Understanding cachexia as a cancer metabolism syndrome.

Metabolic reprogramming occurs in tumors to foster cancer cell proliferation, survival and metastasis, but as well at a systemic level affecting the whole organism, eventually leading to cancer cachexia. Indeed, as cancer cells rely on external sources of nitrogen and carbon skeleton to grow, systemic metabolic deregulation promoting tissue wasting and metabolites mobilization ultimately supports tumor growth. Cachectic patients experience a wide range of symptoms affecting several organ functions such as muscle, liver, brain, immune system and heart, collectively decreasing patients' quality

Physical exercise prepartum to support metabolic adaptation in the transition period of dairy cattle: A proof of concept.

In dairy cattle, the hormonal changes around calving induce large metabolic changes to support milk production. Mobilization of adipose reserves is one of the changes involved, imposing a metabolic load on the liver. We hypothesized that the risk for excessive lipolysis and hepatic lipidosis postpartum can be reduced by starting fat mobilization and processing during the prepartum period by physical exercise, especially in cows with a high body condition score (BCS). As a proof of concept, 32 pregnant Holstein-Friesian dairy cows were selected for a 2 × 2 experimental design. Sixteen cows had

Visualizing Super-Diffusion, Oligomerization, and Fibrillation of Amyloid-β Peptide Chains along Tubular Membranes.

A deeper mechanistic study of peptide amyloidosis on lipid membranes with varying shapes could enhance the comprehensive understanding of the contribution of cellular structures to multiple neurodegenerative diseases, including Alzheimer's disease. We report here the direct visual observation of amyloid-β peptide (Aβ) superdiffusing along tubular lipid membranes via single-molecule tracking (SMT). Such mobility on tubular membranes is critical, as it allows Aβ chains to oligomerize and elongate into fibrils. Factors such as cholesterol that favor Aβ chains with sufficient surface residence tim

Improved solvent extraction procedure and high-performance liquid chromatography-evaporative light-scattering detector method for analysis of polar lipids from dairy materials.

A normal-phase high-performance liquid chromatography-evaporative light-scattering detector method employing dichloromethane, methanol, and acetic acid/triethylamine buffer as the mobile phase was developed for analysis of polar lipids (PLs). This method was applicable for analysis of PLs from both dairy materials and soy lecithin. All of the PLs of interest such as glycolipids, phospholipids, and sphingomyelin were well separated with a total run time of 22.5 min and without necessitating the removal of neutral lipids beforehand. Peak retention times were stable, and the method was reproducib

Fabry disease: impaired autonomic function.

Previous reports of extensive lipid accumulation within neurons of the autonomic nervous system in Fabry disease suggest an anatomicopathologic basis for the peculiar pain, diminished sweating, and gastrointestinal symptoms experienced in this disorder. To further assess autonomic function in Fabry disease, noninvasive clinical tests were performed on 10 patients. Diminished sweating was found in each; the loss was approximately uniform proximally and distally, suggesting sweat gland dysfunction rather than autonomic neuropathy. Impaired pupillary constriction with pilocarpine, and reduced sal

Apolipoprotein-mimetic nanodiscs reduce lipid accumulation and improve liver function in acid sphingomyelinase deficiency.

Acid sphingomyelinase deficiency (ASMD) is a severe lipid storage disorder caused by the diminished activity of the acid sphingomyelinase enzyme. ASMD is characterized by the accumulation of sphingomyelin in late endosomes and lysosomes leading to progressive neurological dysfunction and hepatosplenomegaly. Our objective was to investigate the utility of synthetic apolipoprotein A-I (ApoA-I) mimetics designed to act as lipid scavengers for the treatment of ASMD. We determined the lead peptide, 22A, could reduce sphingomyelin accumulation in ASMD patient skin fibroblasts in a dose dependent man

Lipolysis in lipid turnover, cancer cachexia, and obesity-induced insulin resistance.

