are few interventions. Preclinical studies have identified choline as a potential treatment for cognitive deficits in FASD. We previously conducted three randomized controlled trials (RCTs) in children under age 6 and reported on memory benefits following all three trials. Two RCTs compared a fixed 500 mg daily dose to placebo, and the third RCT tested a weight-adjusted choline dose. Here, we report on a longitudinal follow-up 5 years post-study co

BACKGROUND: Despite public health efforts, prenatal alcohol exposure (PAE) remains a significant concern. Its effects on offspring vary depending on the amount and frequency of drinking, and the risk of Fetal Alcohol Spectrum Disorders (FASDs) increases with heavy or binge drinking. In contrast, the long-term consequences of moderate PAE remain unclear. Although previous studies indicate that moderate PAE may have a limited impact on neurodevelopment, its ability to induce persistent cerebrovascular dysfunction may promote chronic low-level oxidative stress in the brain. Therefore, it is impor

BACKGROUND AND AIMS: Alcohol consumption during pregnancy poses notable risks to maternal and foetal health, yet its prevalence and impact in Nigeria remain understudied. This narrative review aims to initiate conversations around the current state of alcohol intake by expectant mothers in Nigeria and Fetal Alcohol Spectrum Disorder (FASD), while providing recommendations to address this public health issue.
METHODS: Drawing on existing literature, we highlight the prevalence of alcohol intake by expectant mothers in Nigeria and explore the socio-cultural factors influencing drinking behavior

Fetal alcohol spectrum disorders (FASD) exhibit wide phenotypic variability not explained by prenatal alcohol exposure (PAE) alone. While genetic factors may modify vulnerability, there are no prior FASD genome-wide association studies (GWAS). We performed genome-wide single-marker and gene-based analyses of two South African prospective birth cohorts (Cape Town Longitudinal Cohorts (CTLC); n=328 mother-child dyads) and the US Collaborative Initiative on Fetal Alcohol Spectrum Disorders cohort (CIFASD n=429 children only). We examined main genetic and gene-by-PAE interaction effects on working

BACKGROUND: Recent reports indicate a decline in rates of Fetal Alcohol Syndrome (FAS) among Alaska Native and American Indian (AN/AI) infants. Nevertheless, AN/AI infants remain disproportionately impacted by the effects of prenatal alcohol exposure.
METHODS: AN/AI pregnant women in their 3rd trimester completed a questionnaire on demographic data and the amount and frequency of their alcohol consumption in the month prior to conception and during pregnancy. Differences across demographics and trimesters were tested with the Chi-square, Fisher's exact or McNemar's test as appropriate.

INTRODUCTION: International guidelines for diagnosing fetal alcohol spectrum disorder (FASD) differ regarding the required level of prenatal alcohol exposure (PAE). The Australian Guidelines for Assessment and Diagnosis of Fetal Alcohol Spectrum Disorder (2025) introduce a minimum PAE threshold, unlike the 2016 Guide to FASD Diagnosis, which allowed for diagnosis at any exposure level. This retrospective case review evaluates whether applying the minimum PAE threshold would alter diagnostic outcomes for individuals previously assessed for FASD at a specialist neurodevelopmental clinic in Queen

This chapter provides an introduction and overview of animal models that have been used to investigate the teratogenic effects of alcohol. Since the first model was developed in 1899, prenatal alcohol exposure (PAE) has been studied in species ranging from invertebrates to primates. Here, we contextualize and outline critical experimental considerations, including blood alcohol concentration, timing of exposure, and routes of ethanol administration. Detailed comparisons of vertebrate and invertebrate models, particularly rodents, guinea pigs, and non-human primates, highlight their translation

Fetal alcohol spectrum disorders (FASD) represent a significant global health concern and are the leading cause of preventable birth defects, intellectual disabilities, and persistent behavioral deficits. The neurodevelopmental mechanisms underlying FASD are not yet fully understood, but there is growing interest in the role of cortical development in the manifestation of these disorders. The cerebral cortex serves as the master regulator of higher-order functions, including sensory perception, motor planning, decision-making, and general intelligence-processes that are all adversely affected

Prenatal alcohol exposure (PAE) is recognized as a leading preventable cause of birth defects, giving rise to a continuum of cognitive, behavioral, and physical impairments collectively referred to as Fetal Alcohol Spectrum Disorders (FASD). While PAE affects multiple developing organ systems, the fetal brain is particularly vulnerable, exhibiting enduring structural and functional abnormalities in response to alcohol exposure. Recent research highlights mitochondrial dysfunction as an important mechanism in the pathogenesis of alcohol-related neurodevelopmental deficits. Mitochondria are high

Prenatal alcohol exposure impacts fetal development, leading to Fetal Alcohol Spectrum Disorders (FASD) characterized by cognitive, behavioral, and physical impairments. This pre-post, open-label, non-randomized pilot study explores Epigallocatechin-3-Gallate (EGCG), a potent antioxidant and neuronal plasticity modulator, as a therapeutic intervention for FASD improvement. In a 12-month pilot study, patients with 40 FASD (mean age 10 ± 3 years) received 9 mg/kg/day of EGCG, with outcomes assessed through RNA sequencing and neurocognitive evaluations (WISC-IV, CBCL 6-18, and NEPSY-II). Reduced

