BACKGROUND: Coxsackievirus B3 and B4 (CVB) myocarditis was assessed by a more than twofold change in titer of the microneutralization tests against enteroviruses within 3 weeks in all patients, by an endomyocardial biopsy indicative of active myocarditis in eight cases, and by pericardial effusion and acute cardiomegaly in two patients. In all endomyocardial biopsies, immunoglobulin binding to the sarcolemma and to the interstitial tissue was demonstrated irrespective of an infiltrate being at the same focus or not. IgG binding was found in nine, IgM and IgA in seven, C3 in C1q in three, and C

We present an analysis of experimental steady-state properties of charge movements in voltage-clamped frog skeletal muscle; by adopting an approach based on equilibrium statistical mechanics, detailed assumptions about the dynamics of the charge movement were avoided. Different components of the charge movements, as characterized by their sensitivities to local anaesthetics, were taken to correspond to different types of independent subsystem (integral membrane proteins). Quantitative agreement with the data for the q beta and q gamma components was obtained for the simplest subsystems, having

A 35-year-old male patient presented fluctuating bilateral lower extremity weakness for 3 years. Physical examination showed grade 4 proximal muscle weakness in both lower extremities and grade 5 distal muscle weakness. Laboratory data revealed elevated creatine kinase, triglycerides, and cholesterol. Muscle pathology showed deposition of lipid droplet under the sarcolemma. Bone densitometry indicated severe osteoporosis. Next-generation sequencing revealed a pathogenic mutation in the ETFDH gene. The patient was diagnosed with late-onset multiple acyl-CoA dehydrogenase deficiency. After ribof

Prospects of the use of flow cytometry analysis for investigation of forming K(+)- equilibrium membrane potential on the experimental model of myometrium plasma membrane vesicles in the presence of valinomycine using potential-sensitive probe of DiOC6(3). Transmembrane potential magnitude corresponds to magnitude by Nernst's equation. H2O2 and NO2-, probably, increase permeability of membrane for K+ and result in potential dissipation. Given effect is not shown for sodium nitroprusside. The obtained results confirm an assumption as to enhancing the passive transport for K+ through sarcolemma u

We have reported a 58-year-old Japanese female with polymyositis, primary biliary cirrhosis (PBC) and arrhythmia. In contrast to the previously reported 13 cases of polymyositis associated with PBC, symptoms and laboratory data abnormalities responded to oral administration of predonisolone. Interestingly, immunohistochemical and immunoblot analyses of biopsied skeletal muscle revealed diminished expression of dystrophin carboxyl-terminal domain in the sarcolemma, suggesting that, in analogy to Duchenne muscular dystrophy, secondary abnormality of the link between the basal lamina and cytoskel

BACKGROUND: Subcellular localization and function of L-type calcium channels (LTCCs) play an important role in regulating contraction of cardiomyocytes. Understanding how this is affected by the disruption of transverse tubules during heart failure could lead to new insights into the disease.
METHODS: Cardiomyocytes were isolated from healthy donor hearts, as well as from patients with cardiomyopathies and with left ventricular assist devices. Scanning ion conductance and confocal microscopy was used to study membrane structures in the cells. Super-resolution scanning patch-clamp was used to

Duchenne muscular dystrophy (DMD) is a severe X-linked disease characterized by progressive muscle weakness. It is caused by a variety of DMD gene pathogenic variations (large deletions or duplications, and small mutations) which leads to the absence or to a decreased amount of dystrophin protein. The allelic Becker muscular dystrophy is characterized by later onset and milder muscle involvement, and other rarer phenotypes might also be associated, such as dilated cardiomyopathy, cognitive impairment, and other neurological signs. Following the identification of the genetic cause and the disea

The highly organized transverse T-tubule membrane system represents the ultrastructural substrate for excitation-contraction coupling in ventricular myocytes. While the architecture and function of T-tubules have been well described in animal models, there is limited morpho-functional data on T-tubules in human myocardium. Hypertrophic cardiomyopathy (HCM) is a primary disease of the heart muscle, characterized by different clinical presentations at the various stages of its progression. Most HCM patients, indeed, show a compensated hypertrophic disease ("non-failing hypertrophic phase"), with

BACKGROUND: Duchenne muscular dystrophy (DMD) is a fatal muscle-wasting disorder caused by genetic loss of dystrophin protein. Extracellular microRNAs (ex-miRNAs) are putative, minimally invasive biomarkers of DMD. Specific ex-miRNAs (e.g. miR-1, miR-133a, miR-206, and miR-483) are highly up-regulated in the serum of DMD patients and dystrophic animal models and are restored to wild-type levels following exon skipping-mediated dystrophin rescue in mdx mice. As such, ex-miRNAs are promising pharmacodynamic biomarkers of exon skipping efficacy. Here, we aimed to determine the degree to which ex-

