Cancer-associated molecular signature in the tissue samples of patients with cirrhosis.

Several types of aggressive cancers, including hepatocellular carcinoma (HCC), often arise as a multifocal primary tumor. This suggests a high rate of premalignant changes in noncancerous tissue before the formation of a solitary tumor. Examination of the messenger RNA expression profiles of tissue samples derived from patients with cirrhosis of various etiologies by complementary DNA (cDNA) microarray indicated that they can be grossly separated into two main groups. One group included hepatitis B and C virus infections, hemochromatosis, and Wilson's disease. The other group contained mainly

Functional effects of EpCAM N-glycosylation in MDA-MB-468 breast cancer cells.

EpCAM, the epithelial cell adhesion molecule, is a cancer cell marker whose expression is associated with worse prognosis in several epithelial cancers, including breast cancer. Despite being an attractive therapeutic target, it is unclear whether EpCAM plays a causative role in cancer progression and metastasis, and the mechanisms of this possible role remain unknown. To investigate EpCAM, we generated stable human MDA-MB-468 breast cancer cell lines with EpCAM knockout, EpCAM overexpression, or mutant unglycosylated EpCAM expression, in which all three normally N-glycosylated asparagine resi

Expression of cell adhesion molecules in tubulointerstitial nephritis associated with Sjögren's syndrome.

The tissue distribution of cellular adhesion molecules (ICAM-1, ELAM-1, VCAM-1) was studied in specimens from six normal human kidneys and in six biopsies from kidneys with tubulointerstitial nephritis associated with Sjögren's syndrome. In addition, the expression of cellular adhesion molecules was examined both in four renal biopsies from cases of tubulointerstitial nephritis of diverse pathogenesis and in six lip biopsies from cases of Sjögren's syndrome. ICAM-1 was expressed on vascular endothelial cells in normal kidneys, in all specimens of tubulointerstitial nephritis and in salivary gl

Targeting leukocyte migration and adhesion in Crohn's disease and ulcerative colitis.

Crohn's disease and ulcerative colitis are two chronic inflammatory bowel diseases. Current biologic therapies are limited to blocking tumor necrosis factor alpha. However, some patients are primary non-responders, experience a loss of response, intolerance or side effects defining the urgent unmet need for novel treatments. The rapid recruitment and inappropriate retention of leukocytes is a hallmark of chronic inflammation and a potentially promising therapeutic target. We discuss the immunological mechanisms of leukocyte homing and adhesion in the gut mucosa. The interaction of lymphocytes

Identification of an adhesion molecule expressed on adult T cell leukemia cells derived from a patient with gastrointestinal involvement: implication for a possible role of integrin beta 7 in leukemic cell infiltration into intestinal mucosa.

Patients with adult T cell leukemia (ATL) often manifest leukemic cell infiltration into various organs such as lung, liver, skin, and gut. To analyze the mechanism of intestinal infiltration of ATL cells, we made mAbs against ATL-43T, a human T cell line derived from an ATL patient with severe intestinal mucosal infiltration. One of the mAbs, named H920, was noted for a high and relatively specific reactivity with ATL-43T. Molecular cloning was done to identify this molecule and disclosed that the Ag molecule was identical to integrin beta 7. Since integrin beta 7 and its ligand MAdCAM-1 had

Everolimus ameliorates Helicobacter pylori infection-induced inflammation in gastric epithelial cells.

Источник пометил публикацию как отозванную

Helicobacter pylori (H.pylori) infection caused by gastric mucosal inflammation plays a pivotal role in the progression of gastric diseases. The recruitment and attachment of monocytes to the gastric mucosal epithelium are a major event in the early stages of H. pylori-associated gastric diseases. Everolimus is a mechanistic/mammalian target of rapamycin (mTOR) inhibitor used to prevent tumor growth by inhibiting the PI3K signaling pathway. Here, we examined the pharmacological role of Everolimus against H.pylori-induced damage in gastric epithelial cells. Firstly, we found that Everolimus ame

Loss of cutaneous delayed hypersensitivity reactions in nevus anemicus. Evidence for close concordance of cutaneous delayed hypersensitivity and endothelial E-selectin expression.

