Cutaneous Mycobacterioses of Cats and Dogs.

Worldwide, a variety of mycobacterial species have been associated with skin lesions in dogs and cats. Lesions may result from systemic dissemination or local cutaneous inoculation. Only infections with Mycobacterium tuberculosis complex organisms have the potential to be transmitted from companion animals to humans, but even then, zoonotic risk is considered low. Diagnosis of mycobacterial infections is based on identification of intralesional acid-fast bacteria using cytology or histopathology, culture at experienced facilities, and use of PCR-sequencing. Treatment typically requires prolong

Recent Advances in the Detection of Antibiotic and Multi-Drug Resistant Salmonella: An Update.

Antibiotic and multi-drug resistant (MDR) Salmonella poses a significant threat to public health due to its ability to colonize animals (cold and warm-blooded) and contaminate freshwater supplies. Monitoring antibiotic resistant Salmonella is traditionally costly, involving the application of phenotypic and genotypic tests over several days. However, with the introduction of cheaper semi-automated devices in the last decade, strain detection and identification times have significantly fallen. This, in turn, has led to efficiently regulated food production systems and further reductions in food

Phylogenomic and comparative genomic analyses of Aeromonas spp. from South American aquatic systems reveal extensive genomic diversity, antimicrobial resistance, and predicted human pathogenicity.

INTRODUCTION: Aquaculture systems function as dynamic ecological interfaces facilitating the circulation of opportunistic pathogens and antimicrobial resistance (AMR) determinants across environmental, animal, and human compartments. Aeromonas spp. are ubiquitous freshwater bacteria and emerging human opportunistic pathogens, yet genomic data from tropical South American aquaculture remain scarce. METHODS: Nine Aeromonas isolates from Amazonian aquaculture facilities in Peru were subjected to whole-genome sequencing. Species identification was performed using Average Nucleotide Identity and p

Environmental resistome of culturable gram-negative bacilli from the Yaque del Norte River: baseline genomic evidence for one health surveillance.

Antimicrobial resistance (AMR) represents a significant global public health threat. Aquatic ecosystems are increasingly recognized as critical reservoirs of resistance determinants within the One Health framework. Freshwater systems, in particular, facilitate the persistence and dissemination of antimicrobial resistance genes (ARGs) at the human-animal-environment interface. This study characterized the environmental resistome of culturable Gram-negative bacilli isolated from the Yaque del Norte River, the largest river in the Dominican Republic. Water samples were collected along the river b

The zebrafish embryo as a potential 3R-compliant model to identify endocrine activity across estrogenic, androgenic and steroidogenic (EAS) modalities.

Endocrine disrupting compounds (EDCs) are substances that can interfere with hormonal systems, posing risks to wildlife and human health. For wildlife, a substance must exhibit endocrine activity at a relevant target (receptor or enzyme) and, as a direct consequence, cause population-relevant adverse effects in the exposed organism or its offspring to be classified as an EDC. Current regulatory frameworks utilize various in vivo assays to identify potential EDCs related to estrogenic, androgenic and steroidogenesis (EAS) modalities. However, ethical concerns have generated increasing momentum

Comparative lipidomic profiling of African trypanosomes reveals metabolic distinctions impacting pan-species drug development.

African trypanosomes are a group of pathogens that cause disease in humans (human African trypanosomiasis; HAT) and animals (African animal trypanosomosis; AAT) across sub-Saharan Africa, leading to profound impacts on human health as well as economic instability. Achieving sustainable disease control requires a One Health approach, yet the development of urgently required pan-species chemotherapeutics is severely hindered by a lack of unified biological understanding of the causative species. A major bottleneck in drug discovery is the distinct in vitro requirements of the clinically relevant

Combined administration of cocaine and alcohol alters the expression of brain peptide/protein profiles in rats: a MALDI imaging mass spectrometry approach.

While the concurrent use of alcohol and cocaine is common in patterns of polysubstance use, little is known about the combined effects of these substances on the brain. Proteomics approaches enable the identification of potential biomarkers and new pharmacological targets for the diagnosis and treatment of addiction and related psychiatric disorders. The main goal of this study was to explore how the combination of cocaine and alcohol affects brain peptide/protein signatures in the mesolimbic dopaminergic pathway. To this end, we used a chronic and simultaneous intravenous administration of th

Identification and quantification of the sea spray effect on isotopic systems in α-cellulose (δ13C, δ18O), total sulfur (δ34S), and 87Sr/86Sr of European beach grass (Ammophila arenaria, L.) in a greenhouse experiment.

The sea spray effect can severely influence the isotopic signature of terrestrial individuals in coastal regions. To further specify this effect, beach grass was grown in a greenhouse under controlled environmental conditions and sprayed with mineral salt solution containing different mineral salts but only traces of NaCl (group 1). Another group of plants was sprayed with salty water from the Schlei inlet and the Baltic Sea, respectively (group 2). Control plants were only sprayed with tap water. Isotope analyses were conducted on the unwashed and washed plants (δ13Ccellulose, δ18Ocellulose,

Sequence-based detection and typing procedures for Burkholderia mallei: Assessment and prospects.

Although glanders has been eradicated in most of the developed world, the disease still persists in various countries such as Brazil, India, Pakistan, Bangladesh, Nepal, Iran, Bahrain, UAE and Turkey. It is one of the notifiable diseases listed by the World Organization for Animal Health. Occurrence of glanders imposes restriction on equestrian events and restricts equine movement, thus causing economic losses to equine industry. The genetic diversity and global distribution of the causing agent, Burkholderia (B.) mallei, have not been assessed in detail and are complicated by the high clonali

Standardization criteria for an ischemic surgical model of acute hepatic failure in pigs.

