[In vitro antimycobacterial activities of pyrazinamide analogs: results of screening tests].

Attempting to find new drugs, which are more effective than pyrazinamide against Mycobacterium tuberculosis and also active against M. avium complex (MAC), we synthesized various pyrazinamide analogs and pyrazine derivatives and assayed their antimycobacterial activities in vitro against M. tuberculosis, M. avium and M. intracellulare. As is well known, pyrazinamide is more active in acidic medium than in neutral medium, but the growth of mycobacteria in acidic media is poor and inconsistent. Our preliminary experiments revealed that the relative antimycobacterial activities of test-drugs comp

Performance of a targeted next generation sequencing assay for mycobacterial identification and drug resistance from sputum and isolates.

Accurate identification and genotypic drug resistance prediction in mycobacteria are essential for early diagnosis and appropriate clinical management. Targeted next-generation sequencing (tNGS) detects multiple genetic targets and enables direct testing from primary specimens. We evaluated identification using the Deeplex Myc-TB assay for Mycobacterium tuberculosis complex (MTBC) and nontuberculous mycobacteria (NTM) as well as MTBC drug-resistance prediction from 78 isolates and 102 sputum samples. DNA libraries were sequenced (Illumina MiSeq), and results were analyzed using defined accepta

Toxoplasma TgAtg8-TgAtg3 Interaction Primarily Contributes to Apicoplast Inheritance and Parasite Growth in Tachyzoite.

The apicoplast, which harbors key pathways involved in biosynthesis of vital metabolites, is a unique and essential nonphotosynthetic plastid organelle in apicomplexan parasites. Intriguingly, autophagy-related protein 8 (Atg8), a highly conserved eukaryotic protein, can localize to the outermost membrane of the apicoplast and modulate its inheritance in both Toxoplasma and Plasmodium parasites. The Atg8-Atg3 interaction plays a key role in Atg8 lipidation and localization, and our previously work in Toxoplasma has suggested that the core Atg8-family interacting motif (AIM) in TgAtg3, 239FADI2

Potent Protease Inhibitors of Highly Pathogenic Lagoviruses: Rabbit Hemorrhagic Disease Virus and European Brown Hare Syndrome Virus.

Rabbit hemorrhagic disease (RHD) and European brown hare syndrome (EBHS) are highly contagious diseases caused by lagoviruses in the Caliciviridae family. These infectious diseases are associated with high mortality and a serious threat to domesticated and wild rabbits and hares, including endangered species such as riparian brush rabbits (Sylvilagus bachmani riparius). In the United States (U.S.), only isolated cases of RHD had been reported until Spring 2020. However, RHD caused by GI.2/rabbit hemorrhagic disease virus (RHDV)2/b was unexpectedly reported in April 2020 in New Mexico and has s

Structure-Guided Synthesis of FK506 and FK520 Analogs with Increased Selectivity Exhibit In Vivo Therapeutic Efficacy against Cryptococcus.

Calcineurin is an essential virulence factor that is conserved across human fungal pathogens, including Cryptococcus neoformans, Aspergillus fumigatus, and Candida albicans. Although an excellent target for antifungal drug development, the serine-threonine phosphatase activity of calcineurin is conserved in mammals, and inhibition of this activity results in immunosuppression. FK506 (tacrolimus) is a naturally produced macrocyclic compound that inhibits calcineurin by binding to the immunophilin FKBP12. Previously, our fungal calcineurin-FK506-FKBP12 structure-based approaches identified a non

The emerging role of cholesterol metabolism in gynecologic cancer development and therapy.

The significance of cholesterol metabolism in cancer is a topic of renewed interest. Cholesterol is an essential factor for mammal cells, for it is not only involved in constituting the cell membrane, but also serves as a precursor to steroid hormones and bile acids. Numerous studies have provided increasing evidence of its high relevance to cancer progression. Targeting cholesterol metabolism by using cholesterol metabolism inhibitors has offered another therapeutic strategy for reversing drug resistance in tumors. Here, the regulatory process of cholesterol homeostasis under normal physiolog

Antifungal drug resistance in Candida auris: molecular mechanisms and emerging therapeutic approaches.

The human fungal pathogen Candida auris (Candidozyma auris) has emerged as a major global health threat due to its pronounced skin tropism, efficient healthcare-associated transmission, as well as frequent resistance to antifungal therapy. These features have enabled outbreaks in healthcare settings worldwide and have complicated the management of life-threatening invasive infections. Understanding the molecular basis of antifungal multidrug resistance in C. auris is therefore essential for improving clinical management and developing new therapeutic strategies. In this review, we discuss curr

Arrhythmias in Pregnancy: A Comprehensive Review.

Cardiac arrhythmias are increasingly recognized as contributors to maternal morbidity and mortality, particularly in women with preexisting cardiovascular disease. The hemodynamic, hormonal, and autonomic changes of pregnancy can unmask or exacerbate underlying arrhythmogenic conditions. This review summarizes current data on the classification, epidemiology, and management of arrhythmias during pregnancy. Antiarrhythmic therapy requires careful consideration of maternal benefit versus fetal risk, including concerns about teratogenicity and neonatal drug exposure. Management must be individual

Non-parathyroid Hypercalcaemia in a Patient on Lithium Therapy: A Case Report.

Lithium is a commonly used drug in the treatment of several psychiatric disorders. However, due to its narrow therapeutic range, vigilant monitoring is essential to detect adverse effects. While hypercalcaemia is a documented complication of lithium therapy, it is typically associated with hyperparathyroidism. This case report presents a 67-year-old female on prolonged lithium therapy who developed refractory hypercalcaemia that was not attributable to hyperparathyroidism. Discontinuation of lithium resulted in normalisation of calcium levels, confirming lithium-induced hypercalcaemia in the c
