Base barrier cells provide compartmentalization of choroid plexus, brain and CSF.

The choroid plexus (ChP), located in the brain ventricles, is largely composed of ChP epithelial cells that produce the cerebrospinal fluid (CSF) and form the blood-CSF barrier. At the ChP-brain attachment sites, we have discovered unique fibroblasts referred to as ChP base barrier cells (BBCs). We show that ChP BBCs originate from meningeal mesenchymal precursors, arrive early during development, remain throughout life and are conserved across species. ChP BBCs are transcriptionally similar to meningeal arachnoid barrier cells and are interconnected by both adherens and tight junctions. Notab

Compartmentalized Nutrient and Hormone Sensing in β-Cells: the Role of Primary Cilia.

Pancreatic β-cells couple nutrient and hormone sensing to insulin release. Prevailing models depict the β-cell as a homogeneous metabolic unit, overlooking specialized subcellular compartments like the primary cilium. Primary cilia are sensory organelles that function as critical signaling hubs in pancreatic β-cells. Disrupting primary cilia causes diabetes, but the underlying mechanisms remain unclear. In this review, we critically assess evidence supporting the cilium's role in β-cells, including functional studies that implicate cilia in the control of cytosolic Ca2+ dynamics and exocytosis

Hepatitis B virus compartmentalization and single-cell differentiation in hepatocellular carcinoma.

Chronic hepatitis B virus (HBV) infection is a major cause of hepatocellular carcinoma (HCC) world-wide. The molecular mechanisms of viral hepatocarcinogenesis are still partially understood. Here, we applied two complementary single-cell RNA-sequencing protocols to investigate HBV-HCC host cell interactions at the single cell level of patient-derived HCC. Computational analyses revealed a marked HCC heterogeneity with a robust and significant correlation between HBV reads and cancer cell differentiation. Viral reads significantly correlated with the expression of HBV-dependency factors such a

Mechanisms of compartmental purkinje cell death and survival in the lurcher mutant mouse.

The Lurcher mutant mouse is characterized by its ataxic gait and loss of cerebellar Purkinje cells and their afferents, granule cells and olivary neurons, during the first weeks of postnatal development. For the 50 years since its discovery, the heterozygous Lurcher mutant has served as an important model system for studying neuron-target interactions in the developing cerebellum and cerebellar function. The identification of the Lurcher (Lc) gene over 10 years ago as a gain-of-function mutation in the δ2 glutamate receptor (GluRδ2) led to extensive studies of cell death mechanisms in the Lc/+

Compartmentalization of dendritic cell and T-cell interactions in the lymph node: Anatomy of T-cell fate decisions.

Upon receiving cognate and co-stimulatory priming signals from antigen (Ag)-presenting dendritic cells (DCs) in secondary lymphoid tissues, naïve CD4+ T cells differentiate into distinct effector and memory populations. These alternate cell fate decisions, which ultimately control the T-cell functional attributes, are dictated by programming signals provided by Ag-bearing DCs and by other cells that are present in the microenvironment in which T-cell priming occurs. We know that DCs can be subdivided into multiple populations and that the various DC subsets exhibit differential capacities to i

Defining compartmentalized stem cell populations with distinct cell division dynamics in the ocular surface epithelium.

Adult tissues contain label-retaining cells (LRCs), which are relatively slow-cycling and considered to represent a property of tissue stem cells (SCs). In the ocular surface epithelium, LRCs are present in the limbus and conjunctival fornix; however, the character of these LRCs remains unclear, owing to lack of appropriate molecular markers. Using three CreER transgenic mouse lines, we demonstrate that the ocular surface epithelium accommodates spatially distinct populations with different cell division dynamics. In the limbus, long-lived Slc1a3CreER-labeled SCs either migrate centripetally t

Aqueous Two-Phase Bioinks for Discrete Packing and Compartmentalization of 3D Bioprinted Cells.

The unparalleled ability of aqueous two-phase systems (ATPS) to reproduce microscale cellular and biomaterial compartmentalization to selectively modulate cell behavior and functionality is ideal for tissue engineering and regenerative medicine (TERM) purposes. Herein, we introduce new ATPS biomaterial inks for 3D bioprinting of water-in-water (W/W) emulsions, enabling precise cellular crowding for tissue regeneration in vitro and ex vivo. Gelatin methacryloyl (GelMA) was dispersed in an alginic acid phase depending on sodium chloride (NaCl) concentration (0-36 g/L). Emulsion droplet size (12.

Compartmentalized B cell-microglia crosstalk drives ischemic white matter demyelination.

Ischemic white matter injury and the consequent neuroimmune response can contribute to vascular cognitive impairment. The role of B cells in ischemic white matter injury is unclear. Here, we show that crosstalk between meningeal B cells and CNS-resident microglia exacerbates white matter injury and vascular cognitive impairment. The B cell population expands and is activated in the dura mater, correlating with worsened white matter damage and neuroinflammation, whereas B cell depletion alleviates myelin thinning and cognitive impairment following chronic cerebral hypoperfusion in adult male mi

Non-allometric expansion and enhanced compartmentalization of Purkinje cell dendrites in the human cerebellum.

Purkinje cell (PC) dendrites are optimized to integrate the vast cerebellar input array and drive the sole cortical output. PCs are classically seen as stereotypical computational units, yet mouse PCs are morphologically diverse and those with multi-branched structure can receive non-canonical climbing fiber (CF) multi-innervation that confers independent compartment-specific signaling. While otherwise uncharacterized, human PCs are universally multi-branched. Do they exceed allometry to achieve enhanced integrative capacities relative to mouse PCs? To answer this, we used several comparative

Compartmentalization of adenosine metabolism in cancer cells and its modulation during acute hypoxia.

