Brain-derived synaptic vesicles have an intrinsic ability to sequester tubulin.

BACKGROUND: The presence and function of microtubules within the synaptic bouton has long been under investigation. In recent years, evidence has accumulated that connects the synaptic vesicle cluster to the local dynamics of microtubule ends. Nonetheless, one question remains open, namely whether the vesicles influence the availability of tubulin within the synaptic compartment. RESULTS: An analysis of previously published live imaging experiments indicates that tubulin is strongly enriched in the synaptic vesicle cluster. To analyze the vesicle-tubulin interaction directly, we isolated vesi…

Novel trifluoromethylquinoline derivatives as potent tubulin polymerization inhibitors with antitumor activity.

Herein, two series of N-aryl-2-trifluoromethyl-quinoline-4-amine derivatives as tubulin polymerization inhibitors were designed and synthesized. Compounds 14b exhibited potency inhibitory activity against LNCaP cells (IC50 = 0.0027 μM and Selectivity Index = 4.07) which was more effective than the positive control colchicine, while compounds 12, 16a, and 14e exhibited moderate activity against LNCaP cells with IC50 values of 0.28, 0.0413, and 0.43 μM, respectively. Moreover, mechanism studies showed that 14b could disrupt tubulin polymerization effectively, block cell cycle in G2/M phase, and …

Alcohol-induced tubulin post-translational modifications directly alter hepatic protein trafficking.

BACKGROUND: Chronic ethanol exposure leads to enhanced protein acetylation and acetaldehyde adduction. Of the multitude of proteins that are modified on ethanol administration, tubulin is among the best studied. However, an open question is whether these modifications are observed in patient samples. Both modifications have also been implicated in promoting alcohol-induced defects in protein trafficking, but whether they do so directly is also unanswered. METHODS AND RESULTS: We first confirmed that tubulin was hyperacetylated and acetaldehyde-adducted in the livers from ethanol-exposed indiv…

Targeting plasmodium α-tubulin-1 to block malaria transmission to mosquitoes.

Plasmodium ookinetes use an invasive apparatus to invade mosquito midguts, and tubulins are the major structural proteins of this apical complex. We examined the role of tubulins in malaria transmission to mosquitoes. Our results demonstrate that the rabbit polyclonal antibodies (pAb) against human α-tubulin significantly reduced the number of P. falciparum oocysts in Anopheles gambiae midguts, while rabbit pAb against human β-tubulin did not. Further studies showed that pAb, specifically against P. falciparum α-tubulin-1, also significantly limited P. falciparum transmission to mosquitoes. We…

Reduced acetylated α-tubulin in SPAST hereditary spastic paraplegia patient PBMCs.

HSP-SPAST is the most common form of hereditary spastic paraplegia (HSP), a neurodegenerative disease causing lower limb spasticity. Previous studies using HSP-SPAST patient-derived induced pluripotent stem cell cortical neurons have shown that patient neurons have reduced levels of acetylated α-tubulin, a form of stabilized microtubules, leading to a chain of downstream effects eventuating in increased vulnerability to axonal degeneration. Noscapine treatment rescued these downstream effects by restoring the levels of acetylated α-tubulin in patient neurons. Here we show that HSP-SPAST patien…

Tubulin acetylation increases cytoskeletal stiffness to regulate mechanotransduction in striated muscle.

Microtubules tune cytoskeletal stiffness, which affects cytoskeletal mechanics and mechanotransduction of striated muscle. While recent evidence suggests that microtubules enriched in detyrosinated α-tubulin regulate these processes in healthy muscle and increase them in disease, the possible contribution from several other α-tubulin modifications has not been investigated. Here, we used genetic and pharmacologic strategies in isolated cardiomyocytes and skeletal myofibers to increase the level of acetylated α-tubulin without altering the level of detyrosinated α-tubulin. We show that microtub…

Crafting of Neuroprotective Octapeptide from Taxol-Binding Pocket of β-Tubulin.

Microtubules play a crucial role in maintaining the shape and function of neurons. During progression of Alzheimer's disease (AD), severe de … P peptide (davunetide) have entered in AD clinical programs. Therefore, the taxol binding pocket of tubulin could be a potential site for designing of mild and noncytotoxic microtubule stabilizing molecules. Here, we adopted an innovative strategy for the development of a peptide based microtubule stabilizer, considering the taxol binding pocket of β-tubu …

Zampanolide and dactylolide: cytotoxic tubulin-assembly agents and promising anticancer leads.

Zampanolide is a marine natural macrolide and a recent addition to the family of microtubule-stabilizing cytotoxic agents. Zampanolide exhibits unique effects on tubulin assembly and is more potent than paclitaxel against several multi-drug resistant cancer cell lines. A high-resolution crystal structure of αβ-tubulin in complex with zampanolide explains how taxane-site microtubule-stabilizing agents promote microtubule assemble and stability. This review provides an overview of current developments of zampanolide and its related but less potent analogue dactylolide, covering their natural sou…

Potential transition state phosphoramidate inhibitors of beta-tubulin as antifilarial agents.

Transition state phosphoramidate inhibitors of beta-tubulin were designed as potential antifilarial agents. The reaction of 2-aminobenzimidazole with diisopropyl phosphite and carbon tetrachloride at a low temperature gave the unexpected 1-diisopropoxyphosphoryl-2-aminobenzimidazole, which on heating gave the novel benzimidazole derivative, 2-(diisopropoxyphosphoryl)aminobenzimidazole. Both products were fully characterized and the synthetic procedure to both compounds was optimized. The procedure was used to prepare the related 5-benzoyl-2-(diisopropoxyphosphoryl)aminobenzimidazole and 5-benz…

Androgenic regulation of tubulin gene expression in axotomized hamster facial motoneurons.

