Leveraging Extracellular Non-coding RNAs to Diagnose and Treat Heart Diseases.

Extracellular vesicles (EVs), including exosomes and microvesicles, emerge to be crucial mediators of cell-to-cell communication in multiple organs. Non-coding RNAs loaded inside EVs contribute as one major mechanism for remote information transfer among different cell types or organs. Increasing evidence suggests that EV-associated non-coding RNAs derived from cardiovascular or non-cardiac cells regulate cardiovascular pathophysiology in heart development and diseases. The functional relevance of the EV-associated ncRNAs in heart diseases provides an avenue to develop novel diagnostic tools a

Noncoding RNA as Diagnostic and Prognostic Biomarkers in Cerebrovascular Disease.

Noncoding RNAs (ncRNAs), such as microRNAs, long noncoding RNAs, and circular RNAs, play an important role in the pathophysiology of cerebrovascular diseases (CVDs). They are effectively detectable in body fluids, potentially suggesting new biomarkers for the early detection and prognosis of CVDs. In this review, the physiological functions of circulating ncRNAs and their potential role as diagnostic and prognostic markers in patients with cerebrovascular diseases are discussed, especially in acute ischemic stroke, subarachnoid hemorrhage, and moyamoya disease.

Current Diagnostic Methods and Non-Coding RNAs as Possible Biomarkers in Huntington's Disease.

Whether as a cause or a symptom, RNA transcription is recurrently altered in pathologic conditions. This is also true for non-coding RNAs, with regulatory functions in a variety of processes such as differentiation, cell identity and metabolism. In line with their increasingly recognized roles in cellular pathways, RNAs are also currently evaluated as possible disease biomarkers. They could be informative not only to follow disease progression and assess treatment efficacy in clinics, but also to aid in the development of new therapeutic approaches. This is especially important for neurologica

Sperm-borne sncRNAs: potential biomarkers for semen fertility?

Semen infertility or sub-fertility, whether in humans or livestock species, remains a major concern for clinicians and technicians involved in reproduction. Indeed, they can cause tragedies in human relationships or have a dramatic overall negative impact on the sustainability of livestock breeding. Understanding and predicting semen fertility issues is therefore crucial and quality control procedures as well as biomarkers have been proposed to ensure sperm fertility. However, their predictive values appeared to be too limited and additional relevant biomarkers are still required to diagnose s

Therapeutic Potential of Non-Coding RNAs and TLR Signalling Pathways in Rheumatoid Arthritis.

BACKGROUND: Rheumatoid Arthritis (RA) is a common connective tissue disease, characteristic of chronic and invasive synovitis in single or multiple joints and vasculitis. RA is a heterogeneous disease with unclear pathogenesis. Therefore, exploring the etiology and pathogenesis of the disease is essential for identifying new promising treatment strategies for RA. Accumulated data have implicated the significant role of non-coding RNA in RA, some of which are demonstrated to regulate inflammation and autoimmunity in RA through the Toll-Like Receptor (TLR) signaling pathway. To clarify the mecha

The role of non-coding RNA on macrophage modification in tuberculosis infection.

Tuberculosis (TB), a serious disease caused by Mycobacterium tuberculosis (Mtb), remains the world's top infectious killer. It is well-established that TB can circumvent the host's immune response for long-term survival. Macrophages serve as the major host cells for TB growth and persistence and their altered functions are critical for the response of the host defense against TB exposure (elimination, latency, reactivation, and bacillary dissemination). Noncoding RNAs are crucial posttranscriptional regulators of macrophage discrimination. Therefore, this review highlights the regulatory mecha

Role of microRNA-7 in liver diseases: a comprehensive review of the mechanisms and therapeutic applications.

MicroRNA-7 (miR-7) is a small non-coding RNA, which plays critical roles in regulating gene expression of multiple key cellular processes. MiR-7 exhibits a tissue-specific pattern of expression, with abundant levels found in the brain, spleen, and pancreas. Although it is expressed at lower levels in other tissues, including the liver, miR-7 is involved in both the development of organs and biological functions of cells. In this review, we focus on the mechanisms by which miR-7 controls cell growth, proliferation, invasion, metastasis, metabolism, and inflammation. We also summarize the specif

Non-Coding Transcript Heterogeneity in Mesothelioma: Insights from Asbestos-Exposed Mice.

