Five ways to find a parasite: comparison of five molecular assays for the detection of Strongyloides spp.

UNLABELLED: Strongyloidiasis, caused by nematodes of the genus Strongyloides, is a neglected tropical disease with significant public health implications. Accurate diagnostics are essential for disease surveillance and control, yet traditional stool-based and serological methods have limitations in sensitivity and specificity. Molecular assays offer an alternative diagnostic approach. In this study, we compared four probe-based real-time PCR (qPCR) assays and one loop-mediated isothermal amplification (LAMP) assay using human fecal DNA samples from Gabon and Papua New Guinea with both Strongyl

Severe strongyloidiasis and Gram-negative bacterial superinfections: epidemiology, clinical implications, and therapeutic strategies.

PURPOSE OF REVIEW: Strongyloidiasis is a neglected tropical disease affecting over 600 million people worldwide. Its severe forms carry a fatality rate exceeding 60%, yet death is rarely attributable to the parasite alone: the terminal event is often bacterial sepsis or meningitis of enteric origin. We review recent evidence on Gram-negative and other bacterial superinfections in severe strongyloidiasis, and their diagnostic and therapeutic implications. RECENT FINDINGS: Cohort studies show that bacteraemia is the commonest complication of severe disease, affecting 38.3% of cases against 4.1%

Prevalence of Strongyloides stercoralis infection among African children: A systematic review and meta-analysis.

Strongyloides stercoralis is a neglected parasite that causes strongyloidiasis. However, the true prevalence of strongyloidiasis is underreported due to low-sensitivity diagnostic methods. This systematic review assessed the prevalence of S. stercoralis among African children and its variation by diagnostic methods, countries, and study settings. All studies published in African countries on S. stercoralis prevalence up to the year 2025 in the PubMed, Google Scholar, Scopus, and ScienceDirect databases were included. Identification, screening, checking the eligibility, and including the releva

Integration of the Triboelectric Effect into a Yolk-Shell Conjugated Microporous Polymer for Enhanced Artificial Photosynthesis.

Conjugated microporous polymers (CMPs) have demonstrated considerable potential in the field of photocatalytic organic synthesis. However, the issue of intrinsic photogenerated carrier recombination severely limits their potential applications. Herein, a strategy for boosting the photocatalytic performance of CMP through the triboelectric effect is proposed. Specifically, the benzothiadiazole-involved CMP (BTTP) with inherent triboelectric character is processed into yolk-shell SiO2@BTTP materials, in which the BTTP and SiO2 function as tribo-negative and tribo-positive materials, respectively

Cerebral Schistosoma japonicum infection confirmed by histopathology and PCR: Diagnostic and therapeutic challenges in multiple parasitic coinfections.

Cerebral schistosomiasis, a rare manifestation of schistosomiasis, is challenging to diagnose due to non-specificity in the clinical and radiologic findings and cross-reactivity in serologic testing with other helminths. Coinfection with multiple parasites further complicates diagnosis and treatment. A previously healthy 32-year-old Filipino man presented with a seizure. Brain magnetic resonance imaging (MRI) revealed multiple clustered enhancing nodules with surrounding vasogenic edema in the left frontal lobe. Serologic testing was positive for anti-Schistosoma japonicum, Strongyloides, and

Hunting for Helminths: short- and long-read shotgun metagenomics for helminth detection in faecal samples.

Soil-transmitted helminths (STHs) pose significant challenges to public health in endemic areas, necessitating reliable methods for their detection. Shotgun metagenomics enables simultaneous detection of STHs and microbes in a sample without prior knowledge of what is present. However, validation of shotgun metagenomics with known infection intensity or across different sequencing platforms has not been carried out for eukaryote parasites including STHs, and false positives remain a pervasive issue. We validated shotgun metagenomics as a method of STH detection in faecal samples. Using the Str

Profound Hemolytic Anemia in Babesiosis With Lyme Disease Coinfection and Occult Strongyloidiasis.

Babesiosis is a tick-borne infection caused by intraerythrocytic Babesia parasites and may result in severe hemolytic anemia and systemic complications. Coinfection with other Ixodes-borne pathogens may further complicate diagnosis and management. We report a 45-year-old immunocompetent man working as a landscaper in Connecticut who presented with syncope following two weeks of fever, fatigue, diaphoresis, arthralgias, jaundice, and dark urine. Laboratory evaluation revealed profound hemolytic anemia with a hemoglobin of 6.8 g/dL, elevated lactate dehydrogenase (LDH), undetectable haptoglobin,

Dissonance in Kashechewan: Reflections from a physician and the daughter of refugees.

Alterations in the immunologic response induced by human T cell lymphotrophic virus type I (HTLV-I) predispose the development of disseminated strongyloidiasis. We report a case of an atypical clinical presentation of strongyloidiasis in a patient co-infected with HTLV-I causing scrotal and perineal pain and infertility. Strongyloides stercoralis was found in the analysis of the sperm and specific therapy for strongyloidiasis was associated with disappearance of the symptoms.

Inflammatory bowel disease vs strongyloidiasis.

