Melleolide sesquiterpene aryl esters are secondary products of the mushroom genus Armillaria. We compared the cytotoxicity of eleven melleolides--five thereof are new natural products--against four human cancer cell lines. Armillaridin, 4-O-methylarmillaridin, and dehydroarmillylorsellinate were most active, at IC(50) = 3.0, 4.1 and 5.0 μM, respectively, against Jurkat T cells for the former two compounds, and K-562 cells for the latter. Dehydroarmillylorsellinate did not inhibit respiration and RNA-synthesis of K-562 cells at 5 μM. However, replication of DNA dropped to 35% after 120 min at t

Microscope slide preparations made for the observation of the mycorrhiza of the mycoheterotrophic orchid Gastrodia elata were rediscovered and confirmed to be the original materials used by Shunsuke Kusano for his 1911 publication. In this study, we re-examined these historical slides to assess their preservation and scientific significance. Among the 115 slides, 61 clearly displayed the mycorrhiza of G. elata. Intracellular organelles closely resembling the 'vesicle', 'globule', 'tubular sheath', 'globular bodies', and 'lignified papillae' described by Kusano were observed, and staining react

BACKGROUND: Gastrodia elata, known as a rootless, leafless, achlorophyllous and fully mycoheterotrophic orchid, needs to establish symbionts with particular Armillaria species to acquire nutrition and energy. Previous research findings had approved that ethylene (ET) played an important role in plant-fungi interaction and some receptors of ET had been discovered in microorganisms. However, the molecular mechanisms underlying the role of ET in the interaction between G. elata and Armillaria species remain unknown.
METHODS: Exiguous ethephon (ETH) was added to agar and liquid media to observe t

Currently, "medicinal food" is a trending topic of discussion given the various bioactive compounds found within edible mushrooms. However, research on the collection and biological potency of saponins extracted from hazel mushroom, a species of Armillaria, is lacking. We aimed to conduct additional studies with respect to our previous endeavor, which was an effort to determine possible methods that can be used to alleviate ultrasonic-assisted extraction conditions of compounds and determine possible anticancer effects. Response surface methodology can be used to identify good extraction param

A new polysaccharide fraction (ATP) was obtained from Armillariella tabescens mycelium. Structural analysis suggested that the backbone of ATP was →4)-α-D-Glcp(1 → 2)-α-D-Galp(1 → 2)-α-D-Glcp(1 → 4)-α-D-Glcp(1→, which branched at O-3 of →2)-α-D-Glcp(1 → and terminated with T-α-D-Glcp or T-α-D-Manp. Besides, ATP significantly alleviated ulcerative colitis (UC) symptoms and inhibited the production of pro-inflammation cytokines (IL-1β, IL-6). Meanwhile, ATP could improve colon tissue damage by elevating the expression of MUC2 and tight junction proteins (ZO-1, occludin and claudin-1) levels and

Nitrilases participate in the nitrile metabolism in microbes and plants. They are widely used to produce carboxylic acids from nitriles. Nitrilases were described in bacteria, Ascomycota and plants. However, they remain unexplored in Basidiomycota. Yet more than 200 putative nitrilases are found in this division via GenBank. The majority of them occur in the subdivision Agaricomycotina. In this work, we analyzed their sequences and classified them into phylogenetic clades. Members of clade 1 (61 proteins) and 2 (25 proteins) are similar to plant nitrilases and nitrilases from Ascomycota, respe

Armillaridin (AM) is an aromatic ester compound isolated from Armillaria mellea. Treatment with AM markedly reduced the viability of human chronic myelogenous leukemia K562, chronic erythroleukemia HEL 92.1.7, and acute monoblastic leukemia U937 cells, but not normal human monocytes, in a dose- and time-dependent manner. Treatment of K562 cells with AM caused changes characteristic of autophagy. Only a small amount of AM-treated K562 cells exhibited apoptosis. By contrast, AM treatment resulted in extensive apoptotic features in U937 and HEL 92.1.7 cells without evident autophagy. The autophag

