Spatiotemporal regulation of microRNA-mediated gene silencing in cells.

MicroRNAs (miRNAs) are small non-coding RNAs that regulate gene expression by silencing mRNAs with complementary sequences. Over the past few decades, genetic and biochemical studies have revealed the molecular machinery and mechanisms underlying miRNA function. However, the current understanding of their cellular biology remains relatively limited. While the conventional view simplifies the miRNA journey as transcription in the nucleus and function in the cytoplasm, the precise spatiotemporal regulation of miRNAs within the intracellular environment is only beginning to emerge. Impact of Paracoccin Gene Silencing on Paracoccidioides brasiliensis Virulence.

Among the endemic deep mycoses in Latin America, paracoccidioidomycosis (PCM), caused by thermodimorphic fungi of the Paracoccidioides genus, is a major cause of morbidity. Disease development and its manifestations are associated with both host and fungal factors. Concerning the latter, several recent studies have employed the methodology of gene modulation in P. brasiliensis using antisense RNA (AsRNA) and Agrobacterium tumefaciens-mediated transformation (ATMT) to identify proteins that influence fungus virulence. Our previous observations suggested that paracoccin (PCN), a multidomain fung

[Gene silencing approaches for the treatment of Huntington's disease].

Huntington's disease is a rare neurodegenerative disease caused by a pathologic CAG expansion in the exon 1 of the huntingtin (HTT) gene. Aggregation and abnormal function of the mutant HTT (mHTT) cause motor, cognitive and psychiatric symptoms in patients, which lead to death in 15-20 years. Currently, there is no treatment for HD. Experimental approaches based on drug, cell or gene therapy are developed and reach progressively to the clinic. Among them, mHTT silencing using small non-coding nucleic acids display important physiopathological benefit in HD experimental models.

Chitosan hydrogel as siRNA vector for prolonged gene silencing.

BACKGROUND: The periodontitis is one of the most prevalent diseases with alveolar resorption in adult people and is the main cause of the tooth loss. To investigate the possibility for protecting the loss of alveolar bone in periodontal diseases, a RNAi-based therapeutic strategy is applied for silencing RANK signaling using thermosensitive chitosan hydrogel as siRNA reservoir and vector. RESULTS: The thermosensitive chitosan hydrogel was formed from solution (PH = 7.2, at 4°C) at 37°C within 8 minutes. The degradation rates of hydrogel were ~50% and 5% (W remaining/W beginning) in the presen

Temporal control of gene silencing by in ovo electroporation.

The analysis of gene function during embryonic development asks for tight temporal control of gene expression. Classic genetic tools do not allow for this, as the absence of a gene during the early stages of development will preclude its functional analysis during later stages. In contrast, RNAi technology allows one to achieve temporal control of gene silencing especially when used with oviparous animal models. In contrast to mammals, reptiles and birds are easily accessible during embryonic development. We have developed approaches to use RNAi for the analysis of gene function during nervous

NFkappaB gene silencing inhibits wear particles-induced inflammatory osteolysis.

Wear particles-induced periprosthetic osteolysis involved in proinflammatory cytokine production and osteoclastogenesis, is the major cause of prosthetic join implant loosening. Recent advances in our understanding of cellular and molecular mechanisms of periprosthetic osteolysis have highlighted cytokine release and osteoclast function controlled by numerous intracellular signaling pathway, one of which is nuclear factor kappa B (NFkappaB). Direct inhibition of NFkappaB is an efficient therapy to block bone erosion associated with inflammatory arthritis. There are no approved nonoperative tre

Bcl-2 gene silencing in pediatric epithelial liver tumors.

BACKGROUND: Proteins of the Bcl-2 family prevent cells of various tumor types from undergoing apoptosis and thus contribute to their chemotherapy resistance. The phenotype of multidrug resistance is a major factor for poor treatment results of advanced epithelial liver tumors in children. The role of Bcl-2 proteins in these tumors is yet unclear. The purpose of this study was to analyze the influence of Bcl-2 on the chemotherapy resistance of hepatoblastoma (HB) and pediatric hepatocellular carcinoma (HCC). MATERIALS AND METHODS: Bcl-2 expression was analyzed in the HB cell lines HUH6 and Hep

Promoter-Targeting RNA Technologies: An Epigenetic Strategy for Gene Activation and Gene Silencing.

