Synergistic metabolism of Limosilactobacillus fermentum and Acetobacter pasteurianus combined with nutrition supplementation regulating γ-aminobutyric acid biosynthesis in solid-state fermentation of cereal vinegar.

γ-aminobutyric acid (GABA) is one of the important functional compounds in vinegar. However, the underlying GABA biosynthesis mechanisms and regulating strategy in the solid-state fermentation of vinegar remain unclear. Limosilactobacillus fermentum was identified as key microorganism for producing glutamate decarboxylase (GAD). However, the metabolic activity of L.fermentum was limited due to high ethanol and acid stress, resulting in low GAD activity. Meanwhile, the temperature was positive with GABA, while ethanol was negative with it. Therefore, synergistic bioaugmentation combined with nu

Functions of the prp operon in carbon source metabolism and acetic acid resistance in acetic acid bacteria: a case study in Acetobacter pasteurianus CGMCC 1.41.

BACKGROUND: prp operon usually comprises prpB, prpC, prpD and prpE genes, encoding 2-methylisocitrate (2-MIC) lyase, 2-methylcitrate (2-MC) synthase, 2-MC dehydratase and propionyl-CoA synthetase, respectively, which constitute the pathway 2-methylcitrate cycle (2-MCC) well-known for microbial propionate metabolism. Acetic acid bacteria (AAB) represent a group of obligate aerobic and Gram-negative microorganisms. Attributed to the robust ethanol-oxidating and acetic acid-resisting abilities, vinegar production stands for an important AAB's industrial application. Previously, we unveiled the fi

Preparation and characterization of a novel bacterial cellulose film based photothermal sensitivity Zeolitic Imidazolate Framework and acetobacter xylinum and its application in pH monitoring and antibiosis.

Bacterial cellulose has been widely used as a raw material in the manufacture of medical wound dressings. However, its lack of inherent antibacterial properties and the susceptibility of its porous three-dimensional structure to bacterial colonization necessitate the adjunctive use of antimicrobial agents. Furthermore, the singular antibacterial function of the material falls short of meeting the multifunctional requirements of modern medical dressings. In response to these challenges, we have engineered a bacterial cellulose membrane imbued with rapid photothermal sterilization and pH fluores

Soluble branched (1,4)-beta-D-glucans from Acetobacter species enhance antitumor activities against MHC class I-negative and -positive malignant melanoma through augmented NK activity and cytotoxic T-cell response.

We previously found that an extracellular polysaccharide, AC-1, produced by Acetobacter polysaccharogenes composed of (1,4)-beta-D-glucan with branches of glucosyl residues showed a strong activity to induce production of interleukin (IL)-12 p40 and tumor necrosis factor-alpha by macrophage cell lines in vitro via Toll-like receptor-4 signaling. In the present study, we examined the effects of oral administration of AC-1 on protection against 2 types of murine B16 melanoma lines, major histocompatibility complex (MHC) class I-negative B16L and MHC class I gene-transfected B16K(b) cells. Spatiotemporal heterogeneity drives acetic acid fermentation of traditional Chinese aromatic vinegar: Insights into microbiota dynamics and flavor formation.

To address the ecological implications of spatiotemporal heterogeneity on microbial assembly and flavor formation in Chinese aromatic vinegar, this study systematically revealed these dynamics during the solid-state acetic acid fermentation (AAF). Temporally, Lactobacillus, Acetobacter, and Komagataeibacter synergistically drove acid and aroma formation. In the early stage (day 1), Lactobacillus dominated, creating an acidic niche for Acetobacter. During the main fermentation stage (days 7 to 13), Acetobacter oxidized ethanol to acetic acid, while Lactobacillus accumulated lactic acid, facilit

Diversity of acetic acid bacteria present in healthy grapes from the Canary Islands.

The identification of acetic acid bacteria (AAB) from sound grapes from the Canary Islands is reported in the present study. No direct recovery of bacteria was possible in the most commonly used medium, so microvinifications were performed on grapes from Tenerife, La Palma and Lanzarote islands. Up to 396 AAB were isolated from those microvinifications and identified by 16S rRNA gene sequencing and phylogenetic analysis. With this method, Acetobacter pasteurianus, Acetobacter tropicalis, Gluconobacter japonicus and Gluconacetobacter saccharivorans were identified. However, no discrimination be

Identification of Microbial Metabolites Elevating Vitamin Contents in Barley Seeds.

The current investigation analyzes metabolites of Acetobacter aceti to explore chemical compounds responsible for the induction of vitamins in barley seeds. A bioactivity guided assay of bacterial extracts and chromatographic analyses of barley produce revealed 13 chemical compounds, which were subjected to principal component analysis (PCA). PCA determined four chemical compounds (i.e., quinolinic acid, pyridoxic acid, p-aminobenzoate, and α-oxobutanoic acid) highly associated with increased quantities of vitamins. Further experimentations confirmed that quinolinic acid and p-aminobenzoate we

Changes in microbial communities during the spoilage of palm wine stored at room temperature.

This study was to determine the spoilage microbiota of palm wine stored at room temperature in order to define appropriate strategies for shelf-life extension. For this purpose, during the storage of palm wine samples, yeast, lactic acid bacteria (LAB), and acetic acid bacteria (AAB) counts were performed. Then, species were identified by PCR-RFLP followed by sequencing and the overall bacterial community was determined by NGS (Next Generation Sequencing). The results showed that yeast, LAB and AAB counts decreased in the palm wines collected from producers after 24-48 h, while they remained c

Regulation of gut microbiota of Astragali Radix in treating for chronic atrophic gastritis rats based on metabolomics coupled with 16S rRNA gene sequencing.

