BACKGROUND: Genome-wide association studies have implicated the human leukocyte antigen/major histocompatibility complex (HLA/MHC) region in amyotrophic lateral sclerosis (ALS) susceptibility, and immune dysregulation is increasingly recognised as a modifier of disease course. High-resolution HLA data for ALS remain confined to European and East Asian ancestries. We tested whether classical HLA class I and class II variation contributes to ALS susceptibility in a Kuwaiti cohort.
METHODS: We analysed 38 unrelated ALS cases (mean age, 57.4 years; 63.2% male) and 150 population-matched controls

The human leukocyte antigen (HLA) allele and haplotype frequencies were calculated from 263,542 voluntary donors with medium- to high-resolution typing. This is the first study of a large sample with high-resolution HLA typing in the Thai population. The total numbers of two-field single alleles for HLA-A, -B, -C, -DRB1, and -DQB1 were 139, 198, 115, 80, and 24, respectively. The proportion of each HLA allele in Thais ranged from 1.54% to 3.45% of the total two-field HLA alleles in the IPD-IMGT/HLA database. The frequencies of alleles following common, intermediate, and well-documented (CIWD)

BACKGROUND: Behcet's syndrome (BS) was first described in 1937 as a triad of oral aphthae, genital ulceration, and ocular involvement. It is now recognized as a multisystemic vasculitis that can affect central nervous, gastrointestinal, and cardiovascular systems. Despite extensive research, the etiology of BS remains unclear, and genetic, viral, and environmental factors are thought to contribute to its pathogenesis.
OBJECTIVE: This study aimed to investigate the role of interleukin-17 (IL-17) and IL-23 receptor (IL-23R) gene polymorphisms in the etiology of BS and to evaluate their associat

Systemic lupus erythematosus (SLE) is a complex autoimmune disease characterized by dysregulated B cell responses and pathogenic autoantibody production. Given the central role that B cells play in SLE pathogenesis, we aimed to characterize genetic variation within the B cell compartment of African ancestry (AFR) women with SLE who are disproportionately and more severely affected by SLE. Here, we analyzed RNA-seq data from a previously published study of high disease activity AFR SLE individuals and healthy controls (HC) to identify coding variants in B cells with potential functional consequ

Epidemiological and biological evidence indicate a close connection between Alzheimer's disease (AD) and type-2 diabetes mellitus. Glucose transporter 1 (GLUT1), encoded by the SLC2A1 gene, has a major role in glucose metabolism, the dysregulation of which has been implicated in both diseases. We conducted a case-control association study in a sample of 439 non-diabetic patients with late-onset AD and 304 cognitively healthy, non-diabetic elderly controls to determine the potential risk for developing AD associated with SLC2A1 rs841847 polymorphism. The rs841847 C/C genotype occurrence was hig

BACKGROUND: Polygenic risk scores (PRS) have emerged as promising tools for disease risk stratification. However, their validity across different populations remains unclear, particularly for autoimmune diseases, where environmental factors may play crucial roles.
METHODS: We calculated the population-level PRS for Sjögren's syndrome using seven validated genetic variants (PGS001308) and allele frequency data from the 1000 Genomes Project Phase 3 for five European populations (CEU, TSI, FIN, GBR, and IBS). PRS values were correlated with published prevalence estimates from a systematic litera

Graves' ophthalmopathy (GO) is an autoimmune disorder that affects orbital tissues in approximately 30 % of Graves' disease patients. Single nucleotide polymorphisms (SNPs), particularly in immune-related genes, play a crucial role in the development of GO. This study investigates the association between SNPs in co-stimulatory molecules and specific clinical characteristics of GO, including laterality, orbital pain, swelling, diplopia, exophthalmos, redness, and eyelid retraction. Forty-one patients newly diagnosed with GO were analyzed. Genomic DNA was extracted from their blood, and 98 SNPs

BACKGROUND: The cytochrome P450 family 3 subfamily A polypeptide 5 (CYP3A5) gene plays an important role in renal function through its product's involvement in metabolizing endogenous substances and drugs, including immunosuppressants used following kidney transplantation. A single-nucleotide polymorphism, CYP3A5*3 (rs776746), produces a non-functional variant that may influence progression of chronic kidney disease (CKD) by impairing renal filtration. However, the frequency of the CYP3A5*3 allele in the Thai population and its association with renal parameters remain underexplored. This study
