Occipital bone hypoplasia with foramen magnum obstruction and secondary syringomyelia (SM) is a common condition in the Cavalier King Charles Spaniel (CKCS) that is similar to human Chiari type I malformation. A worldwide family tree of more than 5,500 CKCSs spanning a maximum of 24 generations was established by obtaining pedigree information from 120 dogs diagnosed with SM secondary to occipital bone hypoplasia. The ongoing study showed 6 of 8 great grandparents of all affected dogs could be traced back to 2 female ancestors so that all 8 were descended from one or the other or both. The dis

Identifying molecular connections between developmental processes and disease can lead to new hypotheses about health risks at all stages of life. Here we introduce a new approach to identifying significant connections between gene sets and disease genes, and apply it to several gene sets related to human development. To overcome the limits of incomplete and imperfect information linking genes to disease, we pool genes within disease subtrees in the MeSH taxonomy, and we demonstrate that such pooling improves the power and accuracy of our approach. Significance is assessed through permutation.

INTRODUCTION: Metaphyseal dysplasia (Pyle disease) is a rare autosomal recessive disease with impressive and characteristic radiological findings but relatively mild clinical features. It is usually incidentally diagnosed, despite the impressive radiological findings of gross metaphyseal widening and thinning of cortical bone.
CASE REPORT: Herein, we report an exceptionally unusual case of metaphyseal dysplasia in association with chronic facial nerve palsy.
DISCUSSION: Chronic facial nerve palsy due to compression of the facial nerve in a patient with Pyle disease represents an unusual nove

There is a wide variety of hereditary and non-hereditary bone dysplasias, many with unique radiographic findings. Hereditary bony dysplasias include osteopoikilosis, osteopathia striata, osteopetrosis, progressive diaphyseal dysplasia, hereditary multiple diaphyseal sclerosis and pyknodysostosis. Non-hereditary dysplasias include melorheostosis, intramedullary osteosclerosis and overlap syndromes. Although many of these dysplasias are uncommon, radiologists should be familiar with their genetic, clinical and imaging findings to allow for differentiation from acquired causes of bony sclerosis.

Fetal skeletal dysplasias are a heterogeneous group of rare genetic disorders, affecting approximately 2.4-4.5 of 10,000 births. We performed a retrospective review of the perinatal autopsies conducted between the years 2002-2011 at our center. The study population consisted of fetuses diagnosed with skeletal dysplasia with subsequent termination, stillbirth and live-born who died shortly after birth. Of the 2002 autopsies performed, 112 (5.6%) were diagnosed with skeletal dysplasia. These 112 cases encompassed 17 of 40 groups of Nosology 2010. The two most common Nosology groups were osteogen

Incomplete ossification of the humeral condyle (IOHC) is characterized by an intracondylar fissure located where the intercondylar physis is present in growing dogs. Its radiologic and computed tomographic features have been described but the magnetic resonance (MR) features have not been characterized. Our purpose was to further describe the range of MRappearances of IOHC, to assess the diagnostic capability of MRrelative to radiology, and to determine whether MRis able to identify the disease before a fissure forms. Thirty-eight elbow MRscans and radiographs, when available, were reviewed an

Congenital disorders of glycosylation (CDGs) affect multiple systems and present a broad spectrum of clinical features, often including skeletal dysplasia. Exome sequencing has led to the identification of new CDG genes. Immune and skeletal phenotypes associated with mutations in PGM3, encoding a protein that converts N-acetyl-glucosamine-6-phosphate into N-acetyl-glucosamine-1-phosphate, were recently reported. Through exome sequencing, we identified a novel homozygous mutation (c.1135T>C; p.Phe379Leu) in PGM3 in two siblings with bone marrow failure, severe combined immunodeficiency, renal a

Mondini dysplasia is a rare but an important cause for recurrent pyogenic meningitis in children and requires a high index of clinical suspicion for early diagnosis. We present the case of a 7-year-old boy, who presented with 2 episodes of pyogenic meningitis within a span of 1 month. There was no obvious history of hearing abnormalities, but pure tone audiometry suggested profound mixed hearing loss in the left ear. High-resolution computed tomographic scan and magnetic resonance imaging of temporal bones confirmed the diagnosis of Mondini dysplasia in the left ear. Computed tomographic ciste

In reviewing 38 cases of biliary atresia, 8 cases were detected to have osseous abnormalities. We found definite generalized demineralization of bones in 3 cases, metaphyseal abnormalities in 5 cases, and fractures in 4 cases. Marked demineralization of bones was seen in cases in which the clinical course was more than 1 year. No good correlation was observed between osseous changes and severity of liver dysfunction.

Macrophages are important immune cells for cardiac development, homeostasis, and the response to pathological injury and inflammation. Single-cell sequencing and fate-mapping technologies have revealed the heterogeneity and development of cardiac macrophages. They are classed into two main groups: embryonic tissue-resident and bone marrow-derived macrophages. These different macrophage subsets show complex functions in cardiac diseases such as inflammation regulation, tissue repair, metabolic reprogramming and intercellular communication within the cardiac microenvironment. This review discuss

Disturbed innervation of the pelvic muscular diaphragm from lumbosacral myelodysplasia and other osseous dysplasias results in flaccid paralysis of the perineum. This permits excessive downward herniation, most noticeable in neonates, of the lower abdominal and pelvic organs. The radiographic changes occurring with organ descensus may be transitory and diverse, and recognition of these various findings is important to avoid misdiagnosis and unnecessary further investigations. Several examples of the radiographic changes are presented.

BACKGROUND: Vitamin D is important for bone mass accrual during growth. Additionally, it is considered a requirement for a multitude of processes associated with, for example, the development of immunity. Many countries apply vitamin D supplementation strategies in infants, but the guidelines are not based on scientific evidence and aim at prevention of rickets. It remains unclear whether the recommended doses are sufficient for the wide array of other effects of vitamin D. The VIDI trial performed in Finland is the first large randomised controlled study for evaluation of the effects of diffe

The intrauterine-growth-restricted (IUGR) state, particularly the asymmetric one, has been associated with 'Developmental Origins of Health and Disease' (DOHaD) consequences later in life. Several environmental factors, acting during the phase of foetal developmental plasticity interact with genotypic variation, 'programme' tissue function and change the capacity of the organism to cope with its environment. They may be responsible for chronic illness risk in adulthood. Detection of possible future DOHaD consequences at a very early age, by applying relevant biomarkers, is of utmost importance