Astragali Radix (HQ), a common traditional Chinese medicine (TCM), is widely used to treat chronic atrophic gastritis (CAG). However, its mechanism in treating CAG is still not clear. Accumulating evidence highlights the link between gut microbiota and CAG. We hypothesized that the gut microbiota might be involved in the effect of HQ. Ultra-performance liquid chromatography coupled with time-of-flight mass spectrometry (UPLC-Q-TOF/MS) based metabolomics and 16S rRNA gene sequencing techniques of the cecal contents were applied to study its mechanisms. As a result, nine metabolites and fifteen

In this study, we used compound fungal bran koji to enhance the fermentation of Sichuan bran cupi. Analysis of the microbial composition of cupei showed that Aspergillus, Saccharomyces, Wickerhamomyces, Rhizopus, Lactobacillus, and Acetobacter are the primary genera involved in fermentation. The addition of enriched Qu (BQF) for intensive fermentation increased the relative abundance of Wickerhamomyces and Rhizopus during the initial and mid-stages of fermentation while influencing the dynamics of Aspergillus, Lactobacillus, Acetobacter, and other genera. Compared to the BQ group (control grou

Microbial functional differentiation and flavor formation across layer exchange in cereal vinegar fermentation: Multi-omics insights and targeted bioaugmentation.

Spatial heterogeneity is common in cereal vinegar fermentation, but microbial and metabolic changes across the layer-exchange process remain unclear. We combined metagenomics and metatranscriptomics with physicochemical, organic-acid, and volatile profiling to investigate layer-associated changes during Tianjin Duliu mature vinegar (TDMV) fermentation and guide bioaugmentation. Before exchange, the frequently turned upper layer consumed ethanol at 1.14 times the lower-layer rate, rapidly accumulated acetic acid, and was enriched in Acetobacter and Komagataeibacter. The lower layer retained mor

Tailor-made microbial consortium for Kombucha fermentation: Microbiota-induced biochemical changes and biofilm formation.

Kombucha is a very distinct naturally fermented sweetened tea that has been produced for thousands of years. Fermentation relies on metabolic activities of the complex autochthonous symbiotic microbiota embedded in a floating biofilm and used as a backslop for successive fermentations. Here, we designed a tailor-made microbial consortium representative of the core Kombucha microbiota to drive this fermentation. Microbial (counts, metagenetics), physico-chemical (pH, density) and biochemical (organic acids, volatile compounds) parameters were monitored as well as biofilm formation by confocal l

Dynamic changes of quality and flavor characterization of Zhejiang rosy vinegar during fermentation and aging based on untargeted metabolomics.

Zhejiang Rosy Vinegar (ZRV) is a traditional condiment in Southeast China. This study aimed to track the physicochemical, microbiological, sensory changes, and metabolomic profiles of ZRV during fermentation and aging. The increase of acidity and decrease of reducing sugar were associated with the dominant growth of Lactobacillus and Acetobacter. The total 35 volatile compounds were identified in ZRV, mainly containing alcohols, esters, acids, aldehydes, ketones acids, phenols and nitrogen-containing. Compared to phenethyl acetate with sweet aroma in fresh vinegar, the compound with high odor

Drosophila Gut Immune Pathway Suppresses Host Development-Promoting Effects of Acetic Acid Bacteria.

The physiology of most organisms, including Drosophila, is heavily influenced by their interactions with certain types of commensal bacteria. Acetobacter and Lactobacillus, two of the most representative Drosophila commensal bacteria, have stimulatory effects on host larval development and growth. However, how these effects are related to host immune activity remains largely unknown. Here, we show that the Drosophila development-promoting effects of commensal bacteria are suppressed by host immune activity. Mono-association of germ-free Drosophila larvae with Acetobacter pomorum stimulated lar

Characterization of Bacteria in Nigerian Yogurt as Promising Alternative to Antibiotics in Gastrointestinal Infections.

Gastrointestinal infections are endemic in Nigeria and several factors contribute to their continual survival, including bacterial resistance to commonly used antibiotics. Nigerian yogurts do not include probiotics, and limited information is available about the antimicrobial properties of the fermenters in the yogurt against gastrointestinal pathogens. Therefore, the antimicrobial potentials of bacteria in Nigeria-produced yogurts against intestinal pathogens were investigated in this study. Viable counts of lactic acid bacteria (LAB) in 15 brands of yogurt were enumerated and the bacteria id

Spontaneous organic cocoa bean box fermentations in Brazil are characterized by a restricted species diversity of lactic acid bacteria and acetic acid bacteria.

Spontaneous organic cocoa bean box fermentations were carried out on two different farms in Brazil. Physical parameters, microbial growth, bacterial species diversity [mainly lactic acid bacteria (LAB) and acetic acid bacteria (AAB)], and metabolite kinetics were monitored, and chocolates were produced from the fermented dry cocoa beans. The main end-products of the catabolism of the pulp substrates (glucose, fructose, and citric acid) by yeasts, LAB, and AAB were ethanol, lactic acid, mannitol, and/or acetic acid. Lactobacillus fermentum and Acetobacter pasteurianus were the predominating bac