The last two decades have witnessed a remarkable transformation in the integration of artificial intelligence in healthcare. Similarly, our understanding of fetal alcohol spectrum disorders (FASD) has expanded and the data points now being considered are far more granular. In this article, we will review how investigators have attempted to apply technological innovations to enhance our knowledge of FASD and to fill some of the gaps that exist regarding access to experienced diagnosticians. We will summarize the salient studies to date and highlight future directions in terms of how the technol

BACKGROUND: Fetal alcohol spectrum disorder (FASD), caused by prenatal ethanol exposure, encompasses developmental abnormalities such as craniofacial dysmorphism, sensory and motor impairments, and neurocognitive deficits. MicroRNAs have been proposed as key regulators in FASD pathophysiology, yet the specific molecular pathways they influence remain unclear.
METHODS: To better understand the role of microRNAs in the progression of FASD, we have systematically reviewed the effects of prenatal exposure to alcohol on microRNAs. The search was performed through PubMed MEDLINE, web of science, Sc

BACKGROUND: Prenatal alcohol exposure (PAE) increases the risk for serious adverse birth outcomes and long-term neurodevelopmental, physical, and mental health outcomes, known under the umbrella term of fetal alcohol spectrum disorders (FASD). Despite its well-documented adverse effects, PAE continues to be prevalent in the United States (U.S.), where an estimated 13.5% of women report alcohol consumption during pregnancy. Brief interventions to reduce PAE delivered during prenatal care have obtained mixed results, in part due to obstetricians' high workload and short prenatal care visits. To

This case report presents a complex psychiatric profile of a 19-year-old female with diagnoses of Fetal Alcohol Syndrome (FAS), Autism Spectrum Disorder (ASD), Attention-Deficit/Hyperactivity Disorder (ADHD), mild intellectual disability, and mood dysregulation, complicated by trauma history, psychotic features, and repeated hospitalizations. Her neurodevelopmental and psychiatric disorders have contributed to persistent high-risk behaviors, legal involvement, and challenges with independent living, ultimately necessitating group home placement. This case highlights the unique relationship bet

Fetal Alcohol Spectrum Disorder (FASD), caused by prenatal alcohol exposure (PAE), produces lasting physical, cognitive, and behavioral impairments. The present study examined effects of early PAE on the gut microbiome (GM) in adolescent mice to identify targets for early intervention. Female C57Bl/6 dams received 10 % ethanol during the first 10 days of gestation while controls received water. Fecal and blood samples from adolescent offspring were profiled by 16S rRNA sequencing and gas chromatography-mass spectrometry to characterize microbial composition and short-chain fatty acids (SCFAs).

The evaluation and management of children with prenatal alcohol exposure and suspected fetal alcohol spectrum disorders (FASD) requires a multidisciplinary approach, grounded in the pediatric medical home. This process begins with the primary care provider who is often the initial point of contact for children presenting with developmental and behavioral concerns. An evaluation for a FASD requires the primary care provider to consider the effects of biological risk factors (e.g. genetic and mental health risks, maternal pregnancy risks, and history or prenatal substance exposures), and the rol

This study explores the long-term effects of embryonic ethanol exposure on cognitive functions and gene expression in zebrafish (Danio rerio). We hypothesized that ethanol exposure during a critical developmental stage would lead to deficits in executive functions, such as working memory and behavioral flexibility, as well as alterations in neurodevelopmental gene expression. Zebrafish embryos were exposed to ethanol for 2 h at 24 hpf (hours post fertilization), and behavior was assessed at the fry (15 days post-fertilization), juvenile (45 dpf), and adult (90 dpf) stages. The Y-FMP behavioral

OBJECTIVES: Fetal alcohol syndrome (FAS) is frequently undiagnosed or misdiagnosed, particularly among populations at elevated risk of the condition. We examined the prevalence of FAS among children aged 0 to 5 years enrolled in Medicaid, described characteristics of affected children, and evaluated diagnostic timing between children in foster care and children not in foster care.
METHODS: We conducted a retrospective analysis of Medicaid Transformed Analytic Files for children born from 2015 through 2017 with FAS diagnoses (n = 771), following each birth cohort for 5 years. We used descripti

CONTEXT: Foetal Alcohol Syndrome (FAS) is the most severe form of Foetal Alcohol Spectrum Disorders (FASD), caused by prenatal alcohol exposure, a major preventable cause of congenital disability. FASD lack specific prenatal ultrasound signs, making early diagnosis difficult.
OBJECTIVE: To describe prenatal ultrasound findings in pregnancies with reported alcohol use and compare maternal and neonatal characteristics according to the presence of anomalies.
METHODS: We conducted a retrospective multicentre study in three hospitals from September 2013 to April 2025. All women reporting alcohol
