[Assessment of spontaneous motor activity in young infants: an effective method for the detection of brain function disorders].

A method for the assessment of the brain function of young infants was recently introduced. It consists of evaluation of the quality of spontaneously generated generalized movements (general movements, GMs). GMs appear at an early stage of pregnancy and persist until approximately the 4th month after term. Normal GMs are characterized by the triad of complexity, variation and fluency. Mildly abnormal GMs. indicating mild dysfunction of the nervous system, are not fluent but jerky or stiff. Markedly abnormal GMs, indicating major nervous system dysfunction, are characterized mostly by absence o

Cerebellar outflow lesions: a comparison of movement deficits resulting from lesions at the levels of the cerebellum and thalamus.

Previous work has shown that lesions in the lateral cerebellum involving the dentate nucleus impair both reaching and pinching movements in humans and monkeys. This study addressed the question of whether disruption of the cerebellar-thalamo-cortical pathway at the level of the thalamus would produce behavioral deficits similar to those seen after dentate damage. We compared the performance of both reaching and pinching movements in patients with lateral cerebellar lesions and in patients with discrete lesions of the ventrolateral thalamus. The patients with thalamic lesions had minimal or no

The Role of Polysomnography for Children with Attention-Deficit/Hyperactivity Disorder.

Objective: Attention-Deficit/Hyperactivity Disorder (ADHD) is a common neurodevelopmental disorder in children, characterized by inattention, hyperactivity, and impulsive behavior. In recent years, studies have shown that patients with ADHD often experience sleep problems, raising clinical interest in the potential role of polysomnography (PSG) in the diagnosis and management of ADHD. This study examines polysomnographic findings in children with ADHD who present with diverse sleep complaints. Methods: A cohort of children aged younger than 18 years, diagnosed with ADHD based on DSM-5 criteria

Reducing PTSD symptoms through unconscious intervention.

Intrusive traumatic memories are a hallmark symptom of posttraumatic stress disorder (PTSD). Conventional therapies typically require explicit and emotionally distressing reexposure to trauma cues, often leading to high dropout rates among severely affected patients. This study examined whether trauma memory intrusions could be effectively reduced through unconscious intervention, minimizing emotional distress by subliminally presenting trauma-related visual cues during bilateral eye movement intervention, a core component of eye movement desensitization and reprocessing. Four experiments test

Dystonia and Tremor.

Dystonia and tremor are common movement disorders with heterogenous presentations. Their overlap, termed "tremor in dystonia" for this review, is a common phenomenon that is classified inconsistently in practice and the literature. While the two-axis structure of the dystonia and tremor classifications clarified some aspects of tremor in dystonia, confusion and contention remain amongst clinicians. This is due not only to the varied definitions proposed over time and clinical heterogeneity, but the current lack of consensus on the underlying pathophysiology. This paper outlines the existing an…

A novel heterozygous variant of FUS gene associated with juvenile ALS and premature tremor onset: a case report.

Juvenile amyotrophic lateral sclerosis (JALS) is neurodegenerative disease of the upper and lower motor neurons of rare incidence. Although fused in sarcoma (FUS) mutations in JALS patients have been associated with movement disorders, here we described the case of a young girl with very early onset of tremor, years before commencement of weakness; once symptoms of JALS were stablished, a typical rapid disease progression from spinal to bulbar symptoms were noticed. Genetic testing revealed a novel mutation in FUS gene causative of a protein dysfunction. This case emphasizes the fact that some…

A Case Report of Abdominal Myoclonus Following Anterior Cerebral Artery Stroke.

BACKGROUND: Post-stroke movement disorders are rare. Abdominal myoclonus following anterior cerebral artery (ACA) infarction has not been previously reported. CASE REPORT: A 75-year-old man presented with acute right-sided weakness and involuntary, rhythmic contractions of the abdominal muscles. Imaging revealed an acute left ACA infarction involving the precentral gyrus. Scalp electroencephalography (EEG) showed no epileptiform discharges. Other secondary causes of myoclonus were ruled out. Abdominal myoclonus resolved after intravenous levetiracetam. DISCUSSION: This case highlights cortic…

Woodhouse-Sakati syndrome: A review.

Neurodegeneration with brain iron accumulation (NBIA) is a rare and inherited spectrum of movement disorders caused by mutations affecting the function of proteins that participate in the homeostasis of tissue metals such as iron or copper and other metabolic pathways, although the precise function of the proteins encoded are not always known. Woodhouse-Sakati Syndrome (WSS) is one of the rarest NBIAs. Patients with WSS are characterized by endocrinological and neurological manifestations and neuroradiological findings. However, diagnostic criteria have not been published yet. This article rev…

SARAspeech-Feasibility of automated assessment of ataxic speech disturbance.

Ataxias are a group of movement disorders that are characterized by progressive loss of balance, impaired coordination and speech disturbance, which together lead to markedly reduced quality of life. Speech disturbance is clinically diagnosed, but methods for objective assessment of severity are lacking. Using 71 sets of speech recordings from ataxia patients, we developed an automated classification system. With a tolerance of ±1 point, this classification system correctly predicted experts' ratings of speech disturbance according to item 4 of the Scale for Assessment and rating of ataxia (SA…

Dystonia, spastic tetraplegia, and ataxia due to a novel mutation in the dynamin domain of OPA1.

Movement disorders manifest in various hereditary neurodegenerative diseases. We reported a young man who presented with progressive upper limb dystonia, spastic tetraplegia, and ataxia. Whole-exome sequencing (WES) revealed a novel variant, c.2357A > G, in the dynamin domain of OPA1. No mtDNA deletion was detected in muscle by long-range PCR. Atrophy and decreased glucose metabolism of the basal ganglia were discovered. Decreased mtDNA copy number, fragmented mitochondria, slightly impaired oxidative phosphorylation, and increased autophagy were detected in mutant fibroblasts. Evident oxidati…

Brain 18F-FDG PET in Cowden Syndrome.

Cowden disease is associated with neurodevelopmental abnormalities such as macrocephaly, autism spectrum disorder, and developmental delay. Our understanding of neuroimaging anomalies in patients with PTEN mutations is limited to anatomical MRI abnormalities including white matter abnormalities, meningiomas, arteriovenous malformations, and cortical dysplasia. Our current communication extends the neurological Cowden syndrome phenotype by using brain 18F-FDG PET/CT imaging as a useful complementary approach to MRI to explore movement disorders and neuropsychiatric syndromes in a patient with C…

Generation and characterization of three human induced pluripotent stem cell lines (iPSC) from two family members with dilated cardiomyopathy and left ventricular noncompaction (DCM-LVNC) and one healthy heterozygote sibling.

Autophagy serves as a master regulator of cellular homeostasis. Hence, expectedly autophagic dysfunction has been documented in many diseases such as cancer, neurodegeneration and cardiovascular disorders. A novel homozygous mutation in PLEKHM2 gene (mPLEKHM2) resulted in dilated cardiomyopathy with left ventricular noncompaction (DCM-LVNC), probably as result of impaired autophagy due to disruption of lysosomal movement assisted by PLEKHM2. Here we report a generation of three iPSC lines, four clones originated from two patients with homozygous mPLEKHM2 and two from a heterozygote sibling. Al
