Pathogenesis of COVID-19 described through the lens of an undersulfated and degraded epithelial and endothelial glycocalyx.

The glycocalyx surrounds every eukaryotic cell and is a complex mesh of proteins and carbohydrates. It consists of proteoglycans with glycosaminoglycan side chains, which are highly sulfated under normal physiological conditions. The degree of sulfation and the position of the sulfate groups mainly determine biological function. The intact highly sulfated glycocalyx of the epithelium may repel severe acute respiratory syndrome-related coronavirus 2 (SARS-CoV-2) through electrostatic forces. However, if the glycocalyx is undersulfated and 3-O-sulfotransferase 3B (3OST-3B) is overexpressed, as i

Degradation of the Endothelial Glycocalyx Contributes to Metabolic Acidosis in Children Following Cardiopulmonary Bypass Surgery.

OBJECTIVES: Cardiopulmonary bypass surgery is complicated by metabolic acidosis, microvascular dysfunction, and capillary leak. The glycocalyx-a layer of proteins and sugars lining the vascular endothelium-is degraded during cardiopulmonary bypass. We aimed to describe the kinetics of glycocalyx degradation during and following cardiopulmonary bypass. We hypothesized that cleavage of negatively charged fragments of the glycocalyx would directly induce metabolic acidosis through changes in the strong ion gap (defined using Stewart's physicochemical approach to acid-base chemistry). We also inve

Desialylation of the rooster sperm's glycocalyx decreases sperm sequestration following intravaginal insemination of the hen.

Competitive fertilization was used to study sequestration of neuraminidase-treated sperm within the hen's sperm-storage tubules. The feather color inhibitor gene, I, was used to determine paternity of chicks hatched from eggs laid over a 12-day interval following a single intravaginal insemination. The insemination dose was 1 x 10(8) sperm per hen. The insemination of New Hampshire hens (i/i) with a 50:50 ratio of washed Brown (i/i) and washed White (I/I) Leghorn sperm yielded a 45:55 ratio of brown (i/i) to yellow (I/i) chicks. In contrast, a 14:86 ratio of brown to yellow chicks was obtained

The role of glycocalyx-protein corona interactions in the cell type-specific uptake of mRNA nanocarriers.

Non-viral nanocarriers for oligonucleotide delivery have enormous therapeutic potential, but the factors determining cell type-specific uptake remain incompletely understood. One major cell surface component that delivery vectors encounter is the glycocalyx. While membrane glycans are established interactors for many viruses, their impact on non-viral particles has remained underexplored. Here, we show that the role of membrane glycosaminoglycans (GAGs) in mRNA delivery differs across nanocarrier categories and cell types. We employed novel glycosylation inhibitors as well as enzymatic strateg

Early detection of glycocalyx and microvascular damage in suspected sepsis in the emergency department: the EDGE study.

BACKGROUND: The prompt identification of clinical deterioration in emergency department (ED) patients presenting with infection is crucial yet challenging. Microvascular dysfunction has been linked to poor clinical outcome in critically ill patients, but it remains unclear whether its detection can predict clinical deterioration in early sepsis. This study aims to evaluate the utility of quantitative microvascular videomicroscopy for predicting clinical deterioration in patients with suspected sepsis. METHODS: In this prospective observational study, 299 ED patients with suspected infection o

The Role of Cytokines in Vascular Endothelial Glycocalyx Integrity and Impairment Following Open-Heart Surgery.

Open-heart surgery with cardiopulmonary bypass (CPB) is a high-risk procedure with significant morbidity and mortality. CPB, tissue injury, blood loss, endotoxemia and ischemia-reperfusion injury induce a pronounced systemic inflammatory response, leading to endothelial glycocalyx (EG) damage and vascular endothelial dysfunction. Consequently, immune cells, reactive oxygen species, and enzymes gain free access to vascular endothelial cells, resulting in their dysfunction and enhancing inflammation, vascular permeability, and microvascular impairment. EG degradation is most commonly assessed by

Manuscript DMD-D-25-00346 The impact of glycocalyx on partitioning and distribution of basic drugs.

Concentrations of basic drugs are often overpredicted by physiologically based pharmacokinetic models at very early time points after intravenous dosing. An empirical shallow compartment in our recently published permeability- and perfusion-limited PermQ model improved the prediction of this observed early distribution. This study aimed to determine the impact of glycocalyx, a glycoprotein and glycolipid covering on membranes of red blood cells (RBCs) and endothelium, on the partitioning and distribution of basic drugs. Hyaluronidase (50 μg/mL), heparinase (5 U/mL), and neuraminidase (0.1 U/mL

Reduced syncytiotrophoblast-glycocalyx expression is associated with placental malperfusion and the severity of late-onset preeclampsia.

INTRODUCTION: The involvement of syncytiotrophoblast-glycocalyx (STB-GCX) in placental perfusion remains unclear. In this study, we investigated whether the STB-GCX is altered in preeclampsia (PE) and whether its expression correlates with clinical indicators of hypertension and reduced placental blood flow. METHODS: In a cohort of 61 pregnant women {late-onset PE(LOPE), n = 25; uncomplicated controls, n = 36}, placental perfusion was assessed by measuring the Flow Index (FI) using three-dimensional vocal power Doppler sonography to estimate placental blood flow. Maternal circulating levels o

Biomarkers of endothelial glycocalyx dysfunction in pregnancy: a systematic review of clinical relevance and detection techniques.

