Antimicrobial use in breeding kennels and antimicrobial resistance profile of Escherichia coli and Staphylococcus pseudintermedius isolated from healthy breeding bitches in Northern Italy.

Antimicrobials can be overused in dog breeding facilities, leading to increased resistance among commensal bacteria. The aim of this study was to investigate antimicrobial use intensity in a professional canine reproduction setting by assessing the resistance profiles of Escherichia coli and Staphylococcus pseudintermedius isolated from healthy breeding bitches, as well as through a questionnaire for breeders. Five bitches from each of 15 kennels in Northern Italy were sampled from the perivulvar skin and rectum, and the susceptibility of the isolated bacteria to a panel of different antimicro

Rational design of amphiphilic pyrrolidinium derivatives: structure-activity correlation and alkyl chain tuning for superior antimicrobial activity.

A novel amphiphilic pyrrolidine derivatives, 1-alkyl-1-(2-(hexyl carbamoyloxy)ethyl)pyrrolidinium bromides (CPS-n-6, where n = 12, 14, 16, and 18), were synthesized and characterized. Comprehensive investigations demonstrated that the amphiphiles synthesized exhibit multifunctional properties and meet stringent biomedical criteria. CPS-n-6 possess membranotropic activity and are capable of forming mixed lipid/surfactant bilayers, imparting a positive surface charge and stabilizing biological membranes. Surfactant-based supramolecular systems exhibit the properties of micellar nanocontainers, d

Comparison of ex vivo placenta perfusion and in vitro BeWo b30 cell models for assessing transfer of developmental and reproductive toxic compounds.

There is an increasing interest in the prediction of exposure for risk assessment, as next generation risk assessment (NGRA) encompasses an exposure-led approach. Within the field of developmental and reproductive toxicology (DART), data on placental transfer is essential for estimating foetal exposure. However, human-relevant data is often limited or unavailable. Ex vivo human placenta perfusion is considered the gold standard in placental transfer research, but it is hampered by a low success rate, limited availability of tissue, and the inability to allow high throughput data collection. Utilizing whole genome sequencing data for machine learning driven prediction of antibiotic resistance in Escherichia coli.

BACKGROUND: Antimicrobial resistance (AMR) is an escalating global public health threat that substantially undermines the effectiveness of standard anti-infective therapies and increases the risk of adverse clinical outcomes. METHODS: We analyzed whole-genome sequencing (WGS) data from 1952 Escherichia coli isolates with AST phenotypes. Gene, single-nucleotide polymorphisms (SNPs), and k-mer features were extracted to train machine-learning classifiers for predicting resistance to 10 common antibiotics. Model performance was evaluated using the area under the receiver operating characteristic

Environmental occurrence, ecotoxicity, and management perspectives of β-lactam antibiotics.

β-Lactams are the most widely prescribed antibiotic class globally, comprising penicillins, cephalosporins, carbapenems, and monobactams. Their continual release to the environment raises concerns about both direct ecotoxicity and potential indirect effects on ecosystem function. This review highlights three key findings; 1) environmental concentrations can exceed predicted no-effect concentrations for resistance selection; for example ceftriaxone had been reported 6160 µg/L in wastewater influent and 4150 µg/L in effluent in India, indicating that many β-lactams have removal efficiency below

The ribosomal RNA synthesis ratio biomarker in Mycobacterium ulcerans for drug activity evaluation.

Buruli ulcer (BU), a neglected tropical disease caused by Mycobacterium ulcerans (Mul), is treated with an 8-week regimen of rifampicin (RIF) and clarithromycin (CLA). Clinical trials are currently evaluating amoxicillin/clavulanate (AMX/CLV) co-administration to reduce treatment duration. However, conventional methods for assessing in vitro drug efficacy against Mul, like colony-forming units (CFUs), are slow and cumbersome. The ribosomal RNA synthesis ratio (RS-ratio) measures ribosome biogenesis and serves as a proxy of metabolic activity. While it is a promising predictive biomarker for tr

Yeast's vacuole a privileged niche that protects intracellular bacteria against antibiotics.

Detection of Helicobacter pylori, Staphylococcus, Nocardia and Cyanobacteria inside the yeast Candida tropicalis raised the question whether treating yeast with antibiotics mix (ABM) eliminates intracellular bacteria. Live/Dead staining showed occurrence of viable bacteria inside the vacuole of C. tropicalis. Amplification of bacterial 16S rRNA genes from yeast DNA with the size of 521, 750, 606 and 450 bp were similar to those from control H. pylori, Staphylococcus, Nocardia and Cyanobacteria, respectively. To eliminate intracellular bacteria yeast cultures in yeast-glucose (YG) broth were tr