Triglycerides in adipose tissue are rapidly mobilized during times of energy needs via lipolysis, a catabolic process that plays important role in whole body triglyceride turnover. Lipolysis is regulated through cell surface receptors via neurotransmitters, hormones, and paracrine factors that activate various intracellular pathways. These pathways converge on the lipid droplet, the site of action of lipases and cofactors. Fat cell lipolysis is also involved in the pathogenesis of metabolic disorders, and recent human studies have underscored its role in disease states such as cancer cachexia

Real-time monitoring of melittin-induced pore and tubule formation from supported lipid bilayers and its physiological relevance.

The temporal evolution of effects of antimicrobial peptide melittin on supported phospholipid bilayers (SPBs) containing negatively charged phospholipids was monitored by ellipsometry and laser scanning microscopy together with measurements of lipid mobility by Z-scan fluorescence correlation spectroscopy. Under all conditions used in our study, we observed reproducibly two effects. The first one is formation of pores in the SPB, which occupy approximately 40% of the bilayer. The formation of pores was accompanied by a decrease in lateral diffusion coefficient of the lipids to approximately 60

The acyl-CoA synthetase TgACS1 allows neutral lipid metabolism and extracellular motility in Toxoplasma gondii through relocation via its peroxisomal targeting sequence (PTS) under low nutrient conditions.

Apicomplexa parasites cause major diseases such as toxoplasmosis and malaria that have major health and economic burdens. These unicellular pathogens are obligate intracellular parasites that heavily depend on lipid metabolism for the survival within their hosts. Their lipid synthesis relies on an essential combination of fatty acids (FAs) obtained from both de novo synthesis and scavenging from the host. The constant flux of scavenged FA needs to be channeled toward parasite lipid storage, and these FA storages are timely mobilized during parasite division. In eukaryotes, the utilization of F

A rapid and quantitative reversed-phase HPLC-DAD/ELSD method for lipids involved in nanoparticle formulations.

Lipid nanoparticles (LNPs) have shown great success as drug delivery systems, especially for mRNA vaccines, as those developed during the Covid-19 pandemics. Lipid analysis is critical to monitor the formulation process and control the quality of LNPs. The present study is focused on the development and validation of a high-performance liquid chromatography - diode array detector -evaporative light scattering detector (HPLC-DAD/ELSD) based method for the simultaneous quantification of 7 lipids, illustrating the main components of LNPs: ionizable lipids, the neutral co-lipid cholesterol, phosph

Mechanism of depletion of liver glycogen in cancer cachexia.

Mice transplanted with a cachexia-inducing colonic adenocarcinoma (MAC16) show a progressive decrease in liver glycogen in direct proportion to the loss of body weight. Such tumours elaborate a lipid mobilizing factor (LMF), which produces a dose-dependent stimulation, not only of adipocyte adenylate cyclase, but also of hepatocyte adenylate cyclase in a GTP-dependent manner. These results suggest that LMF has the capacity to initiate hepatic glycogenolysis through an increase in cyclic AMP.

Influence of nitric oxide donors on the intrinsic fluorescence of Na+,K+-ATPase and effects on the membrane lipids.

Effects of the nitric oxide donors S-nitroso-glutathione (GSNO) and S-nitroso-N-acetylpenicillamine (SNAP) on Na+,K+-ATPase-rich membrane fragments purified from pig kidney outer medulla were studied using intrinsic fluorescence and ESR of spin-labeled membranes. These S-nitrosothiols differently affected the intrinsic fluorescence of Na+,K+-ATPase: GSNO induced a partial quenching, whereas SNAP produced no alteration. Quenching can be due to a direct modification of exposed tryptophan residues or to an indirect effect caused by reactions of nitrogen oxide reactive species with other residues

Evaluating Cobas® b101 for lipid profile and SCORE2 risk assessment from capillary blood in population screening.