We report the case of a 38-year-old woman with a 10-year history of ulcerative proctosigmoiditis. Two months after traveling to Morocco, she developed gastrointestinal symptoms accompanied by eosinophilia in blood tests. Four months later, she progressed to systemic illness with pulmonary involvement. A colonoscopy revealed aphthous ileitis with nonspecific histology suggestive of Crohn's Disease (CD). Ultimately, serology confirmed Strongyloides stercoralis infection. Strongyloidiasis is underdiagnosed in Spain, and its differentiation from CD presents a diagnostic challenge.

Illustrative case series of eosinophilia in UK Armed Forces personnel and recommended management for non-specialists.

Peripheral eosinophilia (>0.5×109/L) in the full blood count is caused by a variety of infectious and non-infectious aetiologies. Of particular importance to the UK military are parasitic infections, especially in individuals recruited from overseas and those who have deployed to areas that are highly endemic for schistosomiasis, strongyloidiasis and soil-transmitted helminths. These infections may persist for decades without causing symptoms. UK Armed Forces (UKAF) personnel have recently presented with more severe forms of strongyloidiasis, which can be fatal, especially following immunosupp

Taxonomy of Strongyloides in humans, dogs and cats: a comprehensive review from morphology to molecular and population genetics.

The genus Strongyloides (Nematoda; Strongyloididae) comprises over 50 species of nematodes parasitic in terrestrial vertebrates, including humans (Homo sapiens), dogs (Canis lupus familiaris) and cats (Felis catus). Taxonomy of the genus has been shaped by over a century of morphological research, with the most widely adopted framework established in the late 1980s. Advances in molecular genetics have increasingly revealed cryptic diversity and yielded new insights into interspecific and intraspecific relationships within the genus. Despite the rapid expansion of molecular genetic data over th

Retrospective screening reveals the rare occurrence of zoonotic Strongyloides stercoralis in dogs from temperate Australia, 2014-2024.

Strongyloides stercoralis is an intestinal nematode capable of infecting humans and dogs. Occurrence in dogs from temperate, traditionally non-endemic regions remains poorly characterized, often due to the absence of accessible veterinary diagnostic tests. Recent reports of infections from temperate metropolitan areas have raised concerns about the extent of S. stercoralis prevalence in dogs in these regions. This study investigated the presence of S. stercoralis DNA in canine faecal samples from Sydney, New South Wales, Australia, using a real-time quantitative PCR (qPCR) targeting the 18S rR

Seroprevalence of Strongyloides stercoralis Infection and Its Risk Factors in Pregnant Women Referred to Al-Zahra Hospital in Guilan Province, North Part of Iran: A Cross-Sectional Study From 2024 to 2025.

BACKGROUND: In immunocompromised individuals, strongyloidiasis may progress to hyperinfection syndrome or disseminated disease. To date, the death of a pregnant woman infected with Strongyloides stercoralis has been reported. Chronic cases carry a risk of malnutrition, especially among vulnerable populations like pregnant women and children. Pregnancy-related immunosuppression increases susceptibility to severe strongyloidiasis. This study assesses strongyloidiasis seroprevalence in pregnant women at Al-Zahra Hospital, Guilan Province. An Agar-Free, Glass Bead-Based Method for the Culture of Strongyloides stercoralis: An Exploratory Diagnostic Sensitivity Study.

Background/Objectives: The diagnosis of infection with Strogyloides stercoralis, recently targeted for control by the World Health Organization, (WHO), is challenging. Specific coproparasitological methods (agar plate culture [APC], Baermann sedimentation), recommended by the WHO for public health use, are labor-intensive and require bulky disposable materials as well as experienced microscopists. We explored the sensitivity of an alternative stool culture method using recyclable glass beads, followed by microscopy and PCR in comparison to routine APC and PCR performed on uncultured stool. Strongyloidiasis Beyond the Tropics: Updated Epidemiological Evidence from a Historically Endemic Region in Spain.

Strongyloidiasis is traditionally regarded as a tropical disease; however, in the Valencian Community (Spain), it has historically been linked to localized endemic foci considered of declining relevance. This was a multicenter regional case series study across three hospitals including patients ≥16 years with strongyloidiasis defined by positive serology and/or parasitological confirmation diagnosed from 2015 to 2024. A total of 301 patients were included (median age 53 years (quartile 1-quartile 3, 40-72); 135 (44.9%) female). Most cases were autochthonous (176/299, 58.9%), while 123/299 (41.

A case of duodenal strongyloidiasis presenting as a gastric outlet obstruction and endoscopically mimicking malignancy: A case report and mini review of literature.

INTRODUCTION: Strongyloidiasis is a neglected soil-transmitted helminthiasis caused by Strongyloides stercoralis, affecting an estimated 300-600 million people worldwide. Due to its ability to cause autoinfection, the parasite may persist lifelong and lead to severe complications. While gastrointestinal symptoms are common, presentation as gastric outlet obstruction (GOO) is rare and can mimic malignancy. CASE PRESENTATION: A 42-year-old male presented with several months of non-projectile vomiting, epigastric pain, indigestion, anorexia, and significant weight loss. Examination revealed dehy