Medicinal mushrooms have been essential components of traditional Chinese herbal medicines for thousands of years, and they protect against diverse health-related conditions. The components responsible for their anti-inflammatory activity have yet to be fully studied. This study investigates the anti-inflammatory activity of n-hexane, chloroform, ethyl acetate, and methanol extracts of mycelia in submerged culture from 5 commercially available medicinal mushrooms, namely Cephalosporium sinensis, Cordyceps mortierella, Hericium erinaceus, Ganoderma lucidum, and Armillaria mellea. OBJECTIVES: The growth, development, and accumulation of active compounds in Gastrodia elata are influenced by its mycorrhizosphere bacteria, which indirectly enhances host nutrition by stimulating the growth of the orchid mycorrhizal fungus Armillaria gallica. However, data on the mycorrhizosphere bacterial community of G. elata, especially on key functional strains, remain limited, restricting their application in medicinal plant cultivation. Here, we report the complete genome sequence of the plant growth-promoting bacterium Rahnella aceris HPDA25, which was isolated from the mycorrhizosphe

The Gastrodia elata Blume (GE) life cycle is unique, since its successful germination and growth rely on symbiosis with specific fungi (e.g., Armillaria mellea). However, the community succession, tissue specificity and functional potential of endophytic and rhizosphere bacterial communities during the seed formation stage of GE remain unclear. Here, we used high-throughput 16S rRNA gene sequencing to systematically explore the composition, diversity, and dynamic succession of bacterial communities across different stages of seed formation and among various tissues. Our results revealed that t

BACKGROUND: Fungal communities play crucial roles in plant development and metabolite accumulation, especially in fully mycoheterotrophic medicinal plants like Gastrodia elata. While the importance of fungal symbiosis in G. elata is recognized, how fungal community dynamics evolve across its entire growth cycle and how they influence biomass and bioactive compound accumulation remain largely unclear.
RESULTS: High-throughput sequencing combined with multi-omics analyses revealed that developmental progression significantly shapes fungal diversity and composition, thereby influencing biomass a

Neuroinflammation is a prevalent pathological characteristic of numerous neurodegenerative disorders. Inhibition of neuroinflammation can slow the progression of these diseases. Armillariella tabescens is a valuable medicinal fungus that is often used in traditional Chinese medicine to treat acute and chronic hepatitis, appendicitis, otitis media and cholecystitis. Its mycelium polysaccharide (ATMP) has excellent biological activities, including anti-inflammatory, antioxidation and anti-aging. However, the mechanism of ATMP against neuroinflammation has not been elucidated. This study aimed to

BACKGROUND: Fully mycoheterotrophic orchids depend entirely on fungal partners for carbon and mineral nutrition, yet the structural mechanisms that regulate fungal accommodation within host tissues remain poorly understood. This question is particularly relevant in the Gastrodia elata-Armillaria symbiosis, where the fungal partner is a wood-decaying basidiomycete capable of aggressive host colonization and responsible for root rot disease in many tree species. Here, we examined whether host cell wall remodeling and extensin deposition contribute to the establishment and regulation of the host-

This study was performed to evaluate effects of Armillariella tabescens (A. tabescens) on the growth performance and intestinal immune response and microflora in early-weaned pigs when used as feed additive. A. tabescens mycelia were added to basal diets at concentrations of 0%, 0.1%, 0.3% or 0.9% (w/w). A total of 144 commercial cross-bred piglets were randomly allocated to one of these four diets and fed for 30 days. The growth performance of early-weaned piglets displayed improvement with diets containing 0.1% and 0.3% dried mycelia powder from A. tabescens. Supplementing with 0.1% or 0.3%

Fruiting bodies (sporocarps, sporophores or basidiomata) of mushroom-forming fungi (Agaricomycetes) are among the most complex structures produced by fungi. Unlike vegetative hyphae, fruiting bodies grow determinately and follow a genetically encoded developmental program that orchestrates their growth, tissue differentiation and sexual sporulation. In spite of more than a century of research, our understanding of the molecular details of fruiting body morphogenesis is still limited and a general synthesis on the genetics of this complex process is lacking. In this paper, we aim at a comprehen

Mushrooms are valued for their nutritional qualities and have been used in traditional medicine since the Neolithic era. They exhibit various bioactivities, including antioxidant, hypocholesterolemic, immunomodulatory, and anticancer effects. The anticancer effects arise via direct action on tumor cells and indirect modulation of the immune system; the latter is the predominant mechanism. Numerous studies indicate that various mushroom species are potent immunostimulants because their cell wall polysaccharides and proteoglycans are recognized by intestinal immune cells. This enhances antitumor