OBJECTIVE AND DESIGN: The endothelial glycocalyx (EG) is a key regulator of vascular homeostasis, acting as a dynamic barrier between the bloodstream and the endothelium. In pregnancy, structural and functional alterations of the EG have been increasingly implicated in the pathogenesis of endothelial dysfunction, particularly in preeclampsia and other vascular complications. This systematic review critically examines current evidence on circulating biomarkers of EG degradation and their clinical relevance in hypertensive and metabolic disorders of pregnancy. METHODS: We explore the mechanisti

Glycocalyx kinetics and injury during liver procurement and transplantation as predictors of early graft dysfunction.

INTRODUCTION: Ischemia-reperfusion injury causes endothelial damage, partly through degradation of the glycocalyx. This study aimed to evaluate glycocalyx degradation from graft procurement to reperfusion and assess its potential as a biomarker of early graft function (early allograft dysfunction, EAD). METHODS: This single-center observational prospective study was conducted at Paul Brousse Hospital from April 2022 to April 2023. All primary liver transplantation (LT) recipients were included. Glycocalyx degradation was assessed at procurement, at the end of cold ischemia, and during LT in l

Engineering Mechanical Microenvironments: Integration of Substrate and Flow Mechanics Reveals the Impact on the Endothelial Glycocalyx.

The glycocalyx (GCX), a multicomponent coating on endothelial cells (ECs), plays a critical role in various cellular behaviors, including barrier formation, vasodilation, and mechanotransduction. Mechanical perturbations in the vascular environment, such as blood vessel stiffness, are sensed and transduced by ECs via the GCX. Hypertension-induced stiffness disrupts GCX-mediated mechanotransduction, leading to EC dysfunction and atherosclerotic cardiovascular diseases. Understanding GCX-regulated mechanotransduction necessitates an in vitro model that closely mimics in vivo conditions. Existing

Comparing the effects of various fluid resuscitative strategies on Glycocalyx damage in a canine hemorrhage model.

Hemorrhagic shock and subsequent resuscitation can cause significant dysregulation of critical systems, including the vascular endothelium. Following hemorrhage, the endothelial lining (glycocalyx) can shed, causing release of glycocalyx components, endothelial activation, and systemic inflammation. A canine model of hemorrhagic shock was used to evaluate five resuscitation fluids, including Lactated Ringers+Hetastarch, Whole Blood (WB), Fresh Frozen Plasma+packed Red Blood Cells (FFP+pRBC), and two hemoglobin-based oxygen carrier (HBOC) fluids, for their impact on glycocalyx shedding. Sugar-binding profiles of the mesothelial glycocalyx in frozen tissues of mice revealed by lectin staining.

The mesothelium is a non-adhesive protective surface that lines the serosal cavities and organs within the body. The glycocalyx is a complex structure that coats the outer layer of the mesothelium. However, due to the limitations of conventional fixation techniques, studies on glycans are limited. In this study, lectin staining of frozen tissues was performed to investigate the diversity of glycans in the glycocalyx of mesothelial cells in mice. Datura stramonium lectin (DSL), which recognizes lactosamine and binds to Galectin-3 and -1, was broadly bound to the mesothelial cells of the viscera

Fast-track preparation of lung specimens for electron microscope observations of the pulmonary endothelial glycocalyx.

The glycocalyx (GCX) covers the luminal surface of blood vessels and regulates vascular permeability. As GCX degradation predicts various types of vasculopathy, confirming the presence of this structure is useful for diagnosis. Since the GCX layer is very fragile, careful fixation is necessary to preserve its structure. We explored appropriate and feasible methodologies for visualizing the GCX layer using lung tissue specimens excised from anesthetized mice. Each specimen was degassed and immersed in Alcian blue (ALB) fixative solution, and then observed using electron microscopy. Specimens fr

Endothelial Glycocalyx and Cardiomyocyte Damage Is Prevented by Recombinant Syndecan-1 in Acute Myocardial Infarction.

The outer layer of endothelial cells (ECs), consisting of the endothelial glycocalyx (eGC) and the cortex (CTX), provides a protective barrier against vascular diseases. Structural and functional impairments of their mechanical properties are recognized as hallmarks of endothelial dysfunction and can lead to cardiovascular events, such as acute myocardial infarction (AMI). This study investigated the effects of AMI on endothelial nanomechanics and function and the use of exogenous recombinant syndecan-1 (rSyn-1), a major component of the eGC, as recovering agent. ECs were exposed in vitro to s

Protective effect of restrictive resuscitation on vascular endothelial glycocalyx in pigs with traumatic hemorrhagic shock.

BACKGROUND: Hemorrhagic shock is the leading cause of early traumatic death. Research and discussion on restrictive fluid resuscitation have been ongoing for many years. The purpose of this study was to explore whether restrictive resuscitation can inhibit the shedding of vascular endothelial glycocalyx in the prehospital treatment of traumatic hemorrhagic shock pigs. METHODS: Landrace pigs were randomly divided into a restrictive resuscitation group (restrictive group) and a conventional resuscitation group (conventional group), with 6 pigs in each group. The gunshot caused a rupture of the

A Supraceliac Aortic Cross Clamping Model to Explore Remote Lung Injury and the Endothelial Glycocalyx.

BACKGROUND: We hypothesized that supraceliac aortic cross clamping could induce lung injury mediated by an inflammatory ischemia-reperfusion (IR) trigger. We aimed to characterize glycocalyx (GCX), a component of endothelial membrane, participating to remote lung injury. METHODS: Rats underwent supraceliac aortic cross clamping for 40 min and were sacrificed at 0, 3, 6, and 24 hr of reperfusion (n = 10/group). Each group was compared to sham (n = 6/group). GCX products (syndecan-1 [Sdc-1] and heparan sulfate [HS]), tumor necrosis factor-alpha (TNF-α), and interleukin-1β (IL-1β) were measured
