Closing the Surveillance-to-Stewardship Gap in antimicrobial resistance: a narrative review of global lessons from WHO GLASS, UK ESPAUR, and low- and middle-income countries.

BACKGROUND: Antimicrobial resistance (AMR) surveillance has expanded faster than any other component of the global response, yet resistance continues to rise. In a growing number of settings, the principal constraint is the failure to translate available data into action, a disconnect we describe as the Surveillance-to-Stewardship (S2S) Gap. OBJECTIVES: To define the S2S Gap and its component gaps, examine where it binds across health systems of different maturity, analyse why it persists, and propose an operationalisable, testable framework for closing it. SOURCES: A cross-sectional analysi

Parental son preference in childhood and sex differences in the risk of cardiovascular disease in middle-aged and older adults.

INTRODUCTION: Sex differences in cardiovascular disease (CVD) risk are examined through biological and clinical factors, with less attention to early-life social exposures. This study examined associations of childhood parental son preference with CVD risk, sex differences, and mediation by modifiable risk factors. METHODS: This cohort analysis included China Health and Retirement Longitudinal Study participants aged ≥45 years without baseline CVD. Parental son preference was assessed retrospectively in 2014; incident CVD was self-reported physician-diagnosed heart disease or stroke through 2

Neural Organization of Episodic Memory and Navigation in Children and Adults.

Episodic memory and spatial navigation are often viewed as interlinked, in part due to their shared reliance on medial-temporal lobe (MTL) structures. At the same time, there are important differences, for example, natural spatial navigation requires moving one's body through space while episodic memory does not. We investigated whether naturalistically encoded memories that were either episodic or additionally embedded in a navigational experience relied on overlapping or distinct neural networks. Children ages 8-13 years old and young adults viewed pictures of objects that they had previousl

Development and Application of Expressed Sequence Tags (ESTs) for the Improvement of Cotton Crops.

Expressed sequence tags (ESTs), are short cDNA sequences obtained from expressed genes in any plant tissue of Gossypium spp. and represent a suitable approach for studying the transcriptome in the absence of a reference genome. In this study, protocols for EST development were established to explore expressed genes in cotton under diverse biotic and abiotic stress conditions. ESTs were generated by using two approaches: suppression subtractive hybridization (SSH) and next-generation sequencing (NGS) techniques. The process begins with RNA isolation, followed by mRNA purification, cDNA synthesi

RAPD Marker Optimization and Genetic Diversity Assessment in Averrhoa Genus.

The genus Averrhoa includes economically and nutritionally important fruit crops like Averrhoa carambola (star fruit) and Averrhoa bilimbi (bilimbi). However, it has not been studied enough genetically, even though it is important for horticulture. For germplasm conservation, cultivar identification, and crop improvement programs, it is important to be able to accurately measure genetic diversity. This chapter elucidates the optimization and utilization of random amplified polymorphic DNA (RAPD) markers for the analysis of genetic diversity in Averrhoa species. We used a modified CTAB method t

Application of Single Nucleotide Polymorphisms (SNPs) in Sesame (Sesamum indicum L.) for Genetic Diversity, Mapping, and Marker-Assisted Selection.

Sesamum indicum L. is one of the oldest oilseed, cultivated oleaginous species, which is highly valued due to the excellent quality of the oils and owing to its antioxidative effects. Even though it is a major economic crop, there is a historical gap in breeding of sesame compared to the major staple crops, mainly because of a lack of genomic resources. With the emergence of next-generation sequencing methods, single-nucleotide polymorphisms (SNPs) with their noted benefits, compared to older marker systems, such as dense marker coverage, whole-genome coverage, and high-throughput automation c

Non-Sequencing-Based Microsatellite Development in Pomegranate (Punica granatum): Enrichment and Screening Protocol.

The chapter reports an inexpensive, non-sequencing-based technique to develop microsatellite markers in pomegranate (Punica granatum). Microsatellites are also known as the polymorphic genetic markers that are usually used in genetic diversity, breeding, and population genetics. Even though next-generation sequencing (NGS) has quickly demonstrated speedy advancement in the development of markers, the given protocol provides a viable option in species with limited genomic information or in cases when sequencing is not a viable option. The strategy uses the enrichment strategy as a way of select

Application of Restriction Fragment Length Polymorphism (RFLP).

Restriction fragment length polymorphism (RFLP) was the first reliable genetic marker used in plant molecular studies. It is a DNA-based marker that detects variation in DNA sequence at restriction enzyme sites and produces codominant, locus-specific markers that differentiate homologous and heterozygous genotypes. It provides greater accuracy than trait-based methods. The application of RFLP markers includes developing genetic linkage maps, conducting QTL analysis, characterizing germplasm, and detecting somaclonal variations. Despite being the availability of new molecular markers, RFLP stil

SSR Marker-Assisted Selection and Monitoring of High-Metabolite Cell Lines in Plant Tissue Culture: Protocols and Somaclonal Variation Assessment.

Simple Sequence Repeat (SSR) marker-assisted selection has proven to be the most powerful tool for genetic monitoring and selection in in vitro culture systems. It is widely used in detecting somaclonal variation and producing true-to-type in vitro plantlets with superior quality. Genetic fidelity is the critical point in plant tissue culture, and SSR markers act as a supporter to pass this critical point through locus specificity, codominancy and high polymorphism. SSR markers allow the precise detection of genetic changes during callus formation, organogenesis, somatic embryogenesis, and lon

Application of SSR Markers for the Detection of Metal Tolerance in Plants.

The dynamic flow of heavy metals in ecosystems is disturbing the sustainability of environmental quality and the flow of resources in ecosystems. Detection of genotypes with metal-tolerance ability is a prerequisite for mitigating this burning problem, through restoration of degraded ecosystems, progressive crop productivity, and improvement in efficiency of phytoremediation programs. The application of simple sequence repeat (SSR) markers for tracing metal tolerance in plants is gaining importance among researchers. SSRs, as molecular markers, are highly informative and preferable compared to

Chromatin Immunoprecipitation Followed by Next-Generation Sequencing in Ewing Sarcoma.

Chromatin immunoprecipitation followed by sequencing (ChIP-seq) is a widely used technique to study the distribution of transcription factors, histones, posttranslational modifications, or other DNA-associated proteins across the genome. This methodology is based on the isolation of DNA fragments bound by the protein of interest using specific antibodies. Computational analysis of sequencing libraries derived from the isolated DNA fragments will delineate genome-wide maps of protein-DNA interactions. ChIP-seq profiling of EWSR1::FLI1 oncogene has enormously contributed to the understanding of

Effects of Pretreatment on the Isotopic Composition of Structural Carbonate in Ray-Finned Fish Teeth.

RATIONALE: The carbon and oxygen stable isotope ratios (δ13C and δ18O) of carbonate contained in fish tooth dentin have the potential to offer insights into environmental and ecological conditions, making them promising targets for analysis in fossil specimens. However, the tooth dentin of extant specimens contains significant amounts of organic matter that can decompose or produce gases that interfere with isotope measurements. Therefore, a well-developed pretreatment protocol for removing organic components prior to isotopic analysis is essential. METHODS: In this study, we developed pretre
