The problem of scale in the prediction and management of pathogen spillover.

Disease emergence events, epidemics and pandemics all underscore the need to predict zoonotic pathogen spillover. Because cross-species transmission is inherently hierarchical, involving processes that occur at varying levels of biological organization, such predictive efforts can be complicated by the many scales and vastness of data potentially required for forecasting. A wide range of approaches are currently used to forecast spillover risk (e.g. macroecology, pathogen discovery, surveillance of human populations, among others), each of which is bound within particular phylogenetic, spatial

Single-dose intranasal AdC68-vectored vaccines rapidly protect Syrian hamsters against lethal Nipah virus infection.

Nipah virus (NiV) infection is highly lethal in humans, and the development of vaccines that provide rapid protection is critical for addressing NiV outbreaks. In this study, we demonstrate that a single intranasal immunization with the chimpanzee adenoviral-vectored NiV vaccine, AdC68-F, induced robust and sustained cellular and humoral responses in BALB/c mice, and provided complete protection against challenge with the NiV-Malaysia strain (NiV-M) in Syrian hamsters. Notably, AdC68-F, administered at a dose of 5 × 109 viral particles, offered a complete prophylactic protection window as few

Foals of mares vaccinated for Hendra virus have a suboptimal response to HeV vaccination.

Hendra virus (HeV) is lethal to horses and a zoonotic threat to humans in Australia, causing severe neurological and/or respiratory disease with high mortality. An equine vaccine has been available since 2012. Foals acquire antibodies from their dams by ingesting colostrum after parturition, therefore it is assumed that foals of mares vaccinated against HeV will have passive HeV antibodies circulating during the first several months of life until they are actively vaccinated. However, no studies have yet examined passive or active immunity against HeV in foals. Here, we investigated anti-HeV a

Peptide-based vaccine design against Hendra virus through immunoinformatics approach.

The Hendra virus (HeV) has resulted in epidemics of respiratory and neurological illnesses in animals. Humans have contracted diseases with high fatality rates as a result of infected domestic animals, but effective vaccinations and therapies are currently not available against HeV. Herein, we analyzed the proteome of HeV and constructed an effective and innovative multi-epitope vaccine using immunoinformatics techniques. The vaccine construct was generated, targeting one matrix protein, with the help of the five selected B and T cell epitopes, linkers, and adjuvants and evaluated for their im

Comprehensive immunoinformatics and bioinformatics strategies for designing a multi-epitope based vaccine targeting structural proteins of Nipah virus.

BACKGROUND: Nipah virus (NiV) is characterized by recurring outbreaks and causes severe neurological impact, leading to increased mortality rates. Despite the severity of the disease, there is no proven post-exposure treatment available, emphasizing the critical need for the development of an effective vaccine. OBJECTIVE: This study was aimed at designing a multi-epitope based vaccine candidate based on an in-silico approach. METHODS: NiV's Structural proteins were screened for B and T-cell epitopes, assessing characteristics like antigenicity, immunogenicity, allergenicity, and toxicity. Tw

Evaluation of the immunogenicity of an mRNA vectored Nipah virus vaccine candidate in pigs.

Nipah virus (NiV) poses a significant threat to human and livestock populations across South and Southeast Asia. Vaccines are required to reduce the risk and impact of spillover infection events. Pigs can act as an intermediate amplifying host for NiV and, separately, provide a preclinical model for evaluating human vaccine candidate immunogenicity. The aim of this study was therefore to evaluate the immunogenicity of an mRNA vectored NiV vaccine candidate in pigs. Pigs were immunized twice with 100 μg nucleoside-modified mRNA vaccine encoding soluble G glycoprotein from the Malaysia strain of

Distinct respiratory and neurological disease phenotypes in lethal Nipah virus Malaysia versus Bangladesh infection in IFNAR knockout mice.

