Plasma Extracellular Vesicles from Glioblastoma Patients Affect Phenotype and Activate Receptor Tyrosine Kinases in Glioblastoma Cells.

Extracellular vesicles are important transmitters of oncogenic signals between cancer cells. Despite numerous studies of vesicles from model glioblastoma (GBM) cells, information on the effects of vesicles from GBM patients on the development of model GB cells is lacking. In this study, plasma vesicles from healthy donors (HVs) and GBM patients (GVs) were studied to affect expression of differentiation (GFAP and GLT-1) and stemness (CD9 and CD133) markers and expression and activation of the oncogenic receptor tyrosine kinases EGFR and PDGFRβ in GBM cells (primary GBM011 and model U251 MG cell

Formation of Adaptive Defense to Low Temperatures in Arabidopsis thaliana.

The effects of repeated cold stress on physiological, transcriptional, and metabolic processes was studied in Arabidopsis thaliana. An experiment included stages of initial chilling, acclimation, and repeated cold treatment. Repeated cold stress increased electrolyte leakage and decreased the chlorophyll a content, indicating a disruption of the membrane integrity and a restructuring of the photosynthetic apparatus. Expression analysis showed upregulation of the DREB1A, MYC2, COR15A, and COR15B genes, which are associated with the cold response and stress memory formation. Transcription of the

Calcium Signaling Modulator 4-MPTC Reduces Proliferative and Migration Activities of MDA-MB-468 Breast Cancer Cells.

Cell malignant transformation is accompanied by a remodeling of various cell life-support systems, including calcium signaling. Modulation of the calcium signal can significantly sensitize cancer cells, making them more sensitive to anticancer drugs. A search for selective compounds that influence the cell calcium response is a promising area in the development of combination therapies for oncology diseases. The aim of this study was to determine the effect of 4-MPTC on the invasive potential of triple-negative breast cancer cells. 4-MPTC was previously selected as a modulator of the Stim2-dep

Diversity of Toll-like Receptor Gene Transcripts in Hemocytes of Planorbarius corneus Mollusks (Gastropoda, Pulmonata) Uninfected and Infected with Bilharziella polonica.

The hemocyte transcriptome was for the first time obtained and analyzed in uninfected Planorbarius corneus snails and snails naturally infected with Bilharziella polonica parthenites (Schistosomatidae). Transcripts encoding all groups of immune factors previously described in other gastropods were found in assemblies. Pathogen recognition molecules were the most diverse group of transcripts encoding immunity factors. Toll-like receptors (TLRs) of 11 types were identified. The TLR set did not differ between infected and uninfected snails. The data expand the knowledge of snail immune responses

Biologically Active Compounds from Bark of Juniperus communis (Cupressaceae).

The component composition of the bark was studied in Juniperus communis L. (Cupressaceae). The juniper bark was shown to contain 3.5 times more extractives than wood. Individual component compositions of ethanol and petroleum extracts of J. communis bark was determined by gas chromatography-mass spectrometry. The ethanol and petroleum bark extracts were found to differ in component composition. Monosaccharides, disaccharides, and resin acids were major constituents of the J. communis ethanol extract, while resin acids, fatty acids, and sterols were major constituents of the petroleum extract.

Organometric and Biochemical Regularities of Transition to Aging.

In a study of male Wistar rats, age-dependent patterns of body mass index (BMI) dynamics and mass coefficient (MC) involution were identified for nine visceral organs with high metabolic activity at young reproductive age (3 months) and low metabolic activity during the transition to aging (12 months). Epididymal fat (EF) was the only organ to demonstrate a progressive increase. Synchronous involution of the thymus and muscles with a progressive accumulation of visceral fat can serve as an integral criterion of early aging that is more informative than assessment of individual organs. A phasic

Hematological Parameters in the Ontogeny of the Pond Bat Myotis dasycneme Boie, 1825 from the Urals.

Hematological parameters were for the first time studied in female pond bats (Myotis dasycneme Boie, 1825) in the postnatal period. Significant changes in red blood cell indices were found to accompany the growth and development of pond bats, while white blood cell indices remained stable. Significant differences in erythropoiesis were observed between adults and yearlings. With age, red blood cell count, hemoglobin, and platelet aggregation increased in pond bats; blood clotting processes became faster; and platelet involution was detected (p < 0.05). Neutrophil counts were elevated in adult

First Description of Typhlosole in Ascidian Styela clava: Structure and Putative Function.