Mesothelioma is an aggressive, rapidly fatal cancer and a better understanding of its molecular heterogeneity may help with making more efficient therapeutic strategies. Non-coding RNAs represent a larger part of the transcriptome but their contribution to diseases is not fully understood yet. We used recently obtained RNA-seq data from asbestos-exposed mice and performed data mining of publicly available datasets in order to evaluate how non-coding RNA contribute to mesothelioma heterogeneity. Nine non-coding RNAs are specifically elevated in mesothelioma tumors and contribute to human mesoth

Arbovirus-vector protein interactomics identifies Loquacious as a co-factor for dengue virus replication in Aedes mosquitoes.

Efficient virus replication in Aedes vector mosquitoes is essential for the transmission of arboviral diseases such as dengue virus (DENV) in human populations. Like in vertebrates, virus-host protein-protein interactions are essential for viral replication and immune evasion in the mosquito vector. Here, 79 mosquito host proteins interacting with DENV non-structural proteins NS1 and NS5 were identified by label-free mass spectrometry, followed by a functional screening. We confirmed interactions with host factors previously observed in mammals, such as the oligosaccharyltransferase complex, a

RNA helicase DHX29 controls the translation of transcription factors involved in germinal center response and plasma cell differentiation in mice.

The roles of translational control in the immune system are incompletely understood. Using a CRISPR/Cas9-mediated functional screen of RNA helicases in an in vitro system of plasma cell differentiation, we identify in this study DHX29 as a critical regulator of this process. Mice with B-cell-specific deletion of Dhx29 exhibit severely impaired germinal center B-cell formation, plasma cell differentiation, and antibody production. Mechanistically, DHX29 promotes translation of Tcf3 and Tle3 mRNAs via binding to their 5' untranslated regions (UTRs). In the absence of DHX29, B cells exhibit norma

Meta-analysis of genetic regulation of RNA editing in the human brain identifies new genes underlying neurological disease.

Adenosine-to-inosine (A-to-I) RNA editing is among the most abundant post-transcriptional modifications, whose role in homeostasis and disease of the central nervous system (CNS) has been extensively reported. Regulation of A-to-I editing by cis-acting genetic variants (edQTLs) is understudied, especially in the brain. As a part of the BigBrain Project (n=4,471 individuals), which represents a >10-fold increase in sample size from previous edQTL studies, we harmonized A-to-I sites across datasets to perform a meta-analysis that identified 38,731 edQTLs in 3,554 genes. Integrating other QTL sum

The molecular mechanisms that underlie fragile X-associated premature ovarian insufficiency: is it RNA or protein based?

The FMR1 gene contains a polymorphic CGG trinucleotide sequence within its 5' untranslated region. More than 200 CGG repeats (termed a full mutation) underlie the severe neurodevelopmental condition fragile X syndrome, while repeat lengths that range between 55 and 200 (termed a premutation) result in the conditions fragile X-associated tremor/ataxia syndrome and fragile X-associated premature ovarian insufficiency (FXPOI). Premutations in FMR1 are the most common monogenic cause of premature ovarian insufficiency and are routinely tested for clinically; however, the mechanisms that contribute

Transcriptome-wide analysis reveals spatial correlation between N6-methyladenosine and binding sites of microRNAs and RNA-binding proteins.

N6-methyladenosine (m6A), the most prevalent epitranscriptomic modification in eukaryotes, is enriched in 3'-untranslated regions (3'UTRs) of mRNAs. As 3'UTRs are major binding sites of RNA-binding proteins (RBPs) and microRNAs (miRNAs), m6A-dependent local RNA structure change may alter the accessibility of RBPs and miRNAs to their target sites and regulate mRNA function. Using a human transcriptome-wide computational analysis to investigate the relation between m6A, RBPs and miRNAs, we find a strong positive correlation between number of m6A sites, miRNAs and RBPs binding to mRNAs, suggestin

In vivo therapeutic potential of Dicer-hunting siRNAs targeting infectious hepatitis C virus.