Promoter-targeted RNAs (PTRs) are emerging as a transformative approach for future therapeutics, offering the ability to directly modulate gene expression at the transcriptional level. Unlike traditional RNA-based methods that work post transcriptionally, PTRs interact with gene promoter regions to either activate or suppress gene transcription. This strategy offers a key advantage by enabling more stable and long-lasting control of gene expression, since it acts directly at the level of chromatin and epigenetic regulation rather than only degrading RNA or blocking translation. Unlike CRISPR-b

DNA methylation-mediated extracellular matrix gene silencing in colorectal cancer.

Epigenetic alterations, particularly DNA methylation, play a crucial role in colorectal cancer (CRC) development and progression. We aimed to investigate DNA methylation changes across normal colon samples, adenomas, and adenocarcinomas to identify critical gene networks associated with disease progression. We conducted a comprehensive DNA methylation analysis on 2,346 samples. We extracted CRC methylation data of normal colon mucosa, adenomas, and adenocarcinomas from the genomic data commons data portal and the gene expression omnibus. We ranked differentially methylated genes by their area

Control Strategies of Plant Viruses Using Spray-Induced Gene Silencing.

Plant viruses severely limit global crop productivity, yet no pesticide-like antiviral agents are currently available for effective control. Double-stranded RNA (dsRNA) technologies, acting through RNA interference, provide a sequence-specific and non-transgenic strategy to suppress viral replication and have emerged as promising non-transgenic solutions for crop protection. Spray-induced gene silencing (SIGS), which applies externally produced dsRNA through foliar sprays, seed treatments, or root uptake, provides practical advantages over genetic modification, including rapid deployment, envi

Application of Virus-Induced Gene Silencing to Arbuscular Mycorrhizal Fungi.

Downregulation of AM fungal genes using a plant viral vector is feasible. A partial sequence of a target gene is cloned into the multicloning site of the CMV2-A1 vector developed from RNA2 of Cucumber mosaic virus Y strain, and the RNA2, together with RNA1 and RNA3 of the virus, are in vitro-transcribed. Inoculation of Nicotiana benthamiana with these viral RNAs results in reconstitution of the virus in the plant, which triggers silencing of the fungal gene. Here, we describe not only the methods but also several tips for the successful application of virus-induced gene silencing to AM fungi.

Modified pullulan unlocks efficient polyplex-mediated gene silencing following nebulization.

Pullulan (PULL) has emerged as a promising natural material to produce carriers for pulmonary siRNA delivery. In this study, controlled acidic hydrolysis was employed to tailor PULL molecular weight, reducing it from ~405 kDa to ~45 kDa. The resulting polymer was functionalized with 1,2-bis(3-aminopropylamino)ethane (bAPAE) to obtain a cationic copolymer (DD ≈ 33 mol%) and further modified with all-trans-retinoic acid (Ret) (DD ≈ 4.3 mol%) to introduce hydrophobic component. Potentiometric titration of PULL-bAPAE revealed pKa values of 4.73, 7.00, and 9.31, indicating suitable buffering capaci

Targeted gene silencing in the nervous system with CRISPR-Cas13.

Cas13 nucleases are a class of programmable RNA-targeting CRISPR effector proteins that are capable of silencing target gene expression in mammalian cells. Here, we demonstrate that RfxCas13d, a Cas13 ortholog with favorable characteristics to other family members, can be delivered to the mouse spinal cord and brain to silence neurodegeneration-associated genes. Intrathecally delivering an adeno-associated virus vector encoding an RfxCas13d variant programmed to target superoxide dismutase 1 (SOD1), a protein whose mutation can cause amyotrophic lateral sclerosis, reduced SOD1 mRNA and protein

Gene silencing of HIF-2α disrupts glioblastoma stem cell phenotype.