The ascidian Styela clava is an invasive species that easily colonizes new water areas and has a wide distribution. A previously undescribed typhlosole was discovered in S. clava and studied using histological techniques, transmission electron microscopy and computer microtomography. The typhlosole is a large fold of the dorsal intestinal wall that starts in the lower quarter of the stomach and continues to the rectum. Its shape varies greatly: from a rounded protrusion to a spirally twisted fold. The typhlosole is formed by the intestinal epithelium and a thick connective tissue layer, in whi

Generation of Long-Lived CHO Cells Suitable for Production of Afucosylated Antibodies and Fc-Fusion Proteins.

Using genome editing, we created a homozygous α-(1,6)-fucosyltransferase (FUT8-/-) knockout in apoptosis-resistant CHO 4BGD cells, yielding the new 4BGD-F cell line. Combining CRISPR/Cas9 with paired gRNAs and non-specific puromycin selection yielded a cell population with an exceptionally high FUT8 knockout frequency, obviating the need for metabolic enrichment with lentil lectin (Lens culinaris agglutinin, LCA). Despite impaired clonogenicity of the knockout cells, we successfully isolated multiple clonal cell lines harboring extensive biallelic FUT8 deletions. Isolated clones with biallelic

Thioridazine Induces Increase in Expression of the Pyruvate Transporter MPC1 Associated with Immune Infiltration in Malignant Tumors.

The MPC1 gene is involved in the transport of pyruvate into mitochondria, playing an important role in metabolic processes. Recently, it has been reported that higher MPC1 expression correlates with an increased number of immune cells in human cervical and lung cancers, indicating an enhanced antitumor immune response. Reduced MPC1 levels in gastric tumors are associated with a more severe disease course. Correlational analysis of the MPC1 gene in human lung, hippocampus and frontal cortex tissue samples based on data from the GTEx database revealed associations of this gene with schizophrenia

Development and Efficacy Evaluation of Multivalent DNA Vaccine against Brucellosis.

Attenuated live Brucella strains are currently used in vaccines against brucellosis, causing serious side effects in animals and being hardly suitable for human vaccination. To develop a safer vaccine against brucellosis, plasmid DNA was constructed to express six Brucella proteins in mammalian cells. Linear polyethylenimine was used to increase the efficiency of DNA delivery into cells. Data on optimization of the conditions for the formation of DNA-polyethylenimine complexes were described. A single intramuscular or subcutaneous injection of plasmid DNA complexes with polyethylenimine at a l

Synaptic Vesicle Exocytosis and Endocytosis in Motor Nerve Endings of Transgenic Mice Modeling Amyotrophic Lateral Sclerosis upon Antioxidant Treatment and Gene-Cell Therapy.

Exocytosis and endocytosis of synaptic vesicles were studied in experiments with motor nerve endings of diaphragm neuromuscular preparations isolated from transgenic mice with a model of amyotrophic lateral sclerosis (ALS); treatment simulated antioxidant (edaravone) and gene-cell (umbilical cord blood mononuclear cells (UCB-MNCs) producing VEGF, GDNF, and NCAM) therapies. None of the treatments was found to significantly change the FM 1-43 fluorescent dye loading due to synaptic vesicle endocytosis. Gene-cell therapy increased the rate of dye unloading due to synaptic vesicle exocytosis, whil

Etruscan Wolf Canis etruscus (Canidae, Carnivora) from the Early Pleistocene of Crimea (Taurida Cave).

The cranial and mandibular fragments of the medium-sized canid from the Early Pleistocene locality of Taurida cave (about 1.8-1.5 Ma) in Crimea are described and included in the hypodigm of the species Canis etruscus Forsyth Major, 1877, a common member of the Late Villafranchian fauna of the Circum-Mediterranean region. This species is reported for the first time from the Northern Black Sea region. The discovered evidence reinforce previous published idea regarding the periodically use of the Taurida cave by canids and other large predators as a den and shelter.

The Role of BDNF in the Antidepressant Effects of Electroconvulsive Therapy.

Electroconvulsive therapy (ECT) is an effective treatment method for depression therapy. It produces a number of biological effects, including neurotrophic factors regulation. In the present paper, we investigated the ECT response in depressed rats subjected to the variable frequency ultrasound (20-45 kHz) and examined the contribution of brain-derived neurotrophic factor (BDNF) expression changes to the observed effects. The obtained results reflect the therapeutic potential of ECT for the treatment of depressive-like state in rodents and indicate the role of BDNF in these processes. In futur
