Silicone hydrogels grafted with natural amino acids for ophthalmological application.

In this report, protein repelling silicone hydrogels with improved hydrophilicity were prepared by photo-polymerization of silicone-containing monomer and glycidyl methacrylate followed by grafting zwitterionic amino acids. The grafted silicone hydrogels possessed excellent hydrophilic surfaces due to the enrichment of amino acids, which was confirmed by attenuated total reflectance Fourier transform infrared spectroscopy, X-ray photoelectron spectroscopy, contact angle, and equilibrium water content measurements. Remarkable resistance to bovine serum albumin and lysozyme fouling was observed

A novel approach for targeted delivery to motoneurons using cholera toxin-B modified protocells.

BACKGROUND: Trophic interactions between muscle fibers and motoneurons at the neuromuscular junction (NMJ) play a critical role in determining motor function throughout development, ageing, injury, or disease. Treatment of neuromuscular disorders is hindered by the inability to selectively target motoneurons with pharmacological and genetic interventions. NEW METHOD: We describe a novel delivery system to motoneurons using mesoporous silica nanoparticles encapsulated within a lipid bilayer (protocells) and modified with the atoxic subunit B of the cholera toxin (CTB) that binds to ganglioside

Decorating CdTe QD-Embedded Mesoporous Silica Nanospheres with Ag NPs to Prevent Bacteria Invasion for Enhanced Anticounterfeit Applications.

Quantum dots (QDs) as potent candidates possess advantageous superiority in fluorescence imaging applications, but they are susceptible to the biological circumstances (e.g., bacterial environment), leading to fluorescence quenching or lose of fluorescent properties. In this work, CdTe QDs were embedded into mesoporous silica nanospheres (m-SiO2 NSs) for preventing QD agglomeration, and then CdTe QD-embedded m-SiO2 NSs (m-SiO2/CdTe NSs) were modified with Ag nanoparticles (Ag NPs) to prevent bacteria invasion for enhanced anticounterfeit applications. The m-SiO2 NSs, which serve as intermediat

Harnessing silica-coated silver nanoparticles for combating multidrug-resistant Pseudomonas aeruginosa.

Multidrug-resistant (MDR) Pseudomonas aeruginosa poses a critical challenge in clinical settings because of its resistance to conventional antibiotics. This study investigated the antibacterial potential of silica-coated silver nanoparticles (SiO₂@AgNPs) against MDR P. aeruginosa and explored their synergistic interactions with selected antibiotics. A total of 450 pus samples were processed for bacterial isolation, and P. aeruginosa was identified using standard microbiological methods. MDR strains were confirmed using MIC-based VITEK antimicrobial susceptibility testing and RT-PCR for resista

3D-printed skeletal tissue analogues using bone decellularized extracellular Matrix with PCL-bioglass composite: A biomimetic approach.

Skeletal tissue injuries and defects remain a major clinical challenge due to the limited regenerative capacity of bone with critical-sized defects and limitations in current grafting methods. Bone-derived decellularized matrix (dECM) offers a bioactive composition with growth factors that closely replicate the native tissue analogues. dECM functions as bioactive scaffold that can replicate the native extracellular matrix for promoting effective tissue repair. In this study, dECM was processed into a printable bioink along with PCL and bioglass to fabricate a three-dimensional scaffold using e

Impact of antibacterial composite resin on salivary pH dynamics in high-caries-risk children: An ex-vivo study.

OBJECTIVES: Composite resin restorations in primary teeth frequently fail due to secondary caries, which result from bacterial colonization and acid production at the tooth-restoration interface. This study aimed to evaluate whether composite resins containing quaternary ammonium silica (QASi), known for their antibacterial properties, can reduce bacterial acidification and lower the risk of secondary caries in high-caries-risk children compared to conventional restoratives. METHODS: Saliva samples (10 µL saliva, ∼10⁶ bacteria) from 34 high-caries-risk children were applied ex vivo directly t

Additive manufacturing of silicon nitride fiber-reinforced polyetheretherketone composites with enhanced mechanical strength and multifunctional bioactivity for load-bearing bone defect repair.