BACKGROUND: The relationship of adhesion molecules in the dermis to immunologically mediated cutaneous inflammation can be understood by focusing on a serendipitous phenomenon: a lack of dermatitis within the margins of a nevus anemicus (NA) in generalized contact dermatitis. The expression and induction of endothelial and epithelial adhesion molecules with intradermally injected cytokines were investigated. OBSERVATIONS: Nevus anemicus without dermatitis lacked histopathological changes consistent with inflammatory cellular infiltration. The surrounding skin of the dermatitic lesion expresse

Flow cytometry in the detection of circulating tumor cells in neoplastic effusions.

PURPOSE: Despite its limitations, the cytology of body fluids is widely used in diagnosing neoplastic cells. Flow cytometry detects and identifies individual cells, enabling the detection of circulating tumor cells and facilitating diagnosis. This study compared the diagnostic utility of flow cytometry and cytology for detecting cancer cells in peritoneal and pleural fluids. METHODOLOGY: We used flow cytometry and cytology to examine 119 pleural and peritoneal effusions received for routine screening. Antibodies against clusters of differentiation 45 (CD45), 14 (CD14), and Epithelial cell adh

ICAM2 initiates trans-blood-CSF barrier migration and stemness properties in leptomeningeal metastasis of triple-negative breast cancer.

Leptomeningeal metastasis (LM) occurs when tumor cells spread to the leptomeningeal space surrounding the brain and the spinal cord, thereby causing poor clinical outcomes. The triple-negative breast cancer (TNBC) has been associated with symptoms of LM and mechanism remained unclear. Through proteomic analysis, we identified high expression of ICAM2 in leptomeningeal metastatic TNBC cells, which promoted the colonization of the spinal cord and resulted in poor survival in vivo. Two-way demonstration indicated that high levels of ICAM2 promoted blood-cerebrospinal fluid barrier (BCB) adhesion,

Adhesion molecules in normal and pathological corneas. An immunohistochemical study using monoclonal antibodies.

BACKGROUND: Adhesion molecules are cell surface receptors that are probably important in various cell-cell and cell-extracellular matrix interactions of the cornea. METHOD: In this immunohistochemical light-microscopic study we analyzed the expression pattern of adhesion molecules in normal and pathological human corneas (cases of corneal inflammation and degenerative disorders). The analyzed molecules included the beta 1 integrin or VLA family VLA-1-6, the beta 2 integrins or leukocyte integrins LFA-1, Mac-1, and p150,95, the immunoglobulins LFA-3, CD2, ICAM-1 and VCAM-1 and the selectins EL

Characterization of the role for cadherin 6 in the regulation of human endometrial receptivity.

BACKGROUND: The endometrial luminal epithelium is the first point of attachment of embryos during implantation. Failure of embryos to firmly adhere results in implantation failure and infertility. A receptive endometrial luminal epithelium is achieved through the expression of adhesion molecules in the mid-secretory phase and is a requirement for implantation. Cadherin 6 (CDH6) is an adhesion molecule localizing to the endometrial luminal epithelial cell surface in the mid-secretory/receptive phase and knockdown of CDH6 in the Ishikawa cells (receptive endometrial epithelial cell line) comprom

Intrinsic cellular heterogeneity directs divergent epithelial-to-mesenchymal transition outcomes.

Epithelial-to-mesenchymal transition (EMT) is a fundamental process in development and disease; however, its study is often complicated by the intrinsic heterogeneity of commonly used epithelial cell models. Here, we show that pre-existing heterogeneity contributes to divergent outcomes following transforming growth factor-beta 3 (TGFβ3) treatment. Flow cytometry, gene expression profiling and imaging revealed persistent variability in E-cadherin (E-cad) and epithelial cell adhesion molecule (EpCAM) expression, with differentially sorted subpopulations displaying distinct transcriptional respo

Pan-cancer analysis reveals netrin-1 receptors as potential tumor biomarkers and immune-related therapeutic targets.

The signal pathways mediated by axon guiding molecule netrin-1 (NTN1) are transduced via its several membrane-bound receptors that include deleted in colorectal cancer (DCC), UNC5 (A-D), neogenin 1 (Neo1), melanoma cell adhesion molecule (MCAM), and Down syndrome cell adhesion molecule (DSCAM). Most of these genes play a role in the occurrence and progression of some solid tumors. Sufficient systematic studies have not been performed on the expression characteristics of the role of NTN1 and its receptors in the context of pan-cancer. Based on data from 10,437 subjects with 33 types of solid tu

Establishment and characterization of a new immortalized human adenomyosis epithelial-like cell line, tAEC21†.