The development of RNA interference (RNAi)-based therapy faces two major obstacles: selecting small interfering RNA (siRNA) sequences with strong activity, and identifying a carrier that allows efficient delivery to target organs. Additionally, conservative region at nucleotide level must be targeted for RNAi in applying to virus because hepatitis C virus (HCV) could escape from therapeutic pressure with genome mutations. In vitro preparation of Dicer-generated siRNAs targeting a conserved, highly ordered HCV 5' untranslated region are capable of inducing strong RNAi activity. By dissecting th

MicroRNAs and their role in heart transplantation insights into rejection mechanisms: A narrative review.

Heart transplantation is the definitive treatment for patients with advanced heart failure and refractory symptoms. However, allograft rejection-both acute and chronic-remains a major cause of morbidity, leading to graft dysfunction and failure. Traditionally, endomyocardial biopsy (EMB) has been the standard method for screening allograft rejection. MicroRNAs (miRNAs) are small, non-coding RNA sequences that regulate gene expression by binding to the 3' untranslated regions of complementary mRNA transcripts. This review explores the potential of miRNAs as biomarkers for detecting allograft re

Rli51 Attenuates Transcription of the Listeria Pathogenicity Island 1 Gene mpl and Functions as a Trans-Acting sRNA in Intracellular Bacteria.

Listeria pathogenicity island 1 (LIPI-1) is a genetic region containing a cluster of genes essential for virulence of the bacterial pathogen Listeria monocytogenes. Main virulence factors in LIPI-1 include long 5' untranslated regions (5'UTRs), among which is Rli51, a small RNA (sRNA) in the 5'UTR of the Zn-metalloprotease-coding mpl. So far, Rli51 function and molecular mechanisms have remained obscure. Here, we show that Rli51 exhibits a dual mechanism of regulation, functioning as a cis- and as a trans-acting sRNA. Under nutrient-rich conditions, rli51-mpl transcription is prematurely termi

Effect of different immunisation schedules on the excretion and reversion of oral poliovaccine strains.

Excretion of live oral poliovaccine and molecular markers of increased virulence in the viruses isolated were examined in children who were either previously immunised with inactivated or live vaccine or were unimmunised. There appeared to be some effect of previous immunisation with either live or killed vaccine at the second dose of live vaccine. Where an effect was seen it took the form of reduced rates of excretion, shorter time periods of excretion and more rapid and complete reversion of the excreted virus. The data are consistent with the view that poliovirus is able to escape the immun

Emerging role of interaction between m6A and main ncRNAs in pancreatic cancer.

Pancreatic cancer (PC) is a highly lethal malignancy characterized by its rapid progression and poor prognosis. Despite advancements in understanding its pathogenesis, the intricate interplay of various molecular mechanisms remains to be fully elucidated. In recent years, N6-methyladenosine (m6A) modification has emerged as a significant RNA chemical modification, garnering considerable attention for its role in regulating RNA metabolism and gene expression. Concurrently, non-coding RNAs (ncRNAs), particularly microRNAs (miRNAs), long non-coding RNAs (lncRNAs) and circular RNAs (circRNAs) have

Leishmania exosomal tRNA-derived small non-coding RNA fragments modulate host THP-1 derived macrophage proteins: A quantitative proteomic analysis.

Protozoan parasites of the genus Leishmania, the causative agents of human leishmaniasis, have evolved mechanisms to manipulate host cell functions to their advantage. Recently, small non-coding RNAs have been identified as contributors to the pathogenesis of Leishmania infection. An increasing number of studies have demonstrated that Leishmania tRNA-derived small non-coding RNAs, also known as Leishmania tRNA-derived fragments (Ld-tRFs), can be delivered to host cells via exosomes, thereby influencing host cell function. Here, we investigated the potential effects of Leishmania exosomal Ld-tR
