Platycodon D promotes immunogenic cell death in lung cancer cells by targeting NFS1 to induce PANoptosis.

BACKGROUND: Lung cancer remains one of the most lethal malignancies worldwide, with therapeutic resistance and low response rates representing major clinical challenges. The biosynthetic enzyme NFS1 has been implicated in tumour progression across various cancer types. PANoptosis is a recently identified type of cell death that can increase antitumour immunity, resulting in effective tumour suppression. Platycodin D (PD), a triterpenoid saponin isolated from the traditional Chinese medicine Platycodon grandiflorus, has demonstrated anticancer effects through mechanisms such as apoptosis induct

Improvement of pneumonia by curcumin-loaded bionanosystems based on platycodon grandiflorum polysaccharides via calming cytokine storm.

Pneumonia can lead to high morbidity and mortality secondary to uncontrolled inflammation of the lung tissue. Blocking cytokine storm storms may be the key to saving the life of patients with severe pneumonia. According to the medicinal guide theory of Traditional Chinese Medicine (TCM) and the inherent affinity with macrophages for the site of inflammation, we constructed the drug delivery platform (MNPs) derived from macrophage-membrane encapsulated reaction oxygen species (ROS)-responsive Platycodon grandiflorum polysaccharides (PGP) nanoparticles (PNPs) to calm the cytokine storm and impro

Clinical and Cellular Evidence of Glycyrrhiza glabra and Platycodon grandiflorus for Vocal Fold Nodules Complementary Treatment.

Vocal fold nodules (VFNs) are the most frequent cause of hoarseness. The management comprised medical, surgical and physical therapy but the effectiveness is not always satisfactory. In this study, we try to figure out an alternative treatment from our clinical experience summary. We retrospectively reviewed VFNs patients who received traditional Chinese medicine (TCM) treatments from July 2018 to August 2020 and traced their Chinese Voice Handicap Index-10 (VHI-C10) and multidimensional voice program (MDVP) analysis results. For further evaluation, we conducted an inflammatory response of por

Structural characterization and gut microbiota-mediated immunomodulatory effects of a novel fructose enriched polysaccharide from Platycodon grandiflorus.

This paper characterized PGP90, a novel low-molecular-weight (∼3.1 kDa) inulin-type fructan (α-D-Glcp-(1 → [1)-β-D-Fruf-(2]₁₈) isolated from Platycodon grandiflorus (P. grandiflorus), and demonstrated its potent immunomodulatory effects in CTX-induced immunosuppressed mice. Structural and cell cytotoxicity analysis confirmed the composition of PGP90 with less side effects. Oral administration of PGP90 (100 or 200 mg/kg) significantly restored CTX-suppressed immune parameters, including body weight, spleen weight, spleen index, splenic histoarchitecture, and the distribution of CD3+ T and CD19+

Platycodon grandiflorus-Derived Nanovesicles Sensitize Gastric Cancer to T Cell Killing via Ferroptosis-Driven Membrane Mechanical Remodeling.

Plant-derived nanovesicles have emerged as promising natural nanotherapeutics for cancer treatment, yet whether they can overcome the intrinsic resistance of tumor cells to cytotoxic T-cell-mediated killing remains largely unexplored. Here, we show that lipid-enriched Platycodon grandiflorus-derived nanovesicles (PGNs) sensitize gastric cancer to immune attack through ferroptosis-driven plasma membrane mechanical remodeling. Mechanistically, PGNs reprogram tumor lipid composition by coordinately suppressing the SLC7A11-GPX4 antioxidant axis while delivering polyunsaturated fatty acids, particu

Revealing the "Yin-Jing" mystery veil of Platycodon grandiflorum by potentiating therapeutic effects and lung-oriented guidance property.

UNLABELLED: ETHNIC PHARMACOLOGICAL RELEVANCE: "Yin-Jing" medicine (YJM) has been widely used by both ancient and modern Chinese medicine practitioners during long-term clinical practice. However, it remains unclear how to best guide other medicines to the targeted organs in a traditional Chinese medicine (TCM) prescription. Here, in an attempt to explain the scientific connotation of the YJM property (YJMP) attributed to a basic TCM theory, Platycodon grandiflorum (PG) was chosen as a case study to reveal the mystery of YJMP theory. AIM OF THE STUDY: The main purpose of this study is to emplo

Effect of ultrasonic-assisted H2O2/ascorbic acid modification on Platycodon grandiflorum polysaccharide: structural characterization and in vitro functional activities.

To improve the bioavailability of Platycodon grandiflorum polysaccharides (PGS), an ultrasound-assisted hydrogen peroxide/ascorbic acid (H2O2/Vc) modification method was developed. PGS was first extracted using an ultrasound-assisted deep eutectic solvent (DES) method, followed by synergistic H2O2/Vc treatment under ultrasound for 60, 120, and 180 min to obtain three modified fractions, namely DPGS-60, DPGS-120, and DPGS-180. Structural changes in PGS and its modified fractions were characterized by gel permeation chromatography (GPC), high-performance liquid chromatography (HPLC), Fourier tra

Structure Characterization and Antibacterial, Antifungal, and Antiquorum-Sensing Activity of New Metabolites From the Lipophilic Fractions of Platycodon grandiflorum Root.

The root of Platycodon grandiflorum has long been used as a vegetable and traditional medicine. Although the antibacterial activity of the plant's lipophilic (hexanes and dichloromethane) fractions has been reported, the specific antibacterial compounds have not been identified. In this study, chemical analysis of the lipophilic fractions of P. grandiflorum extracts led to the discovery of five new polyacetylenes (1-5) and nine known compounds (6-14). Their structures were elucidated and confirmed based on 1D and 2D NMR data together with mass spectra. In particular, the relative and absolute