Adenomyosis occurs when endometrial glands and stroma grow within the uterine myometrium. As a clinically significant disease, adenomyosis causes substantial pelvic pain and heavy menstrual bleeding. It remains understudied due to a lack of translational research tools and model systems. This study aimed to develop a telomerase-transformed, epithelial-like cell line derived from the eutopic endometrium of a subject with focal adenomyosis. De-identified endometrial tissue was processed through mechanical and enzymatic digestion. Epithelial and stromal populations were separated by selective adh

Differential loss of E-cadherin expression in infiltrating ductal and lobular breast carcinomas.

The epithelial-specific cell-cell adhesion molecule E-cadherin was analyzed immunohistochemically on tissue sections of 89 human primary infiltrating breast carcinomas, using monoclonal antibodies 6F9 (for cryostat sections) and 5H9 (for cryostat and paraffin sections). The tumors included 41 well and moderately differentiated infiltrating ductal carcinomas (IDCs) most of which (78%) showed strong linear staining at the cell borders at a level, as high as luminal cells of normal mammary glands. The 26 poorly differentiated, more highly malignant IDCs examined also were all positive for E-cadhe

Matriptase proteolysis selectively drives disassembly and internalization of the claudin-7 complex while preserving E-cadherin.

Claudin-7 is a basolaterally enriched tight junction protein that participates in membrane complexes with EpCAM and Trop-2, linking epithelial adhesion to barrier organization. Although proteolytic processing of EpCAM/Trop-2 has been proposed to regulate this complex, its functional consequences for claudin-7 remain unresolved. Here, we define the role of matriptase in regulating claudin-7 dynamics in HaCaT human keratinocytes. Under steady-state conditions, matriptase knockout had only modest effects on claudin-7 distribution, suggesting a limited role in initial complex formation. In contras

Mesothelial cells with mesenchymal features enhance peritoneal dissemination by forming a protumorigenic microenvironment.

Malignant ascites accompanied by peritoneal dissemination contain various factors and cell populations as well as cancer cells; however, how the tumor microenvironment is shaped in ascites remains unclear. Single-cell proteomic profiling and a comprehensive proteomic analysis are conducted to comprehensively characterize malignant ascites. Here, we find defects in immune effectors along with immunosuppressive cell accumulation in ascites of patients with gastric cancer (GC) and identify five distinct subpopulations of CD45(-)/EpCAM(-) cells. Mesothelial cells with mesenchymal features in CD45(

Powering single-cell genomics to unravel circulating tumour cell subpopulations in non-small cell lung cancer patients.

BACKGROUND: Circulating tumour cells (CTCs) are attractive "liquid biopsy" candidates that could provide insights into the different phenotypes of tumours present within a patient. The epithelial-to-mesenchymal transition (EMT) of CTCs is considered a critical step in tumour metastasis; however, it may confound traditional epithelial feature-based CTC isolation and detection. We applied single-cell copy number alteration (CNA) analysis for the identification of genomic alterations to confirm the neoplastic nature of circulating cells with only mesenchymal phenotypes. METHODS: We isolated CTCs

Immunocytological characterization of the expression of cell adhesion molecule L1 during early innervation of mouse otocysts.

Doubts exist as to whether afferent nerve fibers exert a neurotrophic effect on the differentiation of sensory cells in the developing vestibular neuroepithelium. To determine whether innervation of hair cells precedes their differentiation, we have used the L1 adhesion molecule as a marker for axons. The detection of L1 on afferent axons in the otic vesicle of mouse embryos on gestation day 11 shows that nerve fibers penetrate the neuroepithelium before the sensory cells differentiate. L1-immunoreactivity of nerve endings also reveals the considerable fiber ramification on gestation days 14 a

Protein kinase A-dependent phosphorylation of Lutheran/basal cell adhesion molecule glycoprotein regulates cell adhesion to laminin alpha5.

Lutheran (Lu) blood group and basal cell adhesion molecule (B-CAM) antigens reside on two glycoprotein (gp) isoforms Lu and Lu(v13) that belong to the Ig superfamily and differ only by the size of their cytoplasmic tail. Lu/B-CAM gps have been recognized as laminin alpha5 receptors on red blood cells and epithelial cells in multiple tissues. It has been shown that sickle red cells exhibit enhanced adhesion to laminin alpha5 when intracellular cAMP is up-regulated by physiological stimuli such as epinephrine and that this signaling pathway is protein kinase A- and Lu/B-CAM-dependent. In this st