Polyether ether ketone (PEEK) is increasingly applied in bone defect repair due to its excellent biocompatibility and absence of artifact formation. However, the bio-inertness and inadequate mechanical properties of untreated PEEK remain significant challenges for PEEK-based implants. Hence, this study prepares a series of silicon nitride (Si3N4) fiber-reinforced PEEK composite porous scaffolds using twin-screw melt mixing-extrusion and material extrusion 3D printing. Comprehensive evaluations assess the mechanical properties, biocompatibility, osteogenic differentiation, angiogenesis activiti

Inflammation-Responsive Mesoporous Silica Nanoparticles with Synergistic Anti-inflammatory and Joint Protection Effects for Rheumatoid Arthritis Treatment.

PURPOSE: Joint destruction is a major burden and an unsolved problem in rheumatoid arthritis (RA) patients. We designed an intra-articular mesoporous silica nanosystem (MSN-TP@PDA-GlcN) with anti-inflammatory and joint protection effects. The nanosystem was synthesized by encapsulating triptolide (TP) in mesoporous silica nanoparticles and coating it with pH-sensitive polydopamine (PDA) and glucosamine (GlcN) grafting on the PDA. The nano-drug delivery system with anti-inflammatory and joint protection effects should have good potency against RA. METHODS: A template method was used to synthes

CdTe QDs-sensitized TiO2 nanocomposite for magnetic-assisted photoelectrochemical immunoassay of SARS-CoV-2 nucleocapsid protein.

A sensitive, reliable, and cost-effective detection for SARS-CoV-2 was urgently needed due to the rapid spread of COVID-19. Here, a "signal-on" magnetic-assisted PEC immunosensor was constructed for the quantitative detection of SARS-CoV-2 nucleocapsid (N) protein based on Z-scheme heterojunction. Fe3O4@SiO2@Au was used to connect the capture antibody to act as a capture probe (Fe3O4@SiO2@Au/Ab1). It can extract target analytes selectively in complex samples and multiple electrode rinsing and assembly steps were avoided effectively. CdTe QDs sensitized TiO2 coated on the surface of SiO2 sphere

Molecularly Imprinted Silica-Coated CdTe Quantum Dots for Fluorometric Determination of Trace Chloramphenicol.

A dual recognition system with a fluorescence quenching of quantum dots (QDs) and specific recognition of molecularly imprinted polymer (MIP) for the detection of chloramphenicol (CAP) was constructed. MIP@SiO2@QDs was prepared by reverse microemulsion method with 3-aminopropyltriethoxysilane (APTS), tetraethyl orthosilicate (TEOS) and QDs being used as the functional monomer, cross-linker and signal sources, respectively. MIP can specifically recognize CAP, and the fluorescence of QDs can be quenched by CAP due to the photo-induced electron transfer reaction between CAP and QDs. Thus, a metho

Smart pH-Regulated Switchable Nanoprobes for Photoelectrochemical Multiplex Detection of Antibiotic Resistance Genes.

Antibiotic resistance, encoded via particular genes, has become a major global health threat and substantial burden on healthcare. Hence, the facile, low-cost, and precise detection of antibiotic resistance genes (ARGs) is crucial in the realm of human health and safety, especially multiplex sensing assays. Here, a smart pH-regulated switchable photoelectrochemical (PEC) bioassay has been created for ultrasensitive detection of two typical subtypes of penicillin resistance genes bla-CTX-M-1 (target 1, labeled as TDNA1) and bla-TEM (target 2, labeled as TDNA2), whereby pH-responsive antimony ta

Bis(zinc(II)-dipicolylamine)-functionalized sub-2 μm core-shell microspheres for the analysis of N-phosphoproteome.

Protein N-phosphorylation plays a critical role in central metabolism and two/multicomponent signaling of prokaryotes. However, the current enrichment methods for O-phosphopeptides are not preferred for N-phosphopeptides due to the intrinsic lability of P-N bond under acidic conditions. Therefore, the effective N-phosphoproteome analysis remains challenging. Herein, bis(zinc(II)-dipicolylamine)-functionalized sub-2 μm core-shell silica microspheres (SiO2@DpaZn) are tailored for rapid and effective N-phosphopeptides enrichment. Due to the coordination of phosphate groups to Zn(II), N-phosphopep
