Aptamer and Peptide-Engineered Polydopamine Nanospheres for Target Delivery and Tumor Perfusion in Synergistic Chemo-Phototherapy of Pancreatic Cancer.

Pancreatic cancer (PC) is the fourth leading cause of cancer death, and the 5 year survival rate is only 4%. Chemotherapy is the treatment option for the majority of PC patients diagnosed at an advanced stage, whereas the desmoplastic stroma of PC could block the perfusion of chemotherapeutic agents to tumor tissues and contribute generally to chemoresistance. Therefore, the clinical status of PC calls for an urgent exploration in the effective treatment strategy. Chemo-phototherapy is an emerging modality against malignant tumors, but owing to the low targeting ability of theranostic agents o

Label-free peptide aptamer based impedimetric biosensor for highly sensitive detection of TNT with a ternary assembly layer.

We report a label-free peptide aptamer based biosensor for highly sensitive detection of TNT which was designed with a ternary assembly layer consisting of anti-TNT peptide aptamer (peptamer), dithiothreitol (DTT), and 6-mercaptohexanol (MCH), forming Au/peptamer-DTT/MCH. A linear relationship between the change in electron transfer resistance and the logarithm of the TNT concentration from 0.44 to 18.92 pM, with a detection limit of 0.15 pM, was obtained. In comparison, the detection limit of the aptasensor with a common binary assembly layer (Au/peptamer/MCH) was 0.15 nM. The remarkable impr

AAV-encoded CaV2.2 peptide aptamer CBD3A6K for primary sensory neuron-targeted treatment of established neuropathic pain.

Transmission of pain signals from primary sensory neurons to secondary neurons of the central nervous system is critically dependent on presynaptic voltage-gated calcium channels. Calcium channel-binding domain 3 (CBD3), derived from the collapsin response mediator protein 2 (CRMP2), is a peptide aptamer that is effective in blocking N-type voltage-gated calcium channel (CaV2.2) activity. We previously reported that recombinant adeno-associated virus (AAV)-mediated restricted expression of CBD3 affixed to enhanced green fluorescent protein (EGFP) in primary sensory neurons prevents the develop

Fluorogenic Peptide Aptamer Generated Using Bioorthogonal cDNA Display Technology for Cellular Imaging of Heat Shock Protein 90α.

Fluorogenic probes are valuable tools for rapid detection and bioimaging of target molecules without requiring bound/free separation. Although antibody-based probes are widely used, their large size, reliance on secondary labeling, and limited accessibility to some intracellular regions can restrict their utility. In contrast, fluorogenic peptide aptamers-compact, chemically synthesizable, and self-reporting-offer a promising alternative. Here, we developed a method to prepare a fluorophore-conjugated peptide library using cDNA display and applied it to the selection of fluorogenic peptides ta

Synthesis and characterization of miniaturized aptamer-based monolithic sorbent for selective extraction of β-amyloid peptides from cerebrospinal fluid.

The ratio between beta-amyloid (Aβ) peptides 40 and 42 is recognized as a biomarker for Alzheimer's disease, playing a significant role in early diagnosis and disease progression monitoring. Aβ peptides are present at trace levels in cerebrospinal fluid, therefore, developing a new selective extraction procedure is essential for isolating targeted biomarkers from the matrix interferents, ensuring accurate identification and quantification. In this study, a hybrid organic-silica monolith was synthesized in a 530 µm inner diameter-capillary and used for the covalent grafting of beta amyloid pept

Targeting Integrins in Triple Negative Breast Cancer: Therapeutic Potential of Monoclonal Antibodies, PARP Inhibitors, Synthetic Peptides, and Aptamers.

Triple-negative breast cancers (TNBCs) are associated with poor prognosis and chemoresistance. Recent reports indicate that extracellular matrix (ECM) molecules, such as integrins, could influence TNBC development. Integrins are adhesion macromolecules that modulate ECM characteristics such as stiffening of ECM and being associated with hypoxia. Therefore, dysregulation in integrins could compromise the efficient delivery of cancer treatments. Some peptides and aptamers can directly target integrins, and they are now being studied; however, most monoclonal antibodies and PARP inhibitors that a

Intrinsic SERS Fingerprints of Aptamer-Peptide Conjugates for Direct High-Specific Profiling Abnormal Protein Levels in Cancer Patients.