Rhabdomyolysis is a common disorder that occurs as a primary disease or as a complication of a broad spectrum of other diseases. Although some cases are caused by hereditary metabolic or structural abnormalities of the skeletal muscle cell, the majority of cases occur in healthy persons as a result of exhaustive exercise, infections, intoxications, deficiency states, or trauma. Although the causes of rhabdomyolysis are diverse, current evidence suggests that there may be a common final pathway that mediates cellular injury. Thus some noxious factor, perhaps a drug that injures the plasma membr

A 58-year-old Japanese woman was diagnosed with anti-signal recognition particle (SRP)-positive dermatomyositis associated with Sjögren's syndrome, rheumatoid arthritis and lung adenocarcinoma. She presented with cutaneous lesions, including ulceration of her right middle finger. Tissue specimens obtained from her right deltoid muscle were positive for CD4+ T-cell infiltration and the sarcolemma showed the upregulation of major histocompatibility complex (MHC) class I antigens. The present case suggests that overlapping autoimmune diseases or complications of malignancy may result in an atypic

Biopsy specimens from patients with treated or untreated polymyositis and muscular dystrophy controls were examined by indirect immunoperoxidase staining with a panel of monoclonal antibodies to human leucocyte antigens. In untreated polymyositis, helper/inducer T cells were the predominant T cell subset. In treated cases few T cells were seen. Overall, few T cells were seen in dystrophic cases, most infiltrating cells being dendritic and lacking T cell antigens. Staining of sarcolemma with anti-HLA class 1 antibody is weak or negative except in areas adjacent to infiltrating leucocytes or whe

Intracellular pH must be maintained within the physiological range (7.15-7.25) to ensure cellular homeostasis. In the heart, excitation-contraction coupling is closely dependent on intracellular pH because its proper regulation helps the correct management of intracellular Ca2+. In addition, it is important to know the spatial distribution of intracellular pH microdomains because it helps us to better understand the compartmentalized regulation of different channels, transporters, and enzymes. Therefore, maintaining cardiac intracellular pH at physiological levels is of crucial importance for

Transient receptor potential canonical 1 (TRPC1) protein is abundantly expressed in cardiomyocytes. While TRPC1 is supposed to be critically involved in cardiac hypertrophy, its physiological role in cardiomyocytes is poorly understood. We investigated the subcellular location of TRPC1 and its contribution to Ca2+ signaling in mammalian ventricular myocytes. Immunolabeling, three-dimensional scanning confocal microscopy and quantitative colocalization analysis revealed an abundant intracellular location of TRPC1 in neonatal rat ventricular myocytes (NRVMs) and adult rabbit ventricular myocytes

AIMS: In heart failure transverse-tubule (t-tubule) remodelling disrupts calcium release, and contraction. T-tubules in human failing hearts exhibit increased labelling by wheat germ agglutinin (WGA), a lectin that binds to the dystrophin-associated glycoprotein complex. We hypothesized changes in this complex may explain the increased WGA labelling and contribute to t-tubule remodelling in the failing human heart. In this study we sought to identify the molecules responsible for this increased WGA labelling.

Bcl2-associated athanogene 3 (BAG3) is a 575 amino acid anti-apoptotic protein that is constitutively expressed in the heart. BAG3 mutations, including mutations leading to loss of protein, are associated with familial cardiomyopathy. Furthermore, BAG3 levels have been found to be reduced in end-stage non-familial failing myocardium. In contrast to neonatal myocytes in which BAG3 is found in the cytoplasm and involved in protein quality control and apoptosis, in adult mouse left ventricular (LV) myocytes BAG3 co-localized with Na(+)-K(+)-ATPase and L-type Ca(2+) channels in the sarcolemma and

A boy with congenital generalized lipodystrophy type 4 with muscular dystrophy presented in infancy with delay in motor milestones and a persistent elevation of CK. There was no associated mental retardation. He was followed up to 3 years and 11 months; he had a homozygous c.696_697insC mutation in polymerase I and transcript release factor (PTRF). He started to walk at 2 years and 6 months although he did not have mental retardation. Insulin resistance appeared at 3 years and 11 months of age. PTRF immunostaining positivity was absent in the muscle but caveolin-3 was preserved in the sarcolem

A study of myocardium of patients with mitral stenosis was performed using ultrahistochemical methods at different stages of disease and in the course of valve replacement operation. A quantitative analysis of degenerative changes was introduced, the results of which correlated with the severity of heart failure. A pronounced heterogeneity in mitochondrial enzyme activity has been detected cytochemically. In some apparently "unaffected" cardiomyocytes an active alterative process was expressed in an increase of sarcolemma permeability for electron-microscopic tracers. An elevated fenestration

We herein report a 56-year-old Japanese woman who had been diagnosed with hereditary angioedema. She experienced progressing muscle weakness and pain in the upper and lower extremities. Blood tests revealed a marked increase in creatine kinase levels; however, myositis-specific autoantibodies were not detected. Serum C1-inhibitor activity and C4 levels were low. A muscle biopsy showed mild muscle fiber necrosis and C5b-9 deposition in the endomysial capillary vessel walls and sarcolemma, mimicking necrotizing myopathy. These results suggest that C1-inhibitor deficiency induces myositis-like sy
