Proteomic Alterations and Antimicrobial Effects of Postbiotics from Probiotic Lactic Acid Bacteria and Yeast Against Multidrug-Resistant Salmonella Typhimurium.

The study conducted presents the antimicrobial and antibiofilm properties of postbiotics obtained from Bifidobacterium bifidum, Lactiplantibacillus plantarum, and Saccharomyces cerevisiae against multidrug-resistant Salmonella enterica serotype Typhimurium. Integrated microbiological and proteomic approaches proved that all postbiotics inhibited bacterial growth as well as biofilm formation with high biocompatibility to mammalian cells. Transcriptional and proteomic analyses indicated that exposure to postbiotics modulated major virulence-associated genes and proteins related to motility, adhe

Therapeutic efficacy of phage vB_SalP_NW15 and cinnamaldehyde against drug-resistant Salmonella Enteritidis in a chicken infection model.

Salmonella is a major Gram-negative pathogen causing infections in human and poultry. Currently, clinical treatment primarily relies on antibiotics, but their overuse has led to the widespread prevalence of multidrug-resistant (MDR) strains. The combination of phages and natural bioactive substances offers a novel approach to combat MDR Salmonella. In this study, a lytic Salmonella phage vB_SalP_NW15 was isolated from the wastewater of a livestock farm and identified as a member of the Siphoviridae family through transmission electron microscopy. The phage demonstrated a broad-spectrum, effect

Antibiotic concentrations induce morphological changes and increase biofilm formation in multi-antibiotic and heavy metal resistant Kluyvera cryocrescens and Serratia fonticola.

Water pollution is the biggest challenge that has rendered existing water resources unusable due to contamination with antibiotics and heavy metals. Antibiotics are often used to treat bacterial diseases. Heavy metals, on the other hand, are micro-pollutants that pose a threat to aquatic systems, especially when they accumulate in nature. Increasing pollution and the uncontrolled use of antibiotics have exposed bacteria to non-lethal concentrations (sub-MIC), potentially leading to resistance. In this study, Kluyvera cryocrescens and Serratia fonticola were isolated from a freshwater source an

Structural insights into the advancements of mobile colistin resistance enzymes.

The plasmid-encoded mobile colistin resistance enzyme (MCR) is challenging the clinical efficacy of colistin as a last-resort antibiotic against multidrug-resistant bacteria. This transferase catalyzes the addition of positively charged phosphoethanolamine to lipid A, and its catalytic domain in the periplasm has been elucidated. To date, there are many works on the catalytic domain and function of this enzyme class. However, the roles of unreported soluble or inter-membrane domains remain undefined, which might cause an inaccurate or even incorrect understanding of substrate recognition and b

Investigation of the impact of widely used pesticides on conjugative transfer of multidrug resistance plasmids.

Plasmid-mediated conjugative transfer has emerged as a major driver accounting for the dissemination of antibiotic resistance genes (ARGs). In addition to the use of antimicrobial agents, there is growing evidence that non-antibiotic factors also play an important role. Pesticides are widely used to protect crops against vectors of diseases, and are indispensable agents in agricultural production, whereas the impact of pesticide pollution on the transmission of antimicrobial resistance remains poorly understood. Here we reveal that the pesticides at environmentally relevant concentrations, esp

Incremental increases in physiological fluid shear progressively alter pathogenic phenotypes and gene expression in multidrug resistant Salmonella.

The ability of bacteria to sense and respond to mechanical forces has important implications for pathogens during infection, as they experience wide fluid shear fluctuations in the host. However, little is known about how mechanical forces encountered in the infected host drive microbial pathogenesis. Herein, we combined mathematical modeling with hydrodynamic bacterial culture to profile transcriptomic and pathogenesis-related phenotypes of multidrug resistant S. Typhimurium (ST313 D23580) under different fluid shear conditions relevant to its transition from the intestinal tract to the blood

The protective role of commensal gut microbes and their metabolites against bacterial pathogens.

Multidrug-resistant microorganisms have become a major public health concern around the world. The gut microbiome is a gold mine for bioactive compounds that protect the human body from pathogens. We used a multi-omics approach that integrated whole-genome sequencing (WGS) of 74 commensal gut microbiome isolates with metabolome analysis to discover their metabolic interaction with Salmonella and other antibiotic-resistant pathogens. We evaluated differences in the functional potential of these selected isolates based on WGS annotation profiles. Furthermore, the top altered metabolites in co-cu

Personalized aerosolised bacteriophage treatment of a chronic lung infection due to multidrug-resistant Pseudomonas aeruginosa.

Bacteriophage therapy has been suggested as an alternative or complementary strategy for the treatment of multidrug resistant (MDR) bacterial infections. Here, we report the favourable clinical evolution of a 41-year-old male patient with a Kartagener syndrome complicated by a life-threatening chronic MDR Pseudomonas aeruginosa infection, who is treated successfully with iterative aerosolized phage treatments specifically directed against the patient's isolate. We follow the longitudinal evolution of both phage and bacterial loads during and after phage administration in respiratory samples. P

Extreme resistance to the novel siderophore-cephalosporin cefiderocol in an extensively drug-resistant Klebsiella pneumoniae strain causing fatal pneumonia with sepsis: genomic analysis and synergistic combinations for resistance reversal.

Cefiderocol (CFDC) is the first-in-class siderophore-cephalosporin. Klebsiella pneumoniae strain that is extremely resistant to CFDC (MIC: 256 µg/ml) was isolated for the first time in the United Arab Emirates from a patient with pneumonia and sepsis. It belonged to sequence-type 14 (ST14), with a novel core genome ST. Resistance was driven by the co-expression of β-lactamases (blaNDM-1, blaOXA-232 and blaCTX-M-15) and a mutation in catecholate-siderophore receptor, utilized by CFDC to enter the bacterial cell. Synergistic combinations (β-lactamase inhibitors, aztreonam plus CFDC) re-sensitize