Nipah virus (NiV) strains Malaysia (NiV-M) and Bangladesh (NiV-B) cause severe respiratory and neurological disease in humans, yet no FDA-approved medical countermeasures currently exist. Since 2000, NiV-B has been responsible for most human outbreaks, underscoring the need for relevant animal models of this strain to support public health preparedness. While mice offer a scalable platform for high-throughput in vivo screening, susceptibility to NiV disease requires immunosuppression, and detailed clinical characterization, particularly for NiV-B, remains limited. Here, we comprehensively asse

Community willingness to participate in a Nipah vaccine trial in Bangladesh.

INTRODUCTION: Nipah virus (NiV) is a deadly zoonotic disease with pandemic potential, prioritized by the World Health Organization for research and vaccine development. Since Bangladesh has faced annual NiV outbreaks and repeated spillovers since 2001, it is likely to be the site of a Phase II vaccine trial. This study explored people's knowledge about NiV and their willingness to participate in a NiV vaccine trial in Bangladesh. METHODS: We conducted a mixed methods study, collecting qualitative and quantitative data from Mirpur township, Dhaka, and NiV-affected Faridpur District. From Decem

Tetherin Inhibits Nipah Virus but Not Ebola Virus Replication in Fruit Bat Cells.

Ebola virus (EBOV) and Nipah virus (NiV) infection of humans can cause fatal disease and constitutes a public health threat. In contrast, EBOV and NiV infection of fruit bats, the putative (EBOV) or proven (NiV) natural reservoir, is not associated with disease, and it is currently unknown how these animals control the virus. The human interferon (IFN)-stimulated antiviral effector protein tetherin (CD317, BST-2) blocks release of EBOV- and NiV-like particles from cells and is counteracted by the EBOV glycoprotein (GP). In contrast, it is unknown whether fruit bat tetherin restricts virus infe

Insurance and epidemics: SARS, West Nile virus and Nipah virus.

Severe acute respiratory syndrome (SARS) reminds us that sudden disease emergence is a permanent part of our world--and should be anticipated in our planning. Historically the emergence of new diseases has had little or no impact beyond a small, localized cluster of infections. However, given just the right conditions, a highly virulent pathogen can suddenly spread across time and space with massive consequences, as has occurred on several occasions in human history. In the wake of the SARS outbreak, we are now forced to confront the unpleasant fact that human activities are increasing the fre

From spillover to preparedness: a One Health analysis of knowledge gaps in Nipah virus surveillance, prevention, and health system response.

BACKGROUND: Nipah virus (NiV) is a highly pathogenic zoonotic henipavirus that continues to pose a significant threat to global health security because of its high case fatality rate, ability to cause human-to-human transmission, and absence of licensed vaccines or specific therapeutics. Although more than two decades of research have advanced the understanding of NiV ecology, epidemiology, and clinical disease, major uncertainties remain regarding spillover mechanisms, environmental drivers, surveillance integration, and outbreak preparedness. These persistent knowledge gaps limit the ability

Next-generation diagnostics: virus capture facilitates a sensitive viral diagnosis for epizootic and zoonotic pathogens including SARS-CoV-2.

BACKGROUND: The detection of pathogens in clinical and environmental samples using high-throughput sequencing (HTS) is often hampered by large amounts of background information, which is especially true for viruses with small genomes. Enormous sequencing depth can be necessary to compile sufficient information for identification of a certain pathogen. Generic HTS combining with in-solution capture enrichment can markedly increase the sensitivity for virus detection in complex diagnostic samples. METHODS: A virus panel based on the principle of biotinylated RNA baits was developed for specific

Sustained antibody response to a linear epitope of Nipah virus fusion protein in human survivor serum samples.

Nipah virus (NiV), a pathogen with pandemic potential, lacks approved treatments or vaccines, highlighting the urgent need for research on immune-targeted antigenic determinants. A significant gap persists in NiV research, as studies on the fusion (F) protein critical for viral entry as well as the B cells epitopes, have primarily focused on computational prediction rather than experimental validation of immunogenic epitopes obtained through immunoinformatics approach. This study focused on the conserved F protein across NiV human isolates and employed an immunoinformatics approach to predict

Establishment of a VSV-Based Pseudovirus Platform for In Vitro and In Vivo Evaluation of Nipah Vaccine-Induced Neutralizing Responses.