OBJECTIVES: Cardiovascular disease (CVD) is a leading cause of mortality worldwide. Emerging lung cancer screening in smokers may offer an opportunity for simultaneous cardiovascular risk evaluation. We developed a mobile CT screening unit and assessed the feasibility of integrating dyslipidemia screening and non-HDL cholesterol-based SCORE2 estimation using the Cobas® b101 point-of-care testing (POCT) analyzer on capillary blood samples. METHODS: Following a preliminary evaluation of two POCT analyzers, the Cobas® b101 was selected for capillary lipid profiling. Lipid results obtained from c

The effect of cholesterol on the intrinsic rate of lipid flip-flop as measured by sum-frequency vibrational spectroscopy.

Cholesterol is a major constituent of biological membranes in mammalian cells. Experiments have shown that cholesterol influences the physical properties of the plasma membrane, such as lateral diffusion and phase equilibrium. In addition to controlling the 2-dimensional phase behaviour and mobility of lipids in membranes, cholesterol has also been implicated in the transbilayer diffusion of lipids across the bilayer. Sum-frequency vibrational spectroscopy (SFVS) is used to measure the intrinsic rate of lipid flip-flop for 1,2-distearoyl-sn-glycero-3-phosphocholine (DSPC) in the presence of ch

Induced videokeratography alterations in patients with excessive meibomian secretions.

PURPOSE: To describe lipid-induced specific videokeratographic (VKG) corneal changes and subsequent resolution after eyelid washing. METHODS: VKG was performed with C-Scan corneal topography. In all patients an excessive meibomian gland lipid secretion was found with or without coexistent chronic posterior blepharitis. After the initial VKG, a meticulous cleaning of the lids with a mild alkali shampoo (10% Johnson's baby shampoo in sterile water) was done, first by gently scrubbing the closed eyelid fissure with the solution to mobilize and emulsify any Meibomian gland secretions followed by

Revealing a Dual Role of Ganglioside Lipids in the Aggregation of Membrane-Associated Islet Amyloid Polypeptide.

Amyloid formation of the human islet amyloid polypeptide (hIAPP) correlates with a loss of insulin-producing beta cells in patients with type II diabetes mellitus. In this study, we investigated the binding of hIAPP to bilayers consisting of ganglioside lipids and dioleoylphosphatidylcholine (DOPC), which is a physiologically relevant lipid species for pancreatic beta cell-associated aggregation. The membrane interactions are studied computationally using a combination of coarse-grained, umbrella sampling, and atomistic molecular dynamics simulations. Herein, we demonstrate how the hIAPP pepti

Characterization of Triacylglycerol Dynamics and Lipid Metabolism in the Beetle Tribolium castaneum.

Lipids are essential for all organisms, and insects can obtain these macromolecules from their diet or synthesize them from carbohydrates and amino acids. These animals store fatty acids in the form of triacylglycerol in lipid droplets in the fat body and, in times of demand, mobilize these reserves as an energy source or for synthesizing structural lipids. However, details about this metabolism are unknown in most species, such as the beetle Tribolium castaneum. Here, we explored the dynamics of triacylglycerol and lipid metabolism during larval and pupal development, after adult emergence, a

Modes of interaction between tapinarof and model lipid membranes mimicking stratum corneum lipid composition and related transdermal transport in human skin.

Tapinarof (3,5-dihydroxy-4-isopropylstilbene, TPN), structurally related to resveratrol, is a nonsteroidal agent that belongs to the class of naturally derived aryl hydrocarbon receptor (AhR) agonists. TPN is an active ingredient of a topical medication (1% cream, VTAMA®). The log Poctanol|water of 2.79 ± 0.08 confirms the relative lipophilic character of TPN. The confirmed hydrophobicity of TPN raises the question of how this drug interacts with model lipidic structures and cell membranes, providing information about its transport mechanisms in the skin. Based on a liposomal model (40 % (w/w)