We have previously demonstrated that systemic administration of testosterone increases the rate of axonal regeneration following facial nerve crush in adult male hamsters. In this investigation, the molecular mechanisms by which androgens may enhance axonal regeneration were examined. Specifically, the following question was addressed using Northern blot and in situ hybridization with three cytoskeletal cDNA probes complementary to beta II-, beta III-, and alpha 1-tubulin mRNA: does exogenous testosterone augment axotomy-induced changes in tubulin mRNA expression in hamster facial motoneurons …

Recombinant monoclonal antibodies for labeling tubulin post-translational modifications in Caenorhabditis elegans.

Post-translational modifications (PTMs) of tubulin regulate microtubule properties and functions. The Verhey lab created recombinant monoclonal antibodies (rMAbs) against tubulin acetylation (rMAb-6-11B-1), tyrosinated α-tubulin (rMAb-YL1/2), and glutamylation (rMAb-GT335) (Blasius et al., 2025; Hotta et al., 2026). Here, we validate these rMAbs in C. elegans hermaphrodites and males. These rMAbs faithfully reproduce the reported cell-type-specific staining patterns of commercial antibodies, providing high-quality, cost-effective resources for studying the tubulin code.

Adaptations in Plasmodium tubulin determine distinct microtubule architectures, mechanics and drug susceptibility.

Microtubules are ubiquitous yet diverse cytoskeleton filaments. However, tubulin conservation presents challenges in understanding the origins of diverse microtubule architectures. The mechanisms by which microtubule architecture varies through the life cycle of the malaria-causing parasite Plasmodium are not understood and provide a valuable framework for exploring how intrinsic properties of tubulin contribute to architectural variety. Using parasite-purified tubulin, we determine P. falciparum microtubule structures by cryo-electron microscopy. Parasite-specific sequences change the tubulin…

Tubulin glutamylation: a key regulator of flagella, cilia, centrosomes, and disease pathways.

Tubulin glutamylation is an essential post-translational modification that expands the functional diversity of microtubules in many cellular structures, including flagella, motile cilia, primary cilia, centrosomes, and neurons. This modification adds variable lengths of glutamate side chains to the C-terminal tails of tubulin, creating a finely tuned biochemical signal that regulates microtubule stability, motor protein movement, the activity of severing enzymes, and the recruitment of key signaling molecules. Growing evidence shows that glutamylation is not uniformly distributed but instead f…

Verubulin and its derivatives: Progress and promise in tubulin-targeted cancer therapy.

Verubulin (MPC-6827) is a microtubule-destabilizing agent that binds to the colchicine site of tubulin and has been primarily investigated for the treatment of glioblastoma and other malignancies. Its ability to interfere with microtubule dynamics, induce mitotic arrest, and retain efficacy in multidrug-resistant tumour cells has positioned it as a valuable scaffold in the development of next-generation anticancer agents. Over the past decade, extensive efforts have been dedicated to optimizing verubulin and its derivatives through modifications that enhance potency and selectivity. Additional…

Affinity-purified Plasmodium tubulin provides a key reagent for antimalarial drug development.

Hirst et al. used a TOG-domain-based affinity-purification approach to reconstitute and define the in vitro dynamics of blood-stage Plasmodium falciparum αβ-tubulin. This provides a key reagent for defining parasite microtubule (MT) dynamics and for evaluating the efficacy of anti-MT drugs throughout the complex parasite life cycle.

Cryo-ET of Toxoplasma parasites gives subnanometer insight into tubulin-based structures.

Tubulin is a conserved protein that polymerizes into different forms of filamentous structures in Toxoplasma gondii, an obligate intracellular parasite in the phylum Apicomplexa. Two key tubulin-containing cytoskeletal components are subpellicular microtubules (SPMTs) and conoid fibrils (CFs). The SPMTs help maintain shape and gliding motility, while the CFs are implicated in invasion. Here, we use cryogenic electron tomography to determine the molecular structures of the SPMTs and CFs in vitrified intact and detergent-extracted parasites. Subvolume densities from detergent-extracted parasites…

Systematic Analysis of Clemastine, a Candidate Apicomplexan Parasite-Selective Tubulin-Targeting Agent.

Apicomplexan parasites, such as Toxoplasma gondii, Plasmodium spp., Babesia spp., and Cryptosporidium spp., cause significant morbidity and mortality. Existing treatments are problematic due to toxicity and the emergence of drug-resistant parasites. Because protozoan tubulin can be selectively disrupted by small molecules to inhibit parasite growth, we assembled an in vitro testing cascade to fully delineate effects of candidate tubulin-targeting drugs on Toxoplasma gondii and vertebrate host cells. Using this analysis, we evaluated clemastine, an antihistamine that has been previously shown t…

A new class of cytotoxic agents targets tubulin and disrupts microtubule dynamics.

Despite the advances in treatment strategies, cancer is still the second leading cause of death in the USA. A majority of the currently used cancer drugs have limitations in their clinical use due to poor selectivity, toxic side effects and multiple drug resistance, warranting the development of new anticancer drugs of different mechanisms of action. Here we describe the design, synthesis and initial biological evaluation of a new class of antimitotic agents that modulate tubulin polymerization. Structurally, these compounds are chalcone mimics containing a 1-(1H-imidazol-2-yl)ethan-1-one moie…