Surface-enhanced Raman spectroscopy (SERS) with ultrasensitive vibrational fingerprints enables quick identification and trace detection of various kinds of molecules. But proteins usually have low Raman cross sections and are difficult to generate recognizable signals in direct SERS detection. Recently, nucleic acid-peptide conjugates are emerging with great potential in structuring, assembling, catalyzing, sensing, etc., and the coupling of aptamers further enables superior biological recognition and programmability. Here, we develop the aptamer-peptide conjugates as a new kind of SERS probe

Primary sensory neuron-specific interference of TRPV1 signaling by AAV-encoded TRPV1 peptide aptamer attenuates neuropathic pain.

BACKGROUND: TRPV1 (transient receptor potential vanilloid subfamily member 1) is a pain signaling channel highly expressed in primary sensory neurons. Attempts for analgesia by systemic TRPV1 blockade produce undesirable side effects, such as hyperthermia and impaired heat pain sensation. One approach for TRPV1 analgesia is to target TRPV1 along the peripheral sensory pathway. RESULTS: For functional blockade of TRPV1 signaling, we constructed an adeno-associated virus (AAV) vector expressing a recombinant TRPV1 interfering peptide aptamer, derived from a 38mer tetrameric assembly domain (TAD

Immunoaffinity-Mimetic Assembly of Peptide-Aptamer Conjugates and Stem Cell-Derived Exosomes into Hierarchical Microgels for Spinal Cord Injury Repair.

Mesenchymal stem cell-derived exosomes (MSC-Exo) containing various paracrine mediators have recently been recognized to promote functional recovery of spinal cord injury (SCI). However, their biomedical applications are hindered by limited possibilities for specific integration within the delivery systems. In this study, inspired by antibody-antigen binding, we present an unprecedented assembly strategy that leverages immunoaffinity-mimetic interactions of peptide-aptamer (Peptide-AptCD63) conjugates targeting CD63 markers on MSC-Exo, yielding a hierarchical microstructure to enhance SCI repa

Enhanced Label-Free Nanoplasmonic Cytokine Detection in SARS-CoV-2 Induced Inflammation Using Rationally Designed Peptide Aptamer.

Rapid and precise serum cytokine quantification provides immense clinical significance in monitoring the immune status of patients in rapidly evolving infectious/inflammatory disorders, examplified by the ongoing severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) pandemic. However, real-time information on predictive cytokine biomarkers to guide targetable immune pathways in pathogenic inflammation is critically lacking, because of the insufficient detection range and detection limit in current label-free cytokine immunoassays. In this work, we report a highly sensitive localized sur

Antimicrobial Peptide LL-37 Facilitates Intracellular Uptake of RNA Aptamer Apt 21-2 Without Inducing an Inflammatory or Interferon Response.

RNA aptamers are synthetic single stranded RNA oligonucleotides that function analogously to antibodies. Recently, they have shown promise for use in treating inflammatory skin disease as, unlike antibody-based biologics, they are able to enter the skin following topical administration. However, it is important to understand the inflammatory milieu into which aptamers are delivered, as numerous immune-modulating mediators will be present at abnormal levels. LL-37 is an important immune-modifying protein upregulated in several inflammatory skin conditions, including psoriasis, rosacea and eczem

Antimicrobial and therapeutic effect of gold Nanoparticle-Aptamer conjugated antimicrobial peptide RW-BP100 in Brucella canis infected mice and RAW 264.7 cells.

Canine brucellosis is a common zoonosis worldwide, but treatment is challenging due to the stealthy and chronic infection caused by the bacterium Brucella (B.) canis. Antimicrobial peptides (AMPs) are promising candidates for combating drug-resistant microbes, and gold nanoparticle aptamer (AuNP-Apt) aids the delivery of peptides to mammalian cells. This study evaluated the therapeutic efficacy of AuNP-AptHis conjugated antimicrobial peptide RW-BP100His (AuNP-AptHis-RW-BP100His) against B. canis in murine macrophage RAW 264.7 cells and BALB/c mice. In vitro, a high concentration of RW-BP100His

Pharmacokinetics, pharmacodynamics and safety of aptamers.