The Nipah virus (NiV) is a zoonotic pathogen characterized by high fatality rates and pandemic potential, whereby there is an urgent need for developing safe and effective vaccines. However, the evaluation of NiV vaccine-induced immunity is hindered by the requirement of Biosafety Level-4 (BSL-4) containment. In this study, we developed a recombinant vesicular stomatitis virus (rVSV)-based pseudovirus-expressing NiV fusion (F) and attachment (G) glycoproteins using a luciferase reporter gene for bioluminescence detection. This pseudovirus was optimized for production in BHK-21 (WI-2) cells, an

Griffithsin Inhibits Nipah Virus Entry and Fusion and Can Protect Syrian Golden Hamsters From Lethal Nipah Virus Challenge.

Nipah virus (NiV) is a highly pathogenic zoonotic paramyxovirus that causes fatal encephalitis and respiratory disease in humans. There is currently no approved therapeutic for human use against NiV infection. Griffithsin (GRFT) is high-mannose oligosaccharide binding lectin that has shown in vivo broad-spectrum activity against viruses, including severe acute respiratory syndrome coronavirus, human immunodeficiency virus 1, hepatitis C virus, and Japanese encephalitis virus. In this study, we evaluated the in vitro antiviral activities of GRFT and its synthetic trimeric tandemer (3mG) against

Transmission routes for nipah virus from Malaysia and Bangladesh.

Human infections with Nipah virus in Malaysia and Bangladesh are associated with markedly different patterns of transmission and pathogenicity. To compare the 2 strains, we conducted an in vivo study in which 2 groups of ferrets were oronasally exposed to either the Malaysia or Bangladesh strain of Nipah virus. Viral shedding and tissue tropism were compared between the 2 groups. Over the course of infection, significantly higher levels of viral RNA were recovered from oral secretions of ferrets infected with the Bangladesh strain. Higher levels of oral shedding of the Bangladesh strain of Nip

A mRNA vaccine encoding for a 60-mer Nipah virus G glycoprotein nanoparticle elicits a robust neutralizing antibodies response against the Nipah virus.

The Nipah virus is a zoonotic pathogen causing encephalitis and acute respiratory illness in humans with very high fatality rates. Here we report a novel messenger RNA vaccine that directly encodes for a nanoparticle displaying 60 head domains of the Nipah virus G (NiV G) glycoprotein that acts as a highly effective antigen. A vaccine encoding for the NiV G nanoparticle elicits high antibody titers against NiV G and a robust neutralizing antibody response with a pseudotyped Nipah virus system. We ultimately find that the proposed mRNA NiV G nanoparticle (mRNA NiV G-NP) provides a flexible plat

Nipah Virus C and W Proteins Contribute to Respiratory Disease in Ferrets.

UNLABELLED: Nipah virus (NiV) is a highly lethal paramyxovirus that recently emerged as a causative agent of febrile encephalitis and severe respiratory disease in humans. The ferret model has emerged as the preferred small-animal model with which to study NiV disease, but much is still unknown about the viral determinants of NiV pathogenesis, including the contribution of the C protein in ferrets. Additionally, studies have yet to examine the synergistic effects of the various P gene products on pathogenesis in animal models. Using recombinant NiVs (rNiVs), we examine the sole contribution of

A recombinant Hendra virus G glycoprotein subunit vaccine protects nonhuman primates against Hendra virus challenge.

UNLABELLED: Hendra virus (HeV) is a zoonotic emerging virus belonging to the family Paramyxoviridae. HeV causes severe and often fatal respiratory and/or neurologic disease in both animals and humans. Currently, there are no licensed vaccines or antiviral drugs approved for human use. A number of animal models have been developed for studying HeV infection, with the African green monkey (AGM) appearing to most faithfully reproduce the human disease. Here, we assessed the utility of a newly developed recombinant subunit vaccine based on the HeV attachment (G) glycoprotein in the AGM model. Four
