Lysosomal storage disorders: emerging therapeutic options require early diagnosis.

Lysosomal storage disorders have been recognised as one of the major groups of genetic disorders affecting children and adults. With over 40 different disorders and a combined prevalence of up to 1:5000 births, this group of disorders is a major public health problem and places an enormous burden on the individuals and families affected. Since the introduction of enzyme replacement therapy for Gaucher disease over 10 years ago there has been considerable progress in the development of enzyme based therapies for other disorders, in addition to alternate therapies including substrate deprivation

Cathodic Quantum Dot Facilitated Electrochemiluminescent Detection in Blood.

The expansion of electrochemical sensors to biomedical applications at point of care requires these sensors to undergo analysis without any pretreatment or extraction. This poses a major challenge for all electrochemical sensors including electrochemiluminescent (ECL)-based sensors. ECL offers many advantages for biomedical applications; however, obtaining results from complex matrixes has proven to be a large hurdle for the application of ECL sensors within this field. This work demonstrates the potential of cathodic ECL to detect and quantify homocysteine (Hcy) with a 0.1 nM limit of detecti

Organoids: generation strategies, applications, and future challenges.

Organoids are microscopic 3D structures that resemble real organs, assembled in vitro from stem cells. Since the Dutch team of Hans Clevers successfully cultured intestinal stem cells to generate intestinal structures in 2009, organoid technology has developed rapidly, and culture protocols covering various organs such as the brain, liver, intestine, kidney, and bone have been established. These micro-organ models retain the cellular heterogeneity, tissue-specific structure, and genetic background of the original tissue. Compared to traditional two-dimensional culture, they provide a more phys

Conductive/Insulating Bioinks with Multitechnology Compatibility and Adjustable Performance.

Conducting/insulating inks have received significant attention for the fabrication of a wide range of additive manufacturing technology. However, current inks often demonstrate poor biocompatibility and face trade-offs between conductivity and mechanical stiffness under physiological conditions. Here, conductive/insulating bioinks based on two-dimensional materials are proposed. The conductive bioink, graphene (GR)-poly(lactic-co-glycolic acid) (PLGA), is prepared by introducing conductive GR into a degradable polymer matrix, PLGA, while the insulating bioink, boron nitride (BN)-PLGA, is synth

Imaging technologies and basic considerations for welfare of laboratory rodents.

Imaging technologies are regularly used in biomedical research to study processes in living animals in a noninvasive manner. But imaging procedures can affect animal physiology, and the need to anesthetize animals for imaging entails potential health risks. In addition, certain imaging modalities require the use of ionizing radiation or the administration of contrast agents or imaging biomarkers, which also have consequences for animal physiology. Finally, procedures associated with imaging, such as animal preparation (e.g., fasting, premedication) and blood sampling, can also affect physiolog

Electromagnetohydrodynamic pulsatile blood flow of Casson nanofluid with stenosis porous artery under periodic body acceleration and slip effects: A Reynolds and Vogel viscosity models.

Nanofluid technology is increasingly relevant in biomedical engineering for enhancing diagnostics and targeted therapies. This study examines the electromagnetohydrodynamic behavior of an Au-blood nanofluid through a tapered, porous, stenosed artery. Blood is modeled as a non-Newtonian Casson fluid with temperature-dependent viscosity, described using Reynolds' and Vogel's models. The governing equations, which incorporate slip, body acceleration, electro-osmosis, magnetic effects, and nanoparticle suspension, are solved using a finite difference scheme and validated against benchmark studies.

Developing and delivering new medical technologies: issues beyond access.

The articles in this issue illuminate psychosocial issues raised by the development and delivery of new medical technologies. Five kinds of questions surface repeatedly: questions about a technology's purpose(s), the value judgments it presumes, the locus of its control, the external forces that drive it, and its long-term social risks. These questions take the discussion of new technologies beyond the challenge of improving access to their benefits. They also raise issues that will become increasingly important as the capabilities of medical technology expand. For example, they suggest resear

Genetic modification of somatic cells for producing animal models and for cellular transplantation.

Great progress has been made in two technologies related to biomedical research: (1) manipulating the genomes of cells; and (2) inducing stem cells in culture to differentiate into potentially useful cell types. These technologies can be used to create animal models of human disease and to provide cells for transplantation to ameliorate human disease. Both embryonic stem cells and adult stem cells have been studied for these purposes. Genetically modified somatic cells provide another source of cells for creating animal models and for cellular transplantation.
