Shedding the Light on Post-Vaccine Myocarditis and Pericarditis in COVID-19 and Non-COVID-19 Vaccine Recipients.

Myocarditis and pericarditis have been linked recently to COVID-19 vaccines without exploring the underlying mechanisms, or compared to cardiac adverse events post-non-COVID-19 vaccines. We introduce an informatics approach to study post-vaccine adverse events on the systems biology level to aid the prioritization of effective preventive measures and mechanism-based pharmacotherapy by integrating the analysis of adverse event reports from the Vaccine Adverse Event Reporting System (VAERS) with systems biology methods. Our results indicated that post-vaccine myocarditis and pericarditis were as…

Lipid- and Polymer-Based Nanoscale Delivery of Anti-Aging Phytochemicals.

Recently, nano-scale delivery approaches have been a field of focus to improve the limitations regarding current traditional delivery systems of phytochemicals to treat age-related disorders. Nanotechnology can help to overcome both the large and small-scale barriers associated with traditional drug and vitamin delivery techniques, such as poor biodistribution, instability, and intracellular trafficking, through cell-targeting and other methods. The main forms of nano-drug delivery systems can be classified under lipid-based nanocarrier systems, polymer-based nanocarrier systems, and inorganic

Engineered platforms for melanogenesis research: Bridging synthetic biology, bioengineering, and biomimetics.

Melanogenesis, the biological process responsible for melanin synthesis, plays a critical role in determining skin pigmentation and providing photoprotection. Dysregulation of this pathway leads to a wide range of pigmentary disorders that affect a significant proportion of the global population. Traditional approaches to studying melanogenesis rely largely on cultured melanocytes and in vivo animal models; however, these systems present several limitations, including concerns related to physiological relevance, ethical constraints, high maintenance costs, and limited scalability. In recent ye

Preclinical models of meningioma: Cell culture and animal systems.

Meningioma is the most common primary intracranial tumor; yet there are no effective systemic or molecular therapies for meningioma patients. One of the primary barriers to understanding meningioma biology and identifying novel therapeutic targets is the lack of tractable preclinical models. While numerous model systems have been created for meningioma, many have fundamental drawbacks. This chapter details the strengths and limitations of existing meningioma models and suggests possible future model systems. Cell culture meningioma models consist of human meningioma cell lines derived from tum

A Tricky Trait: Applying the Fruits of the "Function Debate" in the Philosophy of Biology to the "Venom Debate" in the Science of Toxinology.

The "function debate" in the philosophy of biology and the "venom debate" in the science of toxinology are conceptually related. Venom systems are complex multifunctional traits that have evolved independently numerous times throughout the animal kingdom. No single concept of function, amongst those popularly defended, appears adequate to describe these systems in all their evolutionary contexts and extant variations. As such, a pluralistic view of function, previously defended by some philosophers of biology, is most appropriate. Venom systems, like many other functional traits, exist in natu

Autopoiesis and organ intelligence: toward a cognitive paradigm in medicine.

Modern medicine has been dominated by a neurocentric paradigm that locates cognition exclusively in the brain and treats organs as passive executors of function. Emerging evidence from biology and medicine challenges this view, suggesting that living systems are autopoietic and that organs such as the heart and gut, and the immune system exhibit forms of intrinsic intelligence. This article explores the conceptual foundations and clinical implications of cognitive biology, with particular emphasis on autopoiesis and distributed organ cognition, as a framework for rethinking health, disease, an

Non-linearity, complexity, and quantization concepts in biology.

Founders of quantum mechanics (QM) anticipated that revisions to classical physics due to strange elements of quantum reality, would necessitate similar changes in biology. Complexity theory, systems biology and quantum biology provide possible solutions indicating that subject/object separation is a useful fiction for reductive science. Direct correlates to such QM observational/measurement issues as Complementarity and Uncertainty may justify the introduction of an analog of Heisenberg's uncertainty and the Planck constant for living systems. The phase space of "adjacent possibles" for biolo…

Thiol/disulfide redox states in signaling and sensing.

Rapid advances in redox systems biology are creating new opportunities to understand complexities of human disease and contributions of environmental exposures. New understanding of thiol-disulfide systems have occurred during the past decade as a consequence of the discoveries that thiol and disulfide systems are maintained in kinetically controlled steady states displaced from thermodynamic equilibrium, that a widely distributed family of NADPH oxidases produces oxidants that function in cell signaling and that a family of peroxiredoxins utilize thioredoxin as a reductant to complement the w…

Development and Application of MLB Human Astrovirus Reverse Genetics Clones and Replicons.

Human astroviruses pose a significant public health threat, especially to children, the elderly, and immunocompromised individuals. Nevertheless, these viruses remain largely understudied, with no approved antivirals or vaccines. This protocol focuses on leveraging reverse genetics (RG) and replicon systems to unravel the biology of MLB genotypes, a key group of neurotropic astroviruses. Using reverse genetics and replicon systems, we identified critical genetic deletions linked to viral attenuation and neurotropism, pushing forward vaccine development. We also uncovered novel replication mech

Cyanobacterial type I CRISPR-Cas systems: distribution, mechanisms, and genome editing applications.

Cyanobacteria, renowned for their photosynthetic capabilities, serve as efficient microbial chassis capable of converting carbon dioxide into a spectrum of bio-chemicals. However, conventional genetic manipulation strategies have proven incompatible with the precise and systematic modifications required in the field of cyanobacterial synthetic biology. Here, we present an in-depth analysis of endogenous CRISPR-Cas systems within cyanobacterial genomes, with a particular focus on the Type I systems, which are the most widely distributed. We provide a comprehensive summary of the reported DNA de

Klinefelter syndrome comorbidities linked to increased X chromosome gene dosage and altered protein interactome activity.

Klinefelter syndrome (KS) (47,XXY) is the most common male sex chromosome aneuploidy. Diagnosis and clinical supervision remain a challenge due to varying phenotypic presentation and insufficient characterization of the syndrome. Here we combine health data-driven epidemiology and molecular level systems biology to improve the understanding of KS and the molecular interplay influencing its comorbidities. In total, 78 overrepresented KS comorbidities were identified using in- and out-patient registry data from the entire Danish population covering 6.8 million individuals. The comorbidities extr…
