The Inverted U-Curve Association of Fluoride and Osteoclast Formation in Mice.

The effect of fluoride on osteoclasts is still controversial. In the past, researchers thought that the effects of fluoride on osteoclast and osteoblast formation occurred in a dose-dependent pattern. However, our previous in vitro study showed fluoride elicited a notably different effect on osteoclast formation. To further verify the relationship between fluoride and osteoclast formation in vivo, 60 male C57BL/6 mice were randomly divided into three groups: two treatment groups consuming water supplemented with 50 and 100 mg/L of fluoride, and a third control group with nonsupplemented water.

Otosclerosis 2: the medical management of otosclerosis.

The rationale for medical therapy for otospongiosis is to slow down and eventually stop the phase of bone resorption. There is some increase in the incidence of stapedial otospongiosis in a low-fluoride area compared with a high-fluoride area. Sodium fluoride treatment has a role to play in preventing the onset and progression of hearing loss in patients suffering from otosclerosis. Sodium fluoride therapy has been shown to have some beneficial effect on dizziness associated with otosclerosis. In view of the possibility of systemic side effects of sodium fluoride therapy, a regular follow up o

Differences in loosely bound fluoride formation and anticaries effect of resin-based fluoride varnishes.

OBJECTIVE.  Our in vitro study evaluated calcium fluoride formation in enamel and the anticaries effect of seven resin-based varnishes under cariogenic challenge. METHODS.  Enamel blocks were subjected to pH cycling. The experimental groups received fluoride varnish application, the positive control received topical fluoride gel treatment, and the negative control did not receive any treatment. The pH cycling surface hardness (SH1 ) and integrated loss of subsurface hardness (ΔKHN) were then determined. We measured the amount of fluoride released into the demineralizing and remineralizing (DE-

Relationship between bone fluoride content and histological evidence of calcification defects in osteoporotic women treated long term with sodium fluoride.

Fluoride treatment is used to increase bone formation and cancellous bone mass in patients suffering from postmenopausal osteoporosis with vertebral fractures. Patients submitted to similar therapeutic protocols have shown various histological responses to the treatment, some developing calcification defects and others not. In fact, the bone histological response to fluoride salts depends on the cumulative uptake of fluoride by bone. To clarify the relationship between the presence of calcification defects (identified by the presence of mottled bone and linear formation defects) and the bone f

Delayed fatal hyperkalemia in a patient with acute fluoride intoxication.

A 19-year-old man presented with acute fluoride poisoning. Initially his serum electrolytes were normal, but two hours later he developed ECG evidence of hyperkalemia followed by refractory ventricular fibrillation, suggesting that hyperkalemia may be important in the cardiotoxicity of acute fluoride intoxication. Treatment of fluoride-induced hyperkalemia consists of removal of fluoride from the body by dialysis, binding fluoride with aluminum or calcium, or enhancing fluoride excretion by inducing a metabolic alkalosis. Direct treatment of the hyperkalemia with glucose, insulin, and bicarbon

Effect of polydopamine and fluoride ion coating on dental enamel remineralization: an in vitro study.

BACKGROUND: Fluoride treatment is one of the most effective dental caries prevention methods. To continuously prevent dental caries, stably immobilizing the fluoride on the tooth enamel is highly desirable. This study aimed to evaluate the remineralization of tooth enamels by one-pot coating using polydopamine and fluoride ions. METHODS: To prepare the enamel specimens for polydopamine- and fluoride ion-coating, they were treated with polydopamine- and fluoride-containing gels. The enamel specimens were collected from human molars in a blind manner (n = 100) and were randomized into five trea

Formation of fluoride on enamel in vitro after exposure to fluoridated mouthrinses.

The aim of this study was to quantify the formation of alkali-soluble fluoride (loosely bound fluoride such as calcium fluoride-like material and absorbed fluoride) and alkali-insoluble fluoride (firmly bound fluoride or apatitically bound fluoride) when fluoride mouthrinsing solutions were applied on sound human enamel in vitro. Two commercial products containing 0.2% or 0.05% NaF were used during 30 sec, 60 sec, 5 min, and 60 min. The formation of loosely bound fluoride was determined by KOH extraction and visualized by scanning electron microscopy. The firmly bound fluoride was measured by

Fluoride bioavailability from immediate-release sodium fluoride with calcium carbonate compared with slow-release sodium fluoride with calcium citrate.

The circadian variation in serum fluoride was compared between treatment with immediate-release sodium fluoride (IR-NaF) (30 mg) and calcium carbonate (500 mg calcium) and slow-release sodium fluoride (SR-NaF) (25 mg) and calcium citrate (400 mg calcium) in seven patients with postmenopausal osteoporosis maintained on long-term fluoride treatment. During 12 h following a dose of SR-NaF, serum fluoride levels could be largely kept within the therapeutic window (believed to be 95-190 ng/ml or 5-10 mumol/l). In contrast, IR-NaF produced a wide circadian fluctuation with peak-to-trough change of a

Anabolic effects of fluoride on bone.

Fluoride exerts a biphasic action at the level of osteoblasts, on bone mineral, on bone structure and function, and in the treatment of osteoporosis. At low circulating concentrations, skeletal uptake of fluoride is limited and the effects are beneficial. At higher concentrations and greater skeletal uptake, fluoride may cause the formation of abnormally mineralized bone of impaired quality. A new treatment program entailing intermittent slow release sodium fluoride (SR-NaF) with continuous calcium citrate may capture desirable qualities of fluoride without toxic effects, and be therapeuticall

Effect of fluoride on insulin level of rats and insulin receptor expression in the MC3T3-E1 cells.

Studies on the role of insulin and insulin receptor (InsR) in the process of skeletal fluorosis, especially in osteogenic function, are rare. We evaluated the effect of increasing F⁻ doses on the marker of bone formation, serum insulin level and pancreatic secretion changes in vivo and mRNA expression of InsR and osteocalcin (OCN) in vitro. Wistar rats (n = 50) were divided into two groups, i.e. a control group and fluoride group. The fluoride groups were treated with fluoride by drinking tap water containing 100 mg F⁻/L. The fluoride ion-selective electrode measured the fluoride concentration

Critical evaluation of the composition and use of topical fluorides, with emphasis on the role of calcium fluoride in caries inhibition.

There is evidence that a major part of the fluoride which is retained on teeth during topical application is calcium fluoride or calcium fluoride-like, and that this material is relatively stable in the mouth. This is due to surface adsorption of phosphate (HPO4(2-)) ions onto the calcium fluoride surface. Calcium fluoride releases fluoride during caries challenges due to reduced concentration of HPO4(2-) at acid pH. Normally, the fluoride released from calcium fluoride during caries challenges is subsequently built into hydroxyfluorapatite through dissolution/re-precipitation reactions. It ap

Elemental changes in tooth hard tissues during demineralization and fluoride-induced remineralization: an SEM-EDX study.

BACKGROUND: Tooth enamel and dentin are the most highly mineralized tissues in the human body, and their structural stability depends on the balanced content of calcium (Ca), phosphorus (P), and fluoride (F). Demineralization leads to the loss of these ions, weakening the tooth structure and increasing susceptibility to caries. OBJECTIVE: This study aimed to evaluate elemental changes in Ca, P, and F in intact, demineralized, and remineralized teeth across the enamel, dentin-enamel junction and peripulpal dentin. MATERIALS AND METHODS: 45 extracted human teeth were longitudinally sectioned t