The establishment of a reliable large animal model of acute hepatic failure (AHF) is critical for the evaluation of supportive therapies such as bioartificial liver support systems (BALSS). Large animal surgical models bear some resemblance to the clinical syndrome of AHF in humans. However, these are inherently complex and are predisposed to inter-individual variation. The development of considerable skill is required and no precise standardization criteria for such models have been defined as yet. This study investigates a surgically induced ischemic model of AHF in 15 female Landrace pigs.

Scientific knowledge about gene expression in ruminants under heat stress - A scientometric review.

Heat stress can alter the expression of genes in the individual's molecular response. The identification of these genes makes it possible to better understand the molecular response, identifying biomarker genes and indirect response pathways that can help with genetic improvement studies, animal welfare, separating more thermotolerant varieties and mitigating the effects of heat stress. The aim of this scientometric review was to characterize the state of the art of scientific research into gene expression in ruminants under heat stress, to define the most studied species, biology systems and

High-quality, large-scale, semi-thin, & ultra-thin sections of the optic nerve in large animals: An optimized procedure.

Large animal model of optic nerve (ON) injury is an essential tool for translational medicine. Perfusion fixation with paraformaldehyde is mainly used for preparing the semi-thin (1-2 μm thick) and ultra-thin (<0.5 μm thick) sections of the ON tissues. However, this conventional fixation technique in large animals needs a large volume of fixatives, which increases the risk of toxic exposure and is environmentally unfriendly. Additionally, fixed residual ON cannot be used for other tests that require fresh tissue samples. Although conventional immersion fixation is feasible for preparing a semi

Vasculature submucosal changes at early stages of osteonecrosis of the jaw (ONJ).

Osteonecrosis of the jaw (ONJ), a rare, but potentially severe side effect of anti-resorptive medications, presents as exposed bone in the maxillofacial region lasting for at least 8 weeks. While clinical experience and animal models concur in finding that systemic antiresorptive treatment in conjunction with local risk factors, such as tooth extraction or dental disease may lead to ONJ development, the subclinical molecular changes that precede bone exposure remain poorly understood. The identification of these changes is not only important in understanding disease pathophysiology, but could

Co-occurrence of resistance genes to antibiotics, biocides and metals reveals novel insights into their co-selection potential.

BACKGROUND: Antibacterial biocides and metals can co-select for antibiotic resistance when bacteria harbour resistance or tolerance genes towards both types of compounds. Despite numerous case studies, systematic and quantitative data on co-occurrence of such genes on plasmids and chromosomes is lacking, as is knowledge on environments and bacterial taxa that tend to carry resistance genes to such compounds. This effectively prevents identification of risk scenarios. Therefore, we aimed to identify general patterns for which biocide/metal resistance genes (BMRGs) and antibiotic resistance gene

[Immunology of contact allergy].

Contact allergy is a skin disease that is caused by the reaction of the immune system to low molecular weight chemicals. A hallmark of contact allergens is their chemical reactivity, which is not exhibited by toxic irritants. Covalent binding of contact allergens to or complex formation with proteins is essential for the activation of the immune system. As a consequence antigenic epitopes are formed, which are recognized by contact allergen-specific T cells. The generation of effector and memory T cells causes the high antigen specificity and the repeated antigen-specific skin reaction of cont

Finding causative genes underlying rheumatic disease: could NCF1 pave the way?

The identification of genetic risk factors could facilitate our understanding of the cause and pathogenesis of rheumatic diseases. Maps of susceptibility loci have been established for most complex diseases, including all major rheumatic diseases, but definitive causal variants have been identified only very infrequently. Here, we highlight the need to both position and confirm the role of causative polymorphisms in humans using experimental animals. The approach is best exemplified by the positional cloning of a causative single-nucleotide polymorphism (SNP) (rs201802280), which results in th

[Sound frequency analysis for identification of venous air embolism].

Air emboli occurring during surgery are considered to be life-threatening incidents. With the aim of achieving acoustic identification of venous air emboli, a frequency analysis of the sounds induced by an air embolism (millwheel murmurs) as well as of all other unusual sounds, was undertaken during 20 operations, and in experiments with animals. The frequency spectra of the sounds induced by air emboli are characterised by an increase in the amplitudes in the frequency range 1,100 to 3,000 Hz, while the amplitudes of normal heart sounds continuously decrease with increasing frequency. The fre

Tear biomarkers in veterinary species: An updated review with special focus on dogs and cats.

The tear film is traditionally described as a highly organized, multilayer structure comprising an outer lipid layer, a central aqueous phase, and an inner mucin layer, each contributing distinct structural and functional properties. Beyond its essential roles in maintaining ocular surface homeostasis, tear fluid represents an integrated biological secretion shaped by systemic physiological regulation that might be used as a source of biomarkers. While tear fluid has long been recognized in human medicine as a valuable reservoir of biomarkers, its diagnostic and translational potential in vete

Rapid Detection and Identification of Yersinia pestis from Food Using Immunomagnetic Separation and Pyrosequencing.

Interest has recently been renewed in the possible use of Y. pestis, the causative agent of plague, as a biological weapon by terrorists. The vulnerability of food to intentional contamination coupled with reports of humans having acquired plague through eating infected animals that were not adequately cooked or handling of meat from infected animals makes the possible use of Y. pestis in a foodborne bioterrorism attack a reality. Rapid, efficient food sample preparation and detection systems that will help overcome the problem associated with the complexity of the different matrices and also