AIM: Improved treatment strategies are desperately needed for eradicating cancer stem cells (CSCs), which drive malignancy and recurrence in glioblastoma multiforme. Hypoxic regions within the tumor microenvironment help maintain and promote the proliferation of CSCs. Here, we explored the effects of silencing hypoxia inducible factor-2α (HIF-2α) because of its specificity for CSCs within the hypoxic environment. METHODS: Cancer stem cell neurospheres were formed by enriching from both the glioblastoma cell line U87 and from brain tumor stem cells isolated directly from human brain tumors. Si

Multi-targeted gene silencing strategies inhibit replication of Canine morbillivirus.

BACKGROUND: Canine morbilivirus (canine distemper virus, CDV) is a highly contagious pathogen associated with high morbidity and mortality in susceptible carnivores. Although there are CDV vaccines available, the disease poses a huge threat to dogs and wildlife hosts due to vaccine failures and lack of effective treatment. Thus, the development of therapeutics is an urgent need to achieve rapid outbreak control and reduce mortality in target species. Gene silencing by RNA interference has emerged as a promising therapeutic approach against different human and animal viruses. In this study, pla

Gene methylation and silencing of SOCS3 in mantle cell lymphoma.

The significance of loss of SOCS3, a negative regulator of signalling pathways including those of STAT3 and NF-κB, was examined in mantle cell lymphoma (MCL). The protein expression and gene methylation status of SOCS3 were detected using immunohistochemistry/Western blots and methylation-specific polymerase chain reaction, respectively. To evaluate its functional importance, SOCS3 was restored in two SOCS3-negative MCL cell lines using a lentiviral vector. Loss of SOCS3 protein expression was found in 3/4 MCL cell lines and 18/33 (54.5%) tumours. SOCS3 was found consistently methylated in cel

Methylation silencing of the Apaf-1 gene in acute leukemia.

Apaf-1 is important for tumor suppression and drug resistance because it plays a central role in DNA damage-induced apoptosis. Inactivation of the Apaf-1 gene is implicated in disease progression and chemoresistance of some malignancies. In this study, we attempted to clarify the role of Apaf-1 in leukemogenesis. Apaf-1 mRNA levels were below the detection limit or very low in 5 of 20 human leukemia cell lines (25%) and 5 of 12 primary acute myeloblastic leukemia cells (42%). There were no gross structural abnormalities in the Apaf-1 gene in these samples. Expression of factors regulating Apaf

Gene silencing of the tyrosine phosphatase SHP1 gene by aberrant methylation in leukemias/lymphomas.

High-frequent silencing of hematopoietic cell-specific protein-tyrosine phosphatase SHP1 gene by promoter methylation was detected in various kinds of leukemias and lymphomas, as well as in many hematopoietic cell lines, which is supported by our previous observation of strong decrease of SHP1 mRNA and protein. The promoter methylation of the SHP1 gene was clearly correlated with the clinical stage. Loss of heterozygosity with microsatellite markers near the SHP1 gene was shown in 79% of informative acute lymphoblastic leukemia cases. These results suggest that functional loss of SHP1 is assoc

Cell factories that manufacture microvesicles containing gene silencing RNA prodrugs.

Among the vehicles investigated as delivery agents for antisense RNAs are extracellular vesicles (EVs) released from cultured cells. Arrestin domain-containing 1 (ARRDC1)-mediated microvesicles (ARMMs) are naturally occurring EVs that uniquely are formed by outward budding of cytoplasmic membranes. Previous work has shown that ARMMs production is quantitatively controlled by the ARRDC1 protein and that ARRDC1 and macromolecules attached to it are loaded into nascent ARMMs during ARMM formation. Here, we report a strategy for constructing cell factories that biologically manufacture both ARMMs

TGFBR1 gene silencing attenuates cardiomyopathy in the HFpEF mouse model.

Myocardial fibrosis is a principal factor in the progression of heart failure with preserved ejection fraction (HFpEF). Prior research has substantiated the profibrotic impact of transforming growth factor β1. Nevertheless, the function of the transforming growth factor beta receptor (TGFBR) in HFpEF remains uncertain. In this study, we found an increase in myocardial TGFBR1 expression in mice with HFpEF. Silencing of the TGFBR1 gene improved cardiac function in HFpEF mice by attenuating cardiac fibrosis, reducing myocardial hypertrophy, and ameliorating myocardial remodeling. At the mechanist