Aptamers are synthetic molecules structured as single-stranded DNA or RNA oligonucleotides that can be designed to mimic the functional properties of monoclonal antibodies. They bind to the target molecules (typically soluble or cell-bound proteins) with high affinity (with picomolar to low nanomolar range) and specificity, and therefore can be an alternative to therapeutic antibodies or peptide ligands. This paper reviews published data regarding pharmacokinetics, pharmacodynamics and safety of aptamers from preclinical and clinical studies. Aptamers have been developed for the treatment of a

An integrated microfluidic platform to perform uninterrupted SELEX cycles to screen affinity reagents specific to cardiovascular biomarkers.

As cardiovascular diseases (CVD) are responsible for millions of deaths annually, there is a need for rapid and sensitive diagnosis of CVD at earlier stages. Aptamers generated by systematic evolution of ligands by exponential enrichment (SELEX) processes have been shown to be superior to conventional antibody-based cardiac biomarker detection. However, SELEX is a complicated, lengthy procedure requiring multiple rounds of extraction/amplification and well-trained personnel. To circumvent such issue, we designed an automated, miniaturized SELEX platform for the screening of aptamers towards th

Discovery of the High-Affinity Aptamer for Candidalysin Using a Dual-Mode Colorimetric-SERS Platform.

Candida albicans poses significant health risks through its virulent peptide toxin Candidalysin. As no existing therapeutics specifically target this toxin, developing high-affinity aptamers for its efficient and safe removal is urgently needed. In response, we developed a dual-mode biosensor based on gold nanoparticles (AuNPs) and aptamers for screening high-affinity aptamers for Candidalysin. This biosensor leverages the localized surface plasmon resonance (LSPR) phenomenon and surface-enhanced Raman scattering (SERS) of AuNPs to detect changes in color and Raman signals, respectively, indic

The Evaluation of Pharmacodynamics and Pharmacokinetics of Anti-thrombin DNA Aptamer RA-36.

Anticoagulants are a vital class of drugs, which are applied for short-term surgical procedures, and for long-term treatments for thrombosis prevention in high risk groups. Several anticoagulant drugs are commercially available, but all have intrinsic disadvantages, e.g., bleeding risks, as well as specific ones, e.g., immune response to peptide/protein drugs. Therefore, the search for novel, efficient and safe anticoagulants is essential. Nucleic acid aptamers are an emerging class of contemporary pharmaceuticals which are fully biocompatible and biodegradable; they have low toxicity, and are

Noncompetitive affinity assays of glucagon and amylin using mirror-image aptamers as affinity probes.

The ability to detect picomolar concentrations of glucagon and amylin using fluorescently labeled mirror-image aptamers, so-called Spiegelmers, is demonstrated. Spiegelmers rival the specificity of antibodies and overcome the problem of biostability of natural aptamers in a biological matrix. Using Spiegelmers as affinity probes, noncompetitive capillary electrophoresis affinity assays of glucagon and murine amylin were developed and optimized. The detection limit for glucagon was 6 pM and for amylin was 40 pM. Glucagon-like peptide-1 and -2 did not interfere with the glucagon assay, while the

Development and characterization of high-affinity aptamers for HIV protease detection.

BACKGROUND: There are 39 million people infected by the human immunodeficiency virus (HIV) and 1.3 million new annually infections with more than 150 variants circulating. Early HIV detection is crucial for timely antiretroviral therapy and transmission prevention, but no technique can detect HIV before 10 days of infection. Aptamers, single-stranded oligonucleotides, can recognize specific proteins with high affinity and specificity. New molecular assays based on aptamers selected against highly-conserved peptides in HIV proteins could help to detect the virus earlier. RESULTS: We have selec

DNA aptamers from whole-serum SELEX as new diagnostic agents against gastric cancer.

Gastric cancer is still among the leading causes of cancer deaths worldwide. Despite the improvements in diagnostic methods, the status of early detection has not been achieved so far. Early diagnosis of gastric cancer may significantly improve the cure rate of patients. Therefore, a new diagnostic method is needed. In this study, subtractive SELEX was performed to screen gastric cancer serum-specific DNA aptamers by using gastric cancer serum and normal serum as the target and negative serum, respectively. Four highly specific aptamers generated for gastric cancer serum, Seq-3, Seq-6, Seq-19