Extracellular adenosine mediates diverse anti-inflammatory, angiogenic and vasoactive effects, and has become an important therapeutic target for cancer, which has been translated into clinical trials. This study was designed to comprehensively assess adenosine metabolism in prostate and breast cancer cells. We identified cellular adenosine turnover as a complex cascade, comprising (1) the ectoenzymatic breakdown of ATP via sequential ecto-nucleotide pyrophosphatase/phosphodiesterase-1 (NPP1, officially known as ENPP1), ecto-5'-nucleotidase (CD73, also known as NT5E), and adenosine deaminase r

Compartmentalization of inflammatory cell phenotypes in normal gingiva and peri-implant keratinized mucosa.

The interpretation of studies aimed at understanding the pathophysiology of periodontal breakdown has been hampered by an insufficient understanding of the physiology of host responses in clinically healthy gingiva. This investigation was aimed at the evaluation of the in situ phenotype and topographic distribution of the inflammatory cells in clinically normal gingiva and peri-implant keratinized mucosa (PIKM). Soft tissue biopsies were obtained from clinically healthy gingiva or PIKM in 14 patients. Acetone fixed, cryostat sections were stained with a panel of monoclonal antibodies with a th

Citrate Compartmentalization Controls Calcium-Dependent Cytokine Production in Effector T Cells.

Cytokine production is a core function of effector T cells, yet the mechanisms that regulate cytokine output during an immune response remain incompletely understood. Here, we identify citrate compartmentalization as a cellular mechanism by which CD8 + T cells couple cytokine production to glucose availability. Under glucose-replete conditions, citrate transport from the mitochondria to the cytosol by the citrate carrier SLC25A1 suppresses calcium-dependent transcription factor activity in effector T cells. Either reducing glucose availability or blocking the exchange of citrate across the mit

Cell-matrix interactions drive functional compartmentalization of mitochondria during 3D migration.

Cells migrating through three-dimensional (3D) tissues adapt their mechanical properties in response to extracellular matrix architecture through migratory plasticity. In primary human dermal fibroblasts, matrix elasticity drives distinct low- and high-pressure migration modes in which forces either push or pull the nucleus, respectively. How these mechanically distinct modes of nuclear translocation influence mitochondrial organization and function is not known. Here, we show that mitochondria become enriched anterior to the nucleus during 3D migration and segregate into spatially distinct po

Differential compartmentalization of myeloid cell phenotypes and responses towards the CNS in Alzheimer's disease.

Myeloid cells are suggested as an important player in Alzheimer´s disease (AD). However, its continuum of phenotypic and functional changes across different body compartments and their use as a biomarker in AD remains elusive. Here, we perform multiple state-of-the-art analyses to phenotypically and metabolically characterize immune cells between peripheral blood (n = 117), cerebrospinal fluid (CSF, n = 117), choroid plexus (CP, n = 13) and brain parenchyma (n = 13). We find that CSF cells increase expression of markers involved in inflammation, phagocytosis, and metabolism. Changes in phenoty

Spatial compartmentalization of melanoma cell states reveals CAF-associated tumor cells with enhanced proliferative capacity.

BACKGROUND: Cancer-associated fibroblasts (CAFs) are key components of the melanoma tumor microenvironment and have been implicated in immune evasion and therapy resistance. However, the spatial relationships between CAFs, extracellular matrix (ECM) remodeling, and immune exclusion remain incompletely characterized. METHODS: We integrated single-cell RNA sequencing (scRNA-seq) and spatial transcriptomics from primary melanoma samples (892 regions of interest) to investigate the landscape of CAF-immune interactions. CAF scores were calculated based on five fibroblast-associated genes. Differen

Compartmentalized vascular-mimetic glioma spheroid-on-a-chip for modeling endothelial-stromal cell recruitment.

Glioma progression is closely associated with vascular-related microenvironmental cues, but conventional in vitro models often lack spatial control over tumor spheroids, extracellular matrices, endothelial cells, and stromal support cells. Here, we developed a compartmentalized vascular-mimetic glioma spheroid-on-a-chip for visualizing glioma-vascular-associated cell interactions in a three-dimensional hydrogel microenvironment. The device integrates a central hydrogel channel for pre-formed glioma spheroids, two adjacent medium channels for human umbilical vein endothelial cells (HUVECs) and

Single-cell transcriptomics reveals a compartmentalized antiviral interferon response in the nasal epithelium of mice.

Type III interferons (IFNs) primarily act on epithelial cells and protect against virus infection of the mucosa, whereas type I IFNs act more systemically. To date, it has been unknown which epithelial subtypes in the upper airways, the primary site for initial infection for most respiratory viruses, primarily rely on type III IFN or type I IFNs for antiviral protection. To address this question, we performed a single-cell transcriptomics analysis of the epithelial IFN-mediated response focusing on the upper airways of mice. This work identified nine distinct cell types derived from the olfact

Compartmental surgery for squamous cell carcinoma of the buccal mucosa: description of a new surgical technique.

BACKGROUND: Oral squamous cell carcinoma (OSCC) is the sixth most common cancer globally. Patient survival varies depending on tumour stage and oral cavity subsites. Buccal mucosa neoplasia is rare and burdened by worse prognosis than other oral subsites, showing a high rate of loco-regional relapses within six months after treatment. According to NCCN guidelines, the gold standard treatment is radical surgery. In the oral cavity, the buccal mucosa subsite lacks anatomical barriers opposing neoplastic growth. At this level, the tumour cells could hypothetically spread along the fibres of the p
